        수용성 β-glucan을 함유한 꽃송이버섯 발효액의 제조

        Glucan has been shown to have a significant role in the activation of the immune system, including increased activity of macrophages and so on. Sparassis latifolia (formerly S. crispa) is an edible mushroom abundant in dietary fiber and widely known to contain high levels of β-glucan. In the present study, fermentation broths containing soluble β-glucan were prepared by fermentation with mushrooms with four Lactobacillus species (L. plantarum subsp. Plantarum, L. acidophilus, L. helveticus, and L. delbrueckii subsp. Bulgaricus). After culturing four Lactobacillus spp. in MRS broth, each Lactobacillus was inoculated into MRS broth containing S. latifolia powder 5% (w/v) at 37℃ in an anaerobic incubator for five days. It showed that the β-glucan contents were different in each fermentation sample. The suitable conditions for the preparation of mushroom fermentation broths were investigated and discussed.

        버섯원물과 버섯 추출물 그리고 버섯 유래 화합물을 포함한 식품과 기능성식품

        Mushrooms and their extracts including purified ingredients, are currently used as foods, functional foods (and/or nutraceuticals), and medicines. These products have numerous bioactive compounds such as polysaccharides (mainly β–glucans), glycoproteins, nucleotide analogs, terpenoids, and polyphenols, which have exhibited antioxidant, antimicrobial, anticancer, antiviral, anti-obesity, and immunomodulatory activities. In this review, we discuss the current information on the biactivities of 10 popular mushrooms in Korea. We also summarize the information on mushrooms and the active compounds derived from them, as well as mushroom-based products such as foods, functional foods, and medicines. We believe this review could provide useful information for scientists and consumers who seek to develop new products and promote healthy food habits and lifestyle.

        아미노산 첨가가 꽃송이버섯 균사체 성장 및 베타글루칸, GABA 함량 변화에 미치는 영향

        Sparassis latifolia (formerly S. crispa) is used in food and nutraceuticals or dietary supplements, as rich in flavor compounds and β-glucan. Some previous studies have reported the effects of mushroom on brain function, including its neuroprotective effect. Thus, for this mushroom to be used as an effective nutraceutical for brain function, it would be desirable for it to contain other compounds such as γ-aminobutyric acid (GABA) in addition to β-glucan. In this study, the enhancement of growth and GABA production in the mycelium of medicinal and edible mushroom S. latifolia was investigated. Amino acids were added externally as the main source of nutrition, and the effects of amino acids were investigated using liquid medium, specifically amino acid-free potato dextrose broth (PDB). The amino acids added were L-glutamic acid (named PDBG medium) and L-ornithine (named PDBO medium). The growth of mycelia was determined to be 0.9 ± 0.00 g/L, 2.2 ± 0.16 g/L, and 1.93 ± 0.34 g/L PDBG respectively. The GABA content was 21.3 ± 0.9 mg/100 g in PDB medium, and it in PDBG 1.4% medium, at 115.4 ± 30.2 mg/100 g. However, the PDBO medium was not effective in increasing the GABA content of mycelia. Amino acids had little effect on the β-glucan content of mycelia. The β-glucan content was 39.7 ± 1.4 mg/100 mg, 34.4 ± 0.2 mg/100 mg, and 35.2 ± 9.2 mg/100 mg in PDB, PDBG 1.8% and PDBO 1.4% media, respectively. Addition of glutamic acid and ornithine positively affected the growth of S. latifolia mycelia, and glutamic acid positively affected GABA production; no degradation of GABA was observed with addition of glutamic acid.

