        화종에 따른 국내산 벌꿀 에틸 아세테이트 분획물의 산화방지능 및멜라닌 생성 억제 효과

        Honey is known to have a variety of health-promoting effects. Ethyl acetate fractions (EAFs) of four Korean domestic honeys from basswood, Korean raisin, chestnut, and acacia as well as a New Zealand manuka honey were analyzed comparatively to evaluate their antioxidative and antimelanogenic effects. The EAFs of basswood, Korean raisin, and chestnut honey had higher antioxidant capacities and tyrosinase inhibition activities than those of manuka honey. Pretreatment of B16F1 melanoma cells with EAFs at 100 μg/mL resulted in relative retention of melanin contents as follows: acacia honey (141.0%)>basswood honey (134.3%)>manuka honey (131.5%)>Korean raisin honey (107.3%)> chestnut honey (88.0%). Intracellular tyrosinase activities of B16F1 melanoma cells were significantly (p<0.05) lowered by EAFs of Korean raisin and chestnut honey than by EAF of manuka honey. Consequently, these results suggest that Korean domestic honeys from different floral sources serve as good sources of antioxidative and antimelanogenic agents.

        Changes in phenolics, soluble solids, vitamin C, and antioxidant capacity of various cultivars of hardy kiwifruits during cold storage

        Hardy kiwifruits (Actinidia arguta) containvarious bioactive compounds such as vitamin C and phenolicsand can withstand cold temperatures. Changes insoluble solid, vitamin C, total phenolic, and total flavonoidcontent, and antioxidant capacity of three cultivars of hardykiwifruits (A. arguta 9 A. deliciosa cv. Mansu, A. argutacv. Haeyeon, and A. arguta cv. Chiak) were comparativelyevaluated for 8 weeks of storage at 1 ± 0.5 C. After the8 weeks of storage, soluble solid content of three cultivarsincreased, whereas their vitamin C content decreased. Throughout this storage period, total phenolic and flavonoidcontent of cv. Mansu and cv. Haeyeon remained thesame, while antioxidant capacity of these two cultivars alsoremained similar but with slightly more variations. Cv. Chiak, however, showed a decrease in total phenolic andflavonoid content and antioxidant capacity. These resultssuggest that cold storage of the hardy kiwifruits maintainslevels of bioactive compounds.

        기혼 직장 중년여성의 우울 관련 다 수준 요인

        Objectives: This study is conducted to identify the factors influencing the depression symptoms of married middle-aged working women. Method: This study employed the data of the 8th Korean Survey of Women and Families that was completed by the Korean Women’s Development Institute in 2020. A total of 2,215 married working women between the ages of 40 and 65 years were included in the study. The data were analyzed with the logistic multivariate hierarchy regression analysis using the SAS 9.4. Results: Among the study subjects, 11.51% have depression symptoms between married middle-aged working women which have been defined as CESD-10. In Model 1, which identifies the sociodemographic factors, low subjective health status is associated with depression symptoms. To identify the social support factors in Model 2, having no perceived social status is associated with having depression symptoms. In Model 3, which identifies home factors, low equal gender role and low positive family affected by work are associated with depression symptoms. In Model 4, which is grounded in office work factors, high discrimination in the workspace is associated with depression symptoms. Conclusion: To address married middle-aged working women’s depression symptoms, it may be essential to adopt a multifactor-level approach with ecology.

