        작품논문 : 화예작품 꿈꾸는 아이들의 조형표현에 대한 연구(1)

        They expressed plastically energetic and vivid figures of children in various partterns, using material and guality of vegetable and wire. Expressed as abridged forms intended to emphasize figures of plastic arts, and using material quality and colors as constitutive elements of three-dimensional space, it tried to achieve enlarged esthetic consciousness of floral arts which is good for hightening displaying effects expression.

        한국 고등학교 생물과 교육과정의 목표 변천에 관한 연구

        The Biology education objectives of high school due to the change of curriculum are compared and analyzed as following; 1. The curriculum was reorganized by the need of a nation society, discipline in Korea. 2. The change of the curriculum at high level in objectives area is as following; 1) During syllabus period, it was trained a democratic citizenship. During the course of study period, it was taken serious view of textbook and enforced an education in accordance with the real state of Korean affairs. These periods are called life-centered education and subject-centered curriculum. 2) During the first curriculum period, it was enforced scientific techniques in order to make a scientific citizenship. This period is called experience-centered curriculum. 3) During the second curriculum period, it was accumulated the knowledge due to progress rapid developments of sciences and techniques, realized an idea of The National Education Charter. This period is more emphasis than other periods in the cognitive domain. This periods is called discipline-centered curriculum. 4) Present the third curriculum period, it enables to cope with an unknown future society. This periods is called human-centered curriculum. 3. The change of the curriculum at middle level in objective area is as following; 1) During syllabus and the course of study period have achieved a necessary knowledge from the surrounding environments. 2) The first curriculum period devided into biology I and II. The purpose of biology 1 is to train fundamental facts and principles through the observation and experiments, to cultivate the attitude to apply the knowledge to an actual life and health. the purpose of biology II is to train practical knowledge and cultivate the attitude to apply the knowledge to the development of science. 3) The purpose of the second curriculum period is to understand basic concepts through the process of inquiry in order to contribute to the development of the country. 4) present the third curriculum period' is deviled into biology I and II. The purpose of biology I is to learn basic concepts of the life phenomena which is related with biological structure of human being anti to become men of scientific literacy. The purpose of biology II is to learn the fundamental knowledge and to keep learning for knowledge's sake, 4. The change of the curriculum at low level in objectives area is as following; 1) The purpose of syllabus period is to gain the knowledge of general biology through practice and possess the ability of application. 2) The purpose of the course of study period is to gain the knowledge of plants, animals, and physical hygiene, to emphasize on the human diseases and physical health. 3) The first curriculum period; The purpose of biology I is to understand the living phenomena of organism through experiences and to apply those to human being and to establish to conceptual scheme. The purpose of biology II is to deepen knowledge of biology and to get trained in applied biology. 4) The purpose of the second curriculum period is to make understood the structural knowledge of biology and to emphasize on the ecological phenomena. 5) The third curriculum period; The purpose of biology I is to make understood the phenomenal and macroscopic knowledge of life phenomena of organism. The purpose of biology II is to make understood the microscopic knowledge and to enable to study biology continuously.

        원형질 융합에 의한 전분으로부터 에탄올 발효효모균주의 개량

        에탄올 내성 및 고 알콜 생성 균주인 S. cerevisiae BU-1 와 glucoamylase 생성 우수 균주인 S. diastaticus A15 를 시험 균주로 하여 genetic manipulation 에 의하여 haploid 인 S. cerevisiae BUI.alpha.26($alc^r thr^-$) 와 S. diastaticus A15a6($STA^+ hom$) 균주를 분리하였다. 이등 효모 균주의 원형질체 형성을 위하여 0.8 M sorbitol 의 존재하에서 zymolyase $200\mu$g/ml 와 $400\mu$g/ml 을 2 시간 동안 처리시 두 균주 모두 95% 이상 원형질체를 얻었다. PEG 6000 을 90 분간 반응시킬 경우 융합 빈도는 $ 3.25 {\times} 10^{-3}$ 이었다. 원형질체 융합으로 생성된 융합주의 유전적 분석, 에탄올 내성, glucoamylase 생성을 측정한 결과, 이들 융합주의 유전자형은 STA $alc^r$ thr hom. '/STA' $alc^r$ thr hom로 예상되었으며, glucoamylase 활성도는 A15a6 의 경우 2.7 units 이었으나, 융합주 F7, F10 은 각각 4.2 와 8.4 units 로 나타났다. 에탄올 내성 시험을 하여 선별한 결과, 가장 우수한 균주인 융합주 7개 중 F7 과 F10 을 선별하였다. 5% 의 녹말이 포함된 발효배양액에서 $30^{\circ}C$ 5일간 배양시 생성된 에탄올 생성은 융합주 F7 과 F10 은 각각 2.0% 와 1.85 이였다. Ethanol-tolerant strain, S. eerevisiae BUI a26 ($alc^r thr^-$) and gJucoamylase-producing strain, S diastatieus AI5a6 (STA+ hom-) were prepared by means of genetic manipulation, Protoplast fusion was carried out to introduce STA gene from AI5a6 strain to BUla26 strain, Protoplast formation was shown at 0,8 M sorbitol and 200 Jig/ml to 400 Jig/ml zymolyase treatment for 2 hours incubation, Fusion frequency was $ 3.25 {\times} 10^{-3}$ to the regenerated protoplast number using PEG 6000 for 90 min incubation. The excellent fusants with genotype of STA- $alc^r thr^-$ hom+/STA+ ($alc^s thr^+$ hom- (2n), F7 and FIO, were selected by ethanol-tolerant, ethanol fermentation, and glucoamylase production tests, Glucoamylase production of AI5a6 showed 2,7 units, but 4.2 or 8.4 units for F7 or FIO fusant at $30^{\circ}C$, Ethanol fermentation from 32% glucose by BUla26 was 14,0%(v/v) in fermentaion medium for 5 days incubation, but 14.5% or 15,0% for F7 or FIO strain, respectively. Ethanol fermentation from 5% starch was 2,0% by F7, or 1.8% by FIO strain in fermentation medium for 5 days fermentation.
