The Korean government has invested a tremendous amount of money in the last 10 years to build large public research infrastructures (LPRI). For efficient operation and maintenance of LPRI built with expensive equipment and professional engineers, reasonable budget needs to be allocated. However, it is difficult to fulfill sustainable operation and maintenance (O&M) because there is no standard on budgeting for efficient LPRI operation, including expensive equipment and manpower allocation. There have been a lot of cost assessment studies regarding O&M of high-demand facilities such as hospitals, hotels and residential buildings, but a very few on sustainable O&M of LPRI. Therefore, mid/long-term budget establishment plans for efficient LPRI O&M are required from the initial planning stage and a cost assessment model to support the plans should be developed. The objective of this paper is to propose a cost assessment model for sustainable operation and maintenance of large public research infrastructures. To do so, actual O&M data of 6 LPRI types in operation are collected, and regression analysis model (RAM) is used for development and evaluation a cost assessment model. The study result will support sustainable operation of LPRI from a business perspective and be used as basic data for continuous development of cost assessment models to establish budgets for LPRI operation from an academic perspective.
피부 섬유아세포에서 갈참나무 잎 추출물의 항산화 및 항노화 효능

본 연구는 예부터 약용으로 사용되었거나, 현재 식품공전에 식품원료로 사용이 가능한 것으로 등록된 국내 산림지역에 자생하는 식물을 식품산업에 활용하고자 선행연구에서 우수한 항산화 활성을 보인 갈참나무 잎을 본 연구에서 사용하였다. 70% 에탄올을 이용하여 추출한 갈참나무 잎을 이용하여, hydrogen peroxide로 산화적 스트레스를 유도한 피부 섬유아세포에서의 세포 보호효과, 세포내 항산화 효과 및 항노화 효과를 측정하였다. 세포 독성을 평가한 결과 25, 50 및 $100{\mu}g/mL$의 갈참나무 잎 추출물을 처리하였을 때 모두 독성을 나타내지 않았으며, hydrogen peroxide로 산화적 스트레스를 유도한 상태에서는 세포를 보호하여 농도 유의적으로 세포생존율이 증가하였다. 특히, $100{\mu}g/mL$의 농도에서는 양성대조군으로 사용한 $50{\mu}M$ ascorbic acid 수준까지 세포 생존율이 증가하였다. 세포내 항산화 효과를 확인 하기위해 사용한 $H_2-DCFDA$ assay에서는 형광현미경과 형광흡광도 측정에서 모두 농도 유의적으로 세포내 ROS 저감 활성을 확인하였고 갈참나무 잎 추출물을 $100{\mu}g/mL$ 농도로 처리했을 때는 $50{\mu}M$ ascorbic acid와 비슷한 세포내 항산화 효과를 나타내었다. 또한 SA-${\beta}$-galactosidase assay를 이용한 갈참나무 잎 추출물의 피부 섬유아세포에 대한 항노화활성은 ROS 생성 억제 효과와 유사한 경향으로 갈참나무 잎 추출물의 농도 유의적으로 세포 노화 억제효과를 확인하였다. 이상의 결과를 종합하여 볼 때 갈참나무 잎 추출물이 hydrogen peroxide로 인한 산화적 스트레스 상태에서 세포 보호효과, 항산화 효과 및 항노화 효과가 관찰되어 기능성 식품원료로서의 활용도가 매우 넓을 것으로 판단된다. The skin of the human body occupies the largest surface area of the body and acts as a protection for the person's internal organs. As such, the skin is a major target of oxidative stressors, and these oxidative stressors are known to contribute to skin aging over the course of time. For the most part, an antioxidant is an effective approach to utilize to prevent symptoms related to the reactive oxygen species (ROS)-induced aging of the skin. Therefore, we investigated the antioxidant and anti-aging activity of the leaves of the Quercusaliena Blume extract (QBE). In our study, we confirmed that the cell viability tested with XTT {2,3-bis(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide innersalt} assay was not affected up to a concentration of $100{\mu}g/mL$. In addition, the cell viability of HDF cells induced by hydrogen peroxide was recovered from 81% to 104% after treatment with QBE, which showed the greater protective effect than that of ascorbic acid. Treatments of QBE dose-dependently inhibited reactive oxygen species (ROS) production in HDF cells induced by hydrogen peroxide, which correlated with their protective effects on cell viability. Since QBE treatment exhibited the suppression effect of skin aging by decreasing the ROS production, QBE could be used as a not only natural anti-aging but also antioxidant resource.
LPS로 유도한 RAW 264.7 세포의 염증반응에서 자초(紫草)의 항염증 효과

