      • NF-kB가 매개하는 염증에 의한 당뇨에 있어서 운동의 역할 연구

        The purpose of the present study is to review the role of exercise to influence on the diabetes by Nuclear facotr-kB (NF-kB) mediated inflammation. Inflammation plays a key role in the pathogenesis of the diabetes. NF-kB, which is an transcription factor, is increased in experimental dibetes model. The anti-inflammatory effects of exercise are as follows. 1) Release of interleukin-6 from contracting muscle. 2) Down-regulation of toll-like receptor expression. Exercise-induced muscle glucose uptake requires three steps. 1) Glucose delivery from blood to muscle. 2) Membrane glucose transport. 3) Muscle glucose phosphorylation. Eventually, exercise increases insulin sensitivity through three serial steps in the working muscle. Until now, study on the role of exercise on NF-kB midiated inflammation is not well established. Anti-inflammatory effects of exercise via NF-kB/inflammation pathway will be a potential approach for the therapy of diabetes.

Recent research data for diabetes, exercise and inflammation were collected from Pubmed. Insulin resistance is inflammatory response including immune response. Inflammation plays a key role in the pathogenesis of diabetes. COX 2, which is an inflammation maker, is increased in experimental diabetes model. The anti inflammatory effects of exercise are as follows. 1) Reduction in visceral fat mass. 2) Release of interleukin 6 from contracting muscle. 3) Down regulation of toll like receptor expression.         염증에 의한 비만에 있어서 PGC-1의 역할 연구

        본 연구의 목적은 학교체육프로그램개발을 위한 염증에 의한 비만에 있어서 PGC-1의 역할을 검토하는 것이 다. 비만, 염증, PGC-1 그리고 운동에 관한 최근연구결과들은 MEDLINE 데이터베이스 검색엔진인 PubMed 원문 검색사용법을 통해 수집되었다. 염증은 비만의 병태생리에 있어서 중요한 역할을 한다. 전사인자인 PGC1은 비만상태에 있어 기능이 약화되어 있다. 비만에서 운동의 항염증효과는 다음과 같다. 1) 내장지방감 소 2) Toll-like receptor 발현감소 3) 근육에서의 interleukin-6 분비. PGC-1은 항염증 효과를 가진다. 지금까지 염증에 의한 비만에 있어서 운동을 통한 PGC-1의 역할에 대해서는 잘 알려져 있지 않다. 운동을 통한 PGC-1의 항염증효과는 비만치료에 대한 잠재적 접근이 될것이며 이는 학교체육프로그램개발에 있어서 기초 자료를 제공할 것이다. The purpose of the present study is to review the role of Peroxisome proliferator activated receptorγ coactivator-1 (PGC-1) α on the obesity by inflammation for school exercise program development. Recent research data for obesity, inflammation, PGC-1α and exercise were collected from PubMed, which is a free search engine accessing primarily the MEDLINE database. Inflammation plays a key role in the pathogenesis of the obesity. PGC-1α, which is transcriptional coactivator, is diminished in the obesity. The anti-inflammatory effects of exercise training on the obesity are as follows. 1) Reduction in visceral fat mass 2) Down-regulation of toll-like receptor expression 3) Release of interleukin-6 from muscle. PGC-1α has an anti-inflammatory effect. Until now, a study on the role of PGC-1α via exercise training on the obesity by inflammation is not well established. Anti-inflammatory effects of PGC-1α via exercise will be a potential approach for therapy of obesity and provide basic data for school exercise program development.

        Avenanthramide C suppresses hypoxia-induced cyclooxygenase-2 expression through sirtuin1 activation in non-small-cell lung cancer cells

        Avenanthramide C (AVC), found mainly in oats, mediates anti-inflammatory activities by reducing the anti-inflammatory cytokine levels. This study investigated the effects of AVC on hypoxiainduced cyclooxygenase-2 (COX-2) expression in A549 cells. AVC suppressed the hypoxiainduced increase in COX-2 protein levels and promoter activity. We also observed that the effects of AVC were reversed by a SIRT1 inhibitor, indicating that the inhibitory effects of AVC on hypoxia-induced COX-2 expression are mediated by SIRT1. Therefore, AVC inhibits the hypoxic induction of COX-2 expression via SIRT1 activation. Our results suggest that AVC could be beneficial for preventing lung inflammation under hypoxia.

Exercise is a powerful physiological activator of SIRT1. The inflammation regulation mechanism of SIRT1 activation via exercise on the obesity by inflammation are as follows. 1) SIRT1 protein regulation by exercise in obesity model 2) The regulation of inflammation-related genes by SIRT1 via exercise in obesity model. Anti-inflammatory effects of SIRT1 via exercise will be a balancing approach for detrimental effects of obesity and provide fundamental data for the therapy of obesity.