        Bacillus subtilis를 이용한 국내산 신이대 잎 발효에 따른 영양성분, 폴리페놀, 항산화능 변화

        Bamboo leaf has many nutritional and bioactive compounds such as polyphenols, and it can be used for various foods application. Fermentation is one of appropriate processes that can affect the nutrition, taste and flavor, and antioxidant activities of foods. In this study, a representative domestic bamboo tree (Sasa coreana Nakai) leaves were fermented by Bacillus subtilis KCCM 11315 and the changes of carbohydrates, minerals, amino acids, and phenolic compounds and antioxidant activities were investigated before and after fermentation. During the fermentation period, firstly, the leaf was softened and turned to brown. The constituent carbohydrates were slightly increased from 432.09±5.38 mg/g to 458.42±7.39 mg/g, and free sugars decreased by 95% from 28.12±2.03 mg/g to 1.4±0.14 mg/g. Mineral was 20987.5±345.1 μg/mL, which was slightly increased after the fermentation compared to 20804.1±364.6 μg/mL before that. The total amino acids were increased to 73881.94±137.59 mg/100 g compared to 58464.51±109.12 mg/100 g before fermentation, and free amino acids decreased by more than 85% from 32782.67±92.49 mg/100 g to 4827.83±19.76 mg/100 g. Total polyphenols content (TPC) increased from 25.51±1.04 GAE*mg/g to 35.34±0.91 GAE*mg/g, and the total flavonoid content (TFC) increased to 80.57±0.22 QE*mg/g compared to 69.64±0.26 QE<SUP>*</SUP>mg/g. The total catechin content (TCC) of TFC was decreased from 69.64±0.94 mg/L to 58.23±0.76 mg/L. The DPPH radical IC50 of bamboo leaves decreased to 2624.85±37.03 μg/mL and the ABTS radical IC50 of 187.26±4.78 μg/mL was increased after the fermentation. These results could be used as essential nutritional data before developing processed food products using the bamboo leaf.

        수용성 β-glucan을 함유한 꽃송이버섯 발효액의 제조

        본 연구에서는 꽃송이버섯 건물과 powder를 각각 Yeast Extract broth와 MRS broth에서 Lactoballius 4종(L. plantarum, L. acidophilus, L. helveticus, and L. delbrueckii)과 함께 발효시켰다. 발효액의 final pH는 pH 3.9±0.02로 유산균의 일반적인 final pH 값과 일치함을 통해 유산균의 발효를 확인하였고, 유산균에 의한 꽃송이버섯 발효의 효과를 β-glucan 함량을 통하여 확인하였다. Yeast Extract broth 발효액은 YSB-2LP 1.65 g/100 g으로 β-glucan을 거의 함유하고 있지 않았지만 MRS broth 발효액은 Yeast Extract broth 발효액의 10배에 달하는 MPB-1LP 10.84 g/100 g을 함유하고 있었다. MPB-1LP는 꽃송이버섯 powder와 L.actobacillus plantarum으로 발효한 발효액으로 pH stress로 인해 만들어진 L. plantarum의 β-D-glucosidase에 의해 분해된 고분자량의 수용성 β-glucan이라 사료된다. 꽃송이버섯을 powder 상태로 L. plantarum과 함께 발효시켰을 때 꽃송이버섯의 수용성 β-glucan 추출 효율을 높일 수 있으며, 본 연구의 결과는 꽃송이버섯 유산균 발효물의 면역촉진 작용 연구를 위한 기초연구자료로 활용될 수 있을 것이다.       • 제주도 물영아리오름과 곶자왈의 버섯종 다양성