        지방질다당류로 자극한 RAW 264.7 세포에서 청도반시 땡감 에틸 아세테이트 분획물의 항염증 효과

        본 연구에서는 적과한 청도반시 땡감을 40% (v/v) 에탄올-물 혼합 용액으로 추출하고, 이 추출물을 액체-액체 추출법으로 5개 분획물(노말-헥세인, 클로로폼, 에틸 아세테이트, 노말-뷰탄올, 물)을 얻었다. 5개 분획물 중에서 에틸 아세테이트 분획물이 총 페놀 함량, 총 플라보노이드 함량 및 산화방지능이 가장 높았다. 에틸 아세테이트 분획물은 지방질다당류로 자극한 RAW 264.7 세포에서 산화 질소, IL-6, 산화 스트레스를 감소시켰다. UPLC-ESIMS/MS를 이용하여 땡감 에틸 아세테이트 분획물에서 페놀 화합물로 갈산, 프로토카테츄산, 4-하이드록시벤조산, 쿼서틴-3-O-글루코사이드, 파라-쿠마르산, 쿼서틴을 동정하였다. 본 연구를 통해서 적과 과정에서 부산물로 버려지는 청도반시 땡감이 산화방지능 및 항염증 효과를 보유한 기능성 소재로서 활용될 가능성을 제시하였다. 그러나 향후 적과한 청도반시 땡감의 산화방지능에 기반한 항염증 효과를 좀 더 명확하게 밝히기 위해서는 청도반시 땡감에 존재하는 개별 생리활성물질의 정량 분석 및 항염증에 대한 분자생물학적 메커니즘을 규명할 필요가 있다. Unripe astringent persimmon (Diospyros kaki Thunb. cv. Cheongdo-Bansi) is a by-product produced when thinning out the superfluous fruit of persimmon. We investigated whether unripe astringent persimmon has antioxidative and anti-inflammatory effects. Unripe astringent persimmon extract was fractionated sequentially in n-hexane, chloroform, ethyl acetate, n-butanol, and water. The ethyl acetate fraction had the highest total phenolic content, total flavonoid content, and antioxidant capacity compared to those of the other fractions. Pretreatment of lipopolysaccharide-stimulated RAW 264.7 macrophages with the ethyl acetate fraction reduced nitric oxide, interleukin-6, and intracellular oxidative stress in a dose-dependent manner. Ultra-high-performance liquid chromatography-electrospray ionization-tandem mass spectrometry analysis revealed gallic acid, protocatechuic acid, 4-hydroxybenzoic acid, quercetin-3-O-glucoside, quercetin, and pcoumaric acid as the phenolic compounds of the ethyl acetate fraction. Collectively, these findings suggest that unripe astringent persimmon is a source of functional materials that can promote antioxidative and anti-inflammatory effects.

        창의융합의 학습플랫폼 구조 연구 - 반성적 사고교육 기반 캡스톤 디자인 수업분석을 중심으로 -

        The purpose of this study is to analyze the structure of creative fusion platform in the engineering. For this study, pieces of related literature were reviewed focusing on analyzing the structure of creative fusion learning platform through the analysis of thinking education-based Capstone design class operation. In addition, interviews with five professors in the field of engineering, aesthetics, and social science were analyzed. The key of analysis was the five phases of thinking education process used for Capstone design class. The findings of this study were as follows. Most of the professors recognized the importance of thinking education. However, it was found that the 5-phase thinking education process did not apply systematically to the operation of Capstone design class. Consequently, there was a discussion about the ways to operate Capstone design class in order to improve the class quality through experience-based thinking education instead of simply focusing on outcome-oriented creative learning experience, as well as about valuable outcome expectations for creative products. In addition, based on the study findings, the structure of creative fusion learning platform was proposed as basic structure, process structure, and outcome structure.

        Preserving the inflated structure of lyophilized sporopollenin exine capsules with polyethylene glycol osmolyte

