      • 생쥐에서 오가피에 의한 싸이토카인 유도와 면역반응에 관한 효과

        This experimental study was carried out to evaluate the effects of Acanthopanacis cortex on Cytokine-inducing and and immune response in Mice. In order to investigate the effect of Acanthopanacis cortex, the following was performed; Cytotoxicity, in vitro, the fraction of CD4', CD8', B220' in splenic cell, gene expression of IL-12(p35), IL-12(p40), IFN-γ, and splenic cell proliferation by Acanthopanacis cortex. Analysis of cytokine gene expression was carried out by RT-PCR amplification. Amplified PCR products were electrophoresed on 1.2% agarose gel, and the analysis(Ht) was used to 1D-density program. The results were obtained as follows. Acanthopanacis cortex showed didn't have cell toxicity under 12㎍/㎖ group on mouse lung fibroblast cells. In an in vitro model using mouse peripheral blood mononuclear cells (PBMCs), extract of Acanthopanacis cortex induced multiple cytokine, including interleukin-12 (p35), interleukin-12 (p40), interferon-gamma (IFN-γ). The extract also enhanced the percentages of the CD4', and CD8' in the untrated control were 22.1±3.3 to 38.4±2.1 and 5.0±0.4 to 10.7±0.3%, respectively. From above findings, it is suggested that Acanthopanacis cortex is able to anti-cancer and activate immune response system.

      • 사물탕(四物湯)과 사물탕(四物湯) 구성약물(構成藥物)이 혈구감소증(血球減少症)에 미치는 영향(影響)에 관(關)한 연구(硏究)

        In order to inverstigate interactive effects of Samultang(SMT) and SMT composition and the principle of prescription, I examined the effects of SMT and combination of three components of SMT, In C57BL/6 mouse with oral administration of SMT and cyclophosphamide(CTX) treatment, white blood cells, platelet were counted. Also, those were anti-anemia effect for rat which have hemolytic anemia induced by phenylhydrazine. The results were summerized as follows : 1. In vivo analysis of the effect of the SMT by orally administrated C57BL/6 mice with the SMT and was treated with cyclophosphamide (CTX). When the number of white blood cell. platelet was counted, there was ameliorative effects of leucopenia or thrombocytopenia as a protection to CTX. 2. It was acknowledged that SMT and its composition increased RBC count and hemlytic significantly in rat which have hemolytic anemia. 3. Among SMT composition Angelicae gigantis Radix and Cnidii Rhizoma solution and among the combination group Rehmanniae Radix and Cnidii Rhizoma combination and SMT solution showed significant effects for hemoglobin and weight of spleen in mouse which have hemolytic anemia. Thus it was acknowledged that Rehmanniae Radix and Cnidii Rhizoma combination had effect to improve hemoglobin and weight of spleen. From above these results it was acknowledged that SMT has effects that ameliorative effects of leucopenia or thrombocytopenia as a protection to CTX and anti-anemic effect for the rat which have hemolytic anemia induced by phenylhydrazine and these effects depend upon its combination of SMT composition. Especially it was acknowleged, though there are the slight difference according to combination, that Paeoniae Radix Alba combination had improving effect for physical change. So it is though that Paeoniae Radix Alba is main material for this kind of disease and continuous study is needed.