        제주도는 기후, 식생, 토양 등 자연환경이 독특해서 버섯의 종다양성은 내륙지역과는 크게 다를 것으로 예상된다. 제주도 물영아리오름과 곶자왈에코랜드에서 개최된 한국버섯학회 주관 2016년도 버섯생태조 사에서 51속 66종의 버섯이 관찰되었다. 구름송편버섯(Trametes versicolor) 등 5종은 물영아리오름과 곶 자왈에서 모두 발견되었으며, 물영아리오름에서는 털목이(Auricularia polytricha) 등 34속 36종이 발견되 었으며, 곶자왈(에코랜드)에서는 잔나비불로초(G. applanatum) 등 23속 29종이 발견되었다. 독특한 생태 환경을 보이는 제주도에서 발생되는 야생버섯자원은 새로운 소득품목개발과 신품종을 육성하는 매우 중요한 자산이다. 아울러 지속적인 생태조사를 통해 제주도의 자연환경을 보존하고 기후변화를 예측하 고 대비하는 데 기여할 것으로 전망된다. 물영아리오름은 제주도 서귀포시에 위치하며, 상록낙엽수(예 덕나무, 참식나무, 때죽나무 등), 큰천남성, 섬새우란, 금새우난, 사철난, 찔레나무 울타리, 습지식물인 고마리, 물고추나물, 보풀, 세모고냉이 등 식생이 다양하며, 국내 5번째로 람사조약습지로 지정되어 있 다. 곶자왈은 제주와 같이 용암분출로 형성된 지역에 잘 발달할 수 있으며, 난대림과 온대림을 중심으 로 광범위하게 숲을 형성하고 있으며, 식물종 다양성도 비교적 높다. 곶자왈지역에는 600여종 이상의 관속식물이 있으며, 제주고사리삼, 개가시나무, 으름난초, 수채, 제주물부터 등은 멸종위기식물이다. 섬 휘파람새, 직박구리 등 제주텃새와 긴꼬리딱새나 팔색조 등 희귀철새가 번식한다. *본 초록의 저자는 2016년 한국버섯학회 생태조사 및 분류워크숍에 참여한 분류전문가(고평열), 행사진 행 담당자(버섯과, 충남도원), 생태조사 수상자(학술상:충북도원 김민자, 실용상;용인초당고 김민준, 촬영 상;조선대 조한교, 골든벨상;제주대 이승학)을 대표저자로 선정하였습니다. 생태조사에 참여한 회원 60 명 모든 분들게 감사드립니다.

        Fruit Juice Supplementation Alters Human Skin Antioxidant Levels In Vivo: Case Study of Korean Adults by Resonance Raman Spectroscopy

        Carotenoids, found in many fruits and vegetables, are antioxidants that protect human skin from UV radiation. In humans, fruit and vegetable intake increases carotenoid contents in skin, which are conventionally assessed by invasive blood tests. In this study, 47 healthy Korean subjects (volunteers) consumed fruit juice containing tomato, apple, strawberry, or grape three times per week for 6 weeks. Skin antioxidant levels were measured by non-invasive resonance Raman spectroscopy. The correlation between skin carotenoid (SC) score with demographic data (age, height, weight) and juice supplementation and changes in SC scores among groups were analyzed. Variations in skin antioxidant levels increased with juice supplementation (p < 0.05). Fruit juice intake was significantly correlated with SC score, indicating increased skin antioxidant levels. Grape and tomato increased skin antioxidant levels and showed higher antioxidant activity than other fruits. Fruit juices containing high levels of carotenoids and antioxidants may provide modest benefits to human health.

        꽃송이버섯(Sparassis latifolia) 추출물 소수성 분획의 항암 활성

        The use of mushrooms has immense potential in many diverse applications. Until now, more than 3,000 species are consumed around the world, and more than 100 have shown promising clinical activity against cancer and other chronic diseases. Sparassis latifolia (formerly S. crispa) is an edible mushroom that harbors β-glucan reported to possess immunostimulatory and anticancer properties. However there have been no reports on the anticancer activity of hydrophobic fractions of S. latifolia. In this study, the anticancer activities of S. latifolia extract and hydrophobic fractions were investigated using AGS (stomach cancer), A529 (lung cancer), and HepG2 (liver cancer) cell lines. In cytotoxicity results of A529 cells, fractions of A2, A3, A4, A6, A7, A8, A9, and A10 in all 12 fractions show low IC50 values. For HepG2 cells, A7 fraction results in the lowest IC50 value while A7, A8, and A11 fractions show low IC50 values in AGS cells. S. latifolia extract lead to low cell viability in cancer cells, compared to positive control of paclitaxel. A compound with molecular weight of 181 were detected using HPLCMS but not identified yet. As a result, the hydrophobic fractions of S. latifolia EtOH extract would be a possible candidate as natural anticancer agents in the future.