        Extracted from natural pollen grains, sporopollenin exine capsules (SECs) are robust, chemically inert biopolymer shells that posess highly uniform size and shape characteristics and that can be utilized as hollow microcapsules for drug delivery applications. However, it is challenging to extract fully functional SECs from many pollen species because pollen grains often collapse, causing the loss of architectural features, loading volume, and bulk uniformity. Herein, we demonstrate that polyethylene glycol (PEG) osmolyte solutions can help preserve the native architectural features of extracted SECs, yielding inflated microcapsules of high uniformity that persist even after subsequent lyophilization. Optimal conditions were first identified to extract SECs from cattail (Typhae angustfolia) pollen via phosphoric acid processing after which successful protein removal was confirmed by elemental (CHN), mass spectrometry (MALDI-TOF), and confocal laser canning microscopy (CLSM) analyses. The shape of SECs was then assessed by scanning electron microscopy (SEM) and dynamic image particle analysis (DIPA). While acid-processed SECs experienced high degrees of structural collapse, incubation in 2.5% or higher PEG solutions significantly improved preservation of spherical SEC shape by inducing inflation within the microcapsules. A theoretical model of PEG-induced osmotic pressure effects was used to interpret the experimental data, and the results show excellent agreement with the known mechanical properties of pollen exine walls. Taken together, these findings demonstrate that PEG osmolyte is a useful additive for preserving particle shape in lyophilized SEC formulations, opening the door to broadly applicable strategies for stabilizing the structure of hollow microcapsules.

        삼채의 잎, 인경, 뿌리의 총페놀 함량, 총플라보노이드 함량 및 항산화능

        삼채의 잎, 인경, 뿌리 세 부위를 물, 메탄올-물 혼합용매, 무수 메탄올을 이용하여 추출물을 제조한 후, 총페놀 함량, 총플라보노이드 함량 및 항산화능을 정량 분석하였다. 삼채의 부위별 총페놀 함량은 잎이 240.4-276.6 mg GAE/100 g FW, 인경이 65.5-82.8 mg GAE/100 g FW, 뿌리가 50.0-59.4 mg GAE/100 g FW를 보였다. 삼채의 부위별 총플라보노이드 함량은 잎에서 9.7-34.1 mg CE/100 g FW, 인경에서 5.0-16.7 mg CE/100 g FW, 뿌리에서는 4.4-15.3 mg CE/100 g FW로 나타났다. ABTS법, DPPH법, ORAC법을 통한 항산화능은 잎에서 각각 78.7-103.4, 24.4-59.0, 1,798.8-2,169.7 mg VCE/100 g FW, 인경에서는 35.8-51.1, 9.3-26.3, 415.3-611.7 mg VCE/100 g FW, 뿌리에서는 42.0-55.9, 12.8-24.4, 291.7-429.4 mg VCE/100 g FW을 보였다. 삼채 잎, 인경, 뿌리 중에서 잎이 가장 높은 총페놀 함량, 총플라보노이드 함량 및 항산화능을 가졌다. 6가지 추출용매에서 60% (v/v) 메탄올-물 혼합용매에서 주로 높은 활성을 나타냈다. 항산화능은 총플라보노이드 함량보다 총페놀 함량과 더 높은 상관관계를 보였다. 본 연구의 결과는 삼채가 높은 항산화능 공급할 수 있는 원료로서의 가능성을 보여 주었다는데 의의가 있다. To quantitatively evaluate the total phenolics, total flavonoids, and antioxidant capacity in the leaves, bulbs, and roots of fresh Allium hookeri, they were extracted using various solvents including water, aqueous methanol (20, 40, 60, and 80%; v/v), and absolute methanol. The leaves had the highest levels of total phenolics (240.4-276.6 mg gallic acid equivalents/100 g) and total flavonoids (9.7-34.1 mg catechin equivalents/100 g). The highest antioxidant capacities of 78.7-103.4 mg vitamin C equivalents (VCE)/100 g, 24.4-59.0 mg VCE/100 g, and 1,798.8-2,169.7 mg VCE/100 g in the leaves were also observed using 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 2,2-diphenyl-1-picrylhydrazyl (DPPH), and oxygen radical absorbance capacity (ORAC) assays, respectively. The total phenolics had a higher linear correlation with antioxidant capacity than the total flavonoids. In general, 60% (v/v) aqueous methanol extract had higher levels of total phenolics and flavonoids, and higher antioxidant capacity than any other solvents used. This study suggested that A. hookeri might be a good source of phenolics and antioxidants.