        염증에 의한 비만에 있어서 AMPK의 역할 연구

        본 연구는 학교체육프로그램개발을 위한 염증에 의한 비만에 있어서 AMPK의 역할을 규명하는 종설 연구이다. 염증, 비만, AMPK 그리고 운동에 관한 연구결과는 PubMed 원문 검색엔진을 통해 수집되었다. 운동의 AMPK의 강력한 생리적 활성자이다. 비만에 의한 염증 진행에 있어서 운동의 AMPK의 활성화를 통한 염증조절기전은 다음과 같다. 1) 비만 유발모델에서 운동에 의한 AMPK 단백질 조절 2) 비만 유발모델에서 운동에 의한 AMPK의 유도에 의한 염증관련 유전자의 조절 3) 운동에 의한 AMPK 신호전달조절과 비만으로 인한 염증신호전달체계와의 상호연관성을 규명하고자 한다. 운동에 의한 AMPK 활성화를 통한 항염증효과는 비만의 인체 부정적인 영향에 대한 균형적 접근이 될 것이며 이는 학교체육프로그램 개발에 있어서 기초자료가 될 것이다. Recent data for inflammation, obesity, AMPK and exercise were collected from PubMed. Exercise is a powerful physiological activator of AMPK. The inflammation regulation mechanism of AMPK activation via exercise on the obesity by inflammation are as follows. 1) AMPK protein regulation by exercise in obesity model 2) The regulation of inflammation-related genes by AMPK via exercise in obesity model 3) Identification of interrelationship between AMPK signaling pathway via exercise and inflammation network by obesity. Anti-inflammatory effects of AMPK via exercise will be a balancing approach for detrimental effects of obesity and provide fundamental data for school exercise program development.

      • 렙틴 변형마우스에서 운동에 의한 간세포 자가포식 고찰

        Obesity is a multisystem disease caused by the failure of normal homeostatic mechanisms regulating food intake, fat storage, and energy utilization. Obesity causes the adverse effects in the body, such as inflammation, oxidative stress, and apoptosis. Leptin plays a clear role in obesity etiology, pathophysiology, and health outcomes, but it has not been fully understood. Leptin deficient ob/ob mice are a well-described model for metabolic studies that indicate hyperphagia and early onset obesity. Liver is a central organ with various functions including the elimination of toxins, the production of multiple hormones and proteins, and the storage of vitamins. This largest internal organ also plays a pivotal role in regulating blood sugar, and regulation of lipid storage. Autophagy plays a central role in energy homeostasis, and its induction is essential for lipolysis, a process that is also stimulated by leptin. The purpose of this study was to review the effect of exercise on the autophagy in the liver of leptin deficient ob/ob mice. This review also discuss how exercise in the liver of ob/ob mice activates autophagy and improves Non-alcoholic fatty liver.

        8주간 하체운동이 여성노인의 신체조성과 SRT에 미치는 영향

        본 연구는 8주간의 하체 운동이 여성 노인의 신체조성과 기대수명의 예측인자인 Sitting Rising Test (SRT)에 미치는 영향을 규명하는 것으로서 B 시 거주 65세 이상 여성 노인들을 연구 대상으로 하였다. 총 8주간 주3회에 걸쳐 각각 50분 동안 하체 저항성 운동프로그램을 실시 한 후 트레이닝에 따른 신체조성 (체중, 체지방률, 체지방량, 제지방 량)과 혈압 등의 인체 변인을 포함하여 근지구력 및 SRT 스코어의 변화를 측정하였다. 본 연구 프로그램을 통한 8주간 하체 저항성 운동은 체중과 체지방량을 유의하게 감소시켰으며, 근지구력 및 SRT 지표는 향상되는 것으로 나타났다. 이는 여성노인들의 하체운동을 통해 기대수명을 연장시킬 수 있음을 말해준다. In this study, we examined the effect of lower body exercise on body composition and sitting rising test (SRT) in elderly women. Subjects who were elderly women in B city participated in 8 weeks lower body exercise program. Sessions ran three times weekly for 50 min. Change of body composition, blood pressure, muscle endurance test and SRT were measured and statistically analyzed before and after the lower body exercise program. Lower body exercise decreased body weight and body fat mass, and increased muscle endurance and SRT score. Conclusionally, these data showed that lower body exercise in elderly women may extend life expectancy.

        Effects of estrogen receptor and estrogen on the chromatin structure in estrogen receptor stable transfectants

        The estrogen receptor (ER), a member of the nuclearhormone receptor superfamily recruits coactivatorsthat modify local chromatin structure. Here weinvestigated the effect of the estrogen receptor andestrogen on the global chromatin structure and thelocal chromatin structure of the progesterone recep-tor gene during the process of transcriptional acti-vation using Rat1+ER cells stably expressing theestrogen receptor. The total chromatin was moreaccessible to DNaseI in Rat1+ER cells than in theparental estrogen receptor-negative Rat1 cells. After18 h of estrogen treatment, total chromatin wasmore dispersed in Rat1+ER cells than in Rat1 cells.The chromatin structure of the progesterone recep-tor gene was more sensitive to DNaseI in Rat1+ERcells than in Rat1 cells. However, the chromatinstructure of the progesterone receptor gene did notchange further on estrogen treatment. Our resultssuggest that under certain circumstances unoccu-pied estrogen receptors may play some role in reor-ganizing the repressive chromatin structure toinduce gene activation.

        Development of an efficient endothelial cell specific vector using promoter and 5' untranslated sequences from the human preproendothelin-1 gene

        Rafts, cholesterol- and sphingolipid-rich membrane microdomains, have been shown to play an im-portant role in immune cell activation. More re-cently rafts were implicated in the signal trans-receptor (TNFR) family. In this study, we provide evidences that the raft microdomain has a crucial role in RANK (receptor activator of NF-κB) signaling. We found that the majority of the ectopically expressed RANK and substantial portion of endogenous TRAF2 and TRAF6 were detected in the low-den-sity raft fractions. In adition, TRAF6 association with rafts was increased by RANKL stimulation. The disruption of rafts blocked the TRAF6 trans-teraction betwen RANK and TRAF6. Our obser-vations demonstrate that proper RANK signaling requires the function of raft membrane microdo-mains.
