건조방법에 따른 잣나무(Pinus koraiensis) 잎 추출물의 주름개선 및 미백 효과

This study provide activity for beauty food of water and 80 % ethanol extracts from Pinus koraiensis leaves. Total phenolic content of extracts from Pinus koraiensis leaves were each 12.22 mg/g (Drying under hot air) and 17.93 mg/g (Drying under shade), 14.36 mg/g (Lyophilization) in water extracts (WE) and 11.9 mg/g and 20.63 mg/g, 17.96 mg/g in 80 % ethanol extracts (EE). The 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity of extracts from Pinus koraiensis leaves was 96.20 % in EE from drying under shade at extracts concentration. The 2,2`- azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical decolorization activity of extracts from drying under shade was 99.85 % in WE and 99.80 % in EE at extracts concentration. The antioxidant protection factor (PF) extracts from drying under shade type was 9.63 PF in WE and 10.48 PF in EE at extracts concentration. The thiobarbituric acid reactive substance from Pinus koraiensis leaf was 89.39 % in EE from drying under shade at extracts concentration. The elastase inhibition activity of EE for anti-wrinkle effect showed an excellent wrinkle improvement effect, showing 71.46 % in EE from lyophilization. Collagenase inhibition activity of EE from drying under shade was 97.48 % in extracts. Tyrosinase inhibition activity which was related to antimelanogensis was observed. The tyrosinase inhibitory effect of extracts from lyophilization was confirmed to be 60.4 % in EE more than another drying methods at extracts concentration. Through out all results, it can be expected Pinus koraiensis leaves extracts to use as a functional material for anti-oxidant and functional beauty food.
B16F10 melanoma cell을 이용한 캐모마일(Matricaria chamomilla L.) 추출물의 미백 효과

Matricaria chamomilla L. has been used as a bath agent in Europe because of its sterilization effect on the skin. Flowers contain terpenes, flavonoids are effective in relieving inflammation. Matricaria chamomilla L. has been reported to have various drug efficacies such as sedation, anti-diabetic effect and anti-arthritic effect, but there is little research on the scientific efficacy of whitening effect. The purpose of this study was to examine the whitening effect of Matricaria chamomilla L. extract and to investigate the mechanism of inhibition of melanogenesis. The extracts were used to determine tyrosinase inhibitory activity. The tyrosinase inhibitory activity of extracts was ineffective for water extract but in 60% ethanol extract was shown in a concentration-dependent manner. B16F10 melanoma cell was measured using a powder obtained by lyophilization of 60% ethanol extract. The toxicity was observed at a concentration of 75 μg/mL. And concentration range was selected to be at most 50 μg/ mL. The effect of tyrosinase, MITF, TRP-1 and TRP-2 on the expression of melanin protein was investigated in melanoma cells of B16F10 melanoma cells. As a result, it was confirmed that as the concentration of the extract increased, the melanogenesis level decreased and the protein expression level also decreased in a concentration dependent manner. Therefore, it was concluded that Matricaria chamomilla L. extract inhibited melanogenesis in cells. Based on the above results, it is expected that it will be used as a useful basic data for industrialization of whitening functional food of Matricaria chamomilla L.
번행초(Tetragonia tetragonioides)로부터 추출한 Phenolic Compounds의 미용 식품 활성

본 연구는 물과 ethanol을 추출용매로 사용한 번행초(Tetragonia tetragonioides) 추출물이 미용 식품 소재로 사용 가능한 기능성을 증명하고자 하였다. Phenolic compounds는 물 추출물에서 3.29 mg/g, 50% ethanol 추출물에서 4.14 mg/g의 결과를 각각 나타내었다. 전자공여능 측정 결과 200 μg/mL phenolic compounds의 농도에서 물 추출물은 98.45%, 50% ethanol 추출물은 91.20%의 활성을 나타내었다. ABTS radical 활성 측정 결과 100 μg/mL phenolic compounds의 농도에서 물 추출물은 97.28%, 50% ethanol 추출물은 97.03%의 활성을 나타내었다. Antioxidant protection factor(PF) 측정 결과 200 μg/mL phenolic compounds의 농도에서 물과 ethanol 추출물 모두 1.77 PF의 같은 결과를 나타내었다. Thiobarbituric acid reactive substance(TBARS) 측정 결과 100 μg/mL phenolic compounds의 농도에서 물과 ethanol 추출물은 94.77%와 95.64%의 높은 활성을 나타내었다. 미백과 관련 있는 tyrosinase 저해 활성을 측정한 결과 200 μg/mL phenolic compounds의 농도에서 ethanol 추출물은 34.96%의 억제력을 나타내었다. 주름 개선 효과를 측정하는 elastase 저해 활성과 collagenase 저해 활성을 측정한 결과 200 μg/mL phenolic compounds의 농도에서 ethanol 추출물만 78.90%와 61.3%의 높은 저해 활성을 나타내었다. 수렴 효과를 나타내는 astringent effect에서 200 μg/mL phenolic compounds 농도의 ethanol 추출물은 7.82%의 효과를 나타내었다. Hyaluronidase 저해 효과 측정 결과 물 추출물 200 μg/mL phenolic compounds의 농도에서 81.04%의 염증 억제 효과를 나타내었다. 이러한 결과들로 미루어 보았을 때 번행초 추출물은 항산화 작용과 미용 식품으로서의 기능성을 기대할 수 있다. This study examined the beauty food activities of water and ethanol extracts from Tetragonia tetragonioides. Thiobarbituric acid reactive substances of water and ethanol extracts were 94.77% and 95.64%, respectively, at 100 μg/mL of phenolics. Tyrosinase inhibitory activity, which is related to skin-whitening, was confirmed to be 34.96% for ethanol extracts at 200 μg/mL of phenolics. Elastase inhibitory activity and anti-wrinkle effect of 50% ethanol extracts were 78.9% at 200 μg/mL of phenolics. Collagenase inhibitory activity of ethanol extracts was 61.29% at 200 μg/mL of phenolics. Astringent effect was not detected in water extracts but was 7.82% for 50% ethanol extracts at 200 μg/mL of phenolics. Hyaluronidase inhibitory activity as a measure of anti-inflammation was confirmed to be 81.04% for water extracts at 200 μg/mL of phenolics. Based on these results, Tetragonia tetragonioides extracts can be used as a functional material and functional beauty food with antioxidant effects.

