This study of embryo transfer was carried out to investigate the possibility of introducing new genetic material into closed herds for disease control and of transporting embryos between countries. The results obtained were summarized as follows: 1. On the basis of the number of ovulation points counted, 69.1% of embryos were recovered. 2. A very high proportion (86.6%) of 1-cell embryos were recovered from the oviducts of donor sows at 37 to 41 hours after first mating. 3. Sixty-seven embryos were transferred to 5 recipients. Four recipients farrowed a total of 18 pigs, giving a survival rate of 26.9%. 4. Average pig weight at birth was 1.46㎏.

This is a preliminary study to simplify freezing, thawing and transfer procedure of cattle embryos using mouse embryos. The experiments were designed to investigate the effective concentration of glycerol and sucrose by one-step straw method, and to determine the optimum thawing temperature. Effects of viability among permeable cryoprotec-tants and impermeable sugar dilutions was also studied. Results obtained were summarized as followed; 1. A total of 1,340 embryos were cryopreserved among which 1,390 embryos collected from 136 mice. 2. Post-thaw development to expanded blastocysts, was highest when embryos were frozen in 1.0 M glycerol+0.5 M sucrose. However, proportions of morphologically normal embryos was highest when they were frozen in 0.25 M glycerol+0.5M sucrose. 3. Post-thaw development to expanded blastocysts was highest when embryos were frozen in a lower concentrated sucrose compared to higher concentration. 4. The temperature thawed at 20℃ and 30℃ resulted in a significantly higher embryos survival rate after 72 hr. in culture than did at 35℃. However no significant differences was denoted between 20℃ and 30℃ (p$lt;0.05). 5. In effects of non-permeable sugar dilutions on in-vitro development of frozen-thawed mouse embryos, raffinose and trehalose were compared favorably with sucrose dilution in morphological recovery and survival rates. 6. When the embryos were frozen-thawed rapidly in 1.0 M glycerol and trehalose concentration, the proportions of embryos developed to expended blastocyst were significantly better in 1.25 M and 0.5 M than other higher concentration (p$lt;0.01). 7. In effects of permeable cryoprotectants and sucrose concentration, 1.5 M of permeable freezing media were significantly higher survival rate than that of 2.0 M and propanediol was resulted in slightly higher survival rate than glycerol. However, no significant differences showed among sucrose concentrations exception of null dilution(p$lt; 0.01). The results from this experiment suggested that the use of propanediol and raffinose in the rapid freezing technique offers slightly better results than does the use of glycerol and sucrose.