Objective: Lithospermum Erythrorhizon (LE) has been used as an anti-bacterial and anti-inflammatory agent. However, it is unclear that LE aqueous extract could show the anti-inflammatory effects in RAW 264.7cells. The purpose of this study was to investigate the anti-inflammatory effect of aqueous extract from LE on lipopolysaccharide (LPS) - induced inflammatory response. Methods: To measure out the cytotoxicity of LE, we performed the MTT assay. To evaluate the anti-inflammatory effects of LE, we examined the inflammatory mediators such as nitric oxide (NO), prostaglandin E2 (PGE2) and pro-inflammatory cytokines (tumor necrosis factor (TNF)-a, interleukin, (IL)-1β and (IL)-6) on RAW 264.7 cells. We also examined molecular mechanisms such as mitogen-activated protein kinases (MAPKs) and nuclear factor-B (NF-κB) activation by western blot. Results : Aqueous Extract from LE itself did not have any cytotoxic effect in RAW 264.7 cells. Aqueous extract from LE inhibited LPS-induced productions of inflammatory mediators such as NO, PGE2, and pro-inflammatory cytokines including TNF-a, IL-1β and IL-6 in RAW 264.7cells. In addition, LE inhibited the phosphorylation of p38 kinases (p38), c-Jun NH2-terminal kinase (JNK), and NF-κB activation in RAW 264.7 cells. Conclusion : LE down-regulated LPS-induced production of inflammatory mediators through the inhibition of p38, JNK and NF-κB activation. Taken together, these results could provide the evidence for the anti-inflammatory effects of LE. Therefore, LE may be a novel target in the management of inflammation and help to support a potential strategy for prevention and therapy of inflammatory diseases.
3T3-L1 세포분화 중 지방축적 및 활성산소종 생성에 대한 국내 산림자원 5종 추출물의 효과

본 연구는 국내 산림자생 식물 추출물 5종(밤나무, 진달래, 산수유, 복분자딸기, 갈참나무)에 대한 지방세포 분화 억제 및 활성산소종(ROS) 생성 억제 효능을 평가하였다. 지방세포 분화 중 세포 내 지방축적 및 ROS 생성량을 비교한 결과, 국내 산림 식물 5종 모두 지방축적량 및 ROS 생성량을 농도 유의적으로 감소키는 것으로 나타났다. 특히 복분자딸기 추출물을 처리한 지방세포에서 지방 축적 및 ROS 저감에 가장 크게 나타났으며 양성대조군으로 사용된 N-acetyl-Lcysteine과 유사한 결과를 나타내었다. 또한, 복분자딸기 추출물을 처리함으로써 지방세포의 분화를 조절하는 전사인자인 PPARγ, C/EBPα 및 aP2 발현이 억제됨을 확인하였다. 국내 산림 식물 5종 추출물은 천연물에서 유래한 항비만 및 항산화 활성 기능성식품 소재로 활용 가능할 것으로 생각한다. This study investigate the effects of extracts of five species of Korean native forest plants on lipid accumulation and reactive oxygen species (ROS) production during the differentiation of 3T3-L1 cells. Our results show that Korean native forest plants extracts significantly reduced lipid accumulation and ROS production during adipogenesis in 3T3-L1 cells. Especially, Rubus coreanus Miq. was most effective in the inhibition of lipid accumulation and ROS production at a concentration of 100 μg/mL. Moreover, Rubus coreanus Miq. extracts significantly inhibited adipocyte differentiation, which is dependent on down-regulation of peroxisome proliferator-activated receptor gamma, CCAAT/enhancer-binding protein alpha, and adipocyte-specific fatty acid binding protein, a key adipogenic transcription factor. Therefore, these results suggest that Rubus coreanus Miq. might be a valuable source of bioactive compounds with anti-adipogenic activity.
리놀레인산 자동산화에 미치는 페놀계 산화방지제의 활성 및 잔존량 평가