In Korea, scrub typhus usually occurs in October and November. Hemophagocytic lymphohistiocytosis (HLH) is a distinct clinical entity characterized by a high fever, pancytopenia, hepatosplenomegaly, histiocyte proliferation, and hemophagocytosis. We encountered a summertime case of severe scrub typhus presenting as HLH. A 49-year-old female complained of abdominal pain and fever 3 days in duration. On hospital day 3 she was transferred to the intensive care unit because of clinical deterioration accompanied by severe sepsis. As an eschar was evident on the right shoulder, we commenced doxycycline. Her condition improved dramatically and she was discharged on day 14. Although the indirect immunofluorescence antibody test (IFA) for Orientia tsutsugamushi was negative on admission, a repeat IFA test was positive; the antibody titer was 1:5,120 on hospital day 10. Scrub typhus should be considered during differential diagnosis in a patient with severe sepsis in any season except the fall. (Korean J Med 2017;92:321-325)

Elicitor를 처리한 삼백초의 phenolic compound의 함량은 대조구인 비처리구에 비하여 높아지는 것이 확인되었다. Elicitor 처리한 삼백초 추출물의 항산화 효과는 1.5 mg elicitor/leaf 1 g(elicitor 1.5)에서 3.0 mg elicitor/leaf 1g(elicitor 3.0)으로 elicitor의 처리 농도가 높아질수록 항산화 효과는 높아지는 것으로 확인되었으며, elicitor 처리에 의해 water 추출물보다 70% 에탄올 추출물의 항산화 효과가 더 우수한 효과를 나타내었고, 동일 추출물에서 첨가되는 phenolic 농도 구간별 통계처리 결과 phenolic 농도별로 유의성 있는 활성 증가가 관찰되었다. Elicitor 처리한 삼백초 추출물의 건강기능성 활성은 고혈압 억제 효과가 비처리구에 비해 우수한 억제 효과를 나타내었으며, elicitor 3.0의 경우 억제율이 약 2배나 상승하는 것을 확인할 수 있었고, positive control인 captopril의 억제 효과보다 더 우수한 고혈압 억제 효과를 나타내었다. Elicitor 처리한 삼백초 추출물의 통풍 억제 효과가 비처리구에 비해 우수한 억제 효과를 나타내었으며, positive control인 allopurinol의 억제 효과보다 더 우수한 XOase 억제 효과를 나타내어, 고혈압 억제와 통풍 억제 효과를 이용한 의료산업 및 건강기능성 분야에 매우 유용한 자원으로 활용이 가능할 것으로 판단되었다. Elicitor 처리한 삼백초 추출물의 항균효과는 비처리구에 비해 부분적으로 우수한 항균효과를 나타내었으며, elicitor 1.5에서는 비처리구인 대조구에는 나타나지 않았던 충치균인 S. mutans에 대한 억제 효과가 새로이 확인되었다. Elicitor 3.0에서는 대조구에는 없던 S. mutans에 대한 억제효과뿐만 아니라 다소 약하기는 하지만 위염, 위궤양의 원인균인 H. pylori균에 대한 억제 효과도 추가로 확인되었다. 따라서 삼백초 재배 시에 본 연구에서 사용한 elicitor를 처리하면 수확되는 삼백초의 단위 무게당 생리활성물질인 phenolic compound의 생산량을 높일 수 있고, 생리활성도 증대시킬 수가 있어 산업화를 위한 원료생산에 효율적인 재배기법에 활용할 수 있을 것이라 판단되었다. This study was designed to assess the anti-oxidative and biological activities of extracts from Saururus chinensis. It is more effective to use elicitor than non-treated elicitor when extracting phenolic compounds. Elicitor-treated S. chinensis showed higher total phenolic content than the control group. Anti-oxidative activity of elicitor-treated S. chinensis extract increased in a concentration-dependent manner as concentration increased from 1.5 mg elicitor/leaf 1 g (elicitor 1.5) to 3.0 mg elicitor/leaf 1 g (elicitor 3.0). Moreover, 70% ethanol extract showed higher anti-oxidative activity than water extract. Hypertension inhibitory activity of the elicitor-treated group was much higher than that of the non-treated group. Elicitor 3.0 showed a 2-fold higher inhibitory effect and higher than captopril, which was the positive control. Elicitor-treated S. chinensis extract showed high gout preventative activity compared to the non-treated group. It also showed higher xanthine oxidase inhibitory activity than the positive control allopurinol. Thus, it can be expected to be used as a source in the medical industry for hypertension inhibition and gout prevention. Elicitor-treated S. chinensis extract showed partial anti-microbial activity. Elicitor 1.5 showed Streptococcus mutans inhibitory activity against cavities, which was not observed in the control group. Elicitor 3.0 not only showed cavity preventive activity but also a Helicobacter pylori inhibitory effect, which is directly related to stomach ulcer and gastritis. Therefore, elicitor treatment during S. chinensis cultivation will increase total phenolic compound content in the plant source and increase bioactivity, which will yield high quality for industrialization.