본 연구는 리놀레인산에 7종의 산화방지제를 각각 0.02% 농도로 첨가하여 50°C의 dry oven에서 8일간 자동산화 시키면서 이들 산화방지제의 잔존량 및 산화안정성을 측정하였다. 잔존량은 HPLC를 사용하여 측정하였으며, 산화안정성은 과산화물가를 이용하여 분석하였다. 잔존량은 0일째(100%)에서 시간에 따른 변화율을 측정하였고, 또한 잔존량을 이용하여 100-잔존량(%)으로 손실률(%)을 계산하였다. 산화방지제 손실률과 과산화물가의 상관관계를 분석하여 연관성을 확인하였다. 잔존량 분석 결과 tert-butylhydroquinone(TBHQ)> dodecyl gallate(DG)> octyl gallate(OG)> propyl gallate(PG)> butylated hydroxyanisole(BHA)> 2,4,5-trihydroxybutyrophenone(THBP)> butylated hydroxytoluene(BHT) 순으로 낮은 잔존량을 나타내었으나, BHA 같은 경우 다른 실험군에 비해 8일째의 잔존량이 4일째의 잔존량에 비하여 감소량이 크지 않았으며 TBHQ은 4일째의 잔존량이 이미 0에 가까운 값을 나타내어 8일째의 잔존량과 큰 차이가 없었다. 과산화물가 측정 결과 대조군> TBHQ> DG> OG> PG> BHA> THBP> BHT 순으로 높은 과산화물가를 보였으며 산화방지제 손실률과 과산화 물가의 상관관계 분석 결과 상관관계(correlation coefficient, R<SUP>2</SUP>)는 0.3913~0.9963의 범위로, 특히 BHT가 가장 높은 상관관계(R<SUP>2</SUP>=0.9963)를 보였다. BHA와 TBHQ를 제외한 대부분의 실험군에서 R2 값은 0.8769 이상의 높은 상관관계를 보였다. 본 연구에서 페놀계 산화방지제는 자동산화 과정에서 리놀레인산 기질 내 산화방지제의 잔존량과 기질의 산화안전성 간의 높은 상관관계를 보여주었다. This study assessed the antioxidant activity and residue level of phenolic antioxidants in autoxidation of linoleic acid. The antioxidant activity of phenolic antioxidants was measured based on peroxide value of linoleic acid at 50°C for 8 days. We further evaluated the residue level of phenolic antioxidants in the autoxidation period by HPLC-UV. The residue level of antioxidants changed with time starting on day 0 (100%) and was determined by 100-remaining of antioxidants (%). Our results showed that peroxide values ranged from 0.33 to 10.18 meq/kg in propyl gallate, from 0.67 to 11.01 meq/kg in dodecyl gallate, from 0.01 to 10.34 meq/kg in octyl gallate, from 0.01 to 4.17 meq/kg in butylated hydroxytoluene (BHT), from 1.00 to 5.85 meq/kg in butylated hydroxyanisole (BHA), from 0.33 to 4.18 meq/kg in 2,4,5-trihydroxybutyrophenone, and from 1.00 to 11.01 meq/kg in tert-butylhydroquinone (TBHQ). Among the residue levels of antioxidants, on day 8, BHT showed the highest level while TBHQ showed the lowest. BHT showed the highest correlation coefficient, whereas BHA showed the lowest. This study proves that the residual level of phenolic antioxidants has a good correlation with the degree of autoxidation in linoleic acid.

The purpose of this study was to demonstrate the quality characteristics of Brassica juncea cultivated in Jeongseon (BJJ), South Korea. We analyzed the nutritional components and antioxidant activity of BJJ. As a result of the free sugar analysis, the contents of glucose and fructose in BJJ were $0.29{\pm}0.02g/100g$ and $0.10{\pm}0.00g/100g$, respectively. The major fatty acids were palmitic acid, octadecenoic acid and stearic acid. The palmitic acid was the highest at 31.22% of all fatty acids. The major minerals were identified as Ca, P, K, Mg and Na. The contents of vitamin $B_1$, vitamin $B_2$, vitamin $B_6$, vitamin C and vitamin E in BJJ were $0.02{\pm}0.00mg/100g$, $0.087{\pm}0.01mg/100g$, $0.02{\pm}0.00mg/100g$, $0.56{\pm}0.06mg/100g$ and $0.20{\pm}0.03mg\;{\alpha}-TE/100g$, respectively. As a result of the free amino acid analysis, total amino acid contents in BJJ were $2,801.21{\pm}115.38mg/100g$. L-proline content was the highest ($744.30{\pm}119.06mg/100g$) in BJJ. BJJ extract inhibits reactive oxygen species production in 3T3-L1 adipocytes. Also, BJJ extract exhibits a protective effect on oxidative stress in $H_2O_2$-induced human dermal fibroblast. These results indicate that BJJ comprises various valuable nutrients which can be used as functional food ingredients.
술폰화 poly(arylene ether sulfones) 양이온 교환막의 술폰산기 농도 변화에 따른 특성

이온 교환막의 고정이온 농도변화에 따른 막 특성을 알아보기 위해 matrix인 PSF (polysulfone)과 술폰화제인 CSA(chlorosulfuric acid)의 몰 비를 달리하여 SPSF (sulfonated polysulfone) 양이온 교환수지를 합성하였다. 합성된 SPSF의 이온교환용량, 함수율, 고정이온 농도, 분배계수 등을 측정하였다. DSC, TGA분석으로부터 SPSF의 술폰산기 농도 증가에 따라 벌키한 술폰산기의 도입으로 인하여 유리전이 온도는 증가하고 중량 감소비는 감소하는 것으로 나타났다. 본 실험조건에서는 SPSF막의 구조가 SEM, AFM등의 분석을 통하여 비대칭인 것으로 나타났다. 교류 임피던스로부터 측정한 SPSF 막을 통한 $Na^+$ 이온의 겉보기 확산계수는 술폰산기 농도 증가에 따라 증가하였다. Sulfonated Polysulfone (SPSF) cation-exchange membranes were synthesized by introducing various ratio of chlorosulfuric acid (CSA) onto the main chain of polysulfone (PSF). Properties such as ion exchange capacity, water content, liked ion concentration, and partition coefficient were measured, respectively. Through the analysis of DSC and TGA, it has been shown that glass transition temperature increased and weight loss decreased as sulfuric acid group concentration increased. Structure of membrane measured by AFM and SEM was seen to be asymmetric. Apparent diffusion coefficient of sodium ions through SPSF membrane by AC impedance was increased as sulfuric acid group concentration increased.