      • 사향추출물이 생쥐 대식세포의 염증 유발 싸이토카인 유전자 발현에 미치는 영향

        To investigate the capacity of anti-inflammatory cytokines and biological resonse modifiers(BRMs) to induce IL-β,IL-6, TNF-α gene overexpression from mouse macrophages, we isolated the resident peritoneal macrophages from BALB/c mouse (8 week old) and incubated for 6 h with lipopolysaccaride (LPS) and Moschus moschiferus (MOMS) extracts. Analysis of inflammatory cytokines gene expression was carried out by RT-PCR amplification. Amplified PCR products were electrophoresed on 1.2% agarose gel, and the analysis(Ht) was used to 1D-density program. 1.LPS and MOMS extract treatments resulted in a significant decrease in IL-1β, IL-6, TNF-α mRNA expression level compared with the LPS treatment. 2.Among four sample of MOMS, Inhibitory effects of MOMS-A and MOMS-D for inflammatory cytokines gene expression were to be fine compared with the MOMS-B and MOMS-C. According to the above data, Because the anti- tumoral and anti-inflammatory response activities of macrophage are known to be dependent on the production of inflammatory cytokines(IL-1β, IL-6, TNF-α) by macrophages, we suggest that evaluations of BRM for the reduction of inflammatory cytokines production by macrophages are important for clinical application.

        산사자의 크로마토그램 다변량(多變量) 패턴 분석(分析)

        It is well known that Crataegus Fructus strengthens the Stomach and Spleen, helps digestion, tonify stomachache, and decreases bleeding. A major component isolated from this herb consist of carotene, riboflavin, quercetin, chlorogenic acid, amygdalin, ursolic acid etc. In this study, we aimed to analyze the changes of liquid chromatogram pattern, one of major standardization method, to determine the quality of Crataegus Fructus.

      • 牛膝이 齒牙 및 齒周疾患에 미치는 影響

        Achyranthis Bidentatae Radix(ABR) is important prescriptions that have been used in oriental medicine for stomatitis and wound healing. The study was done to evaluate the inhibitory effects of cytotoxicity, formation of superoxide on the macrophage and neutrophil, prostaglandins(PGE_2), interleul-ins(IL-1β), collagenase activity and synthesis of collagen and DNA. The results were obtained as follows: 1. ABR was not showed the proliferation difference of human fibroblast and monocyte in 0.01% and 0.001% concentrations to be experimented and in result, it was concluded that they have no cytotoxicity but showed cytotoxicity in 0.1% concentrations. 2. ABR inhibited the formation of superoxide to 48% at the concentration of 0.001% in the mouse monocyte. 3. ABR inhibited the formation of superoxide to 40% at 0.001%, 58% at 0.0001% as compared with control in the human monocyte. 4. ABR inhibited the formation of superoxide to 58% at 0.0001%, 40% at 0.001% in the human neutrophil. 5. ABR was not showed the proliferation difference of human monocyte in all concentrations to be experimented and in result, it was concluded that they inhibited the formation of prostaglandins(PGE_2) in the human monocyte stimulated with E. coli. 6. ABR showed the all concentration of inhibiting the production of interleukins(IL-1β) in the human monocyte stimulated with E. coli. 7. ABR didn't influence on collagen synthesis and total protein in fibroblasts. 8. ABR inhibited the collagenase activity to 84% at 0.1%, 69% at 0.2%, 76% at 0.5%, 91% at 0.001% respectively.

        사향(麝香)의 수용성분(水容性分)이 생쥐 복강내(腹腔內) 거식세포(巨食細胞)의 활성(活性)에 미치는 영향(影響)

        The musk has been reported to have significant anti-inflammatory activities in clinical use and several animal models and we examined the effects of water soluble fraction(WSF) of the musk on murine peritoneal macrophages. WSF decreased the production of nitric oxide from the lipopolysaccharide(LPS)-treated murine peritoneal macrophages and also reduced the phagocytic activity of macrophages on the opsonized sheep red blood cells(SRBC). Transcriptional expression level of the inducible nitric oxide synthase(iNOS) was also decreased and the viability of the treated macrophages was not affected by WSF, suggesting that the effects could be partly explained by transcriptional regulation. Contrary to down-regulating iNOS expression, WSF slightly increased the release of tumor necrosis factor-alpha$(TNF-{\alpha})$, which implied its selective action on cellular pathways activated by LPS. Our results showed that anti-inflammatory activities of the musk could be partly explained by the inhibitory effects of the water soluble fraction on the macrophageal activation.

      • 상백피 추출물이 당뇨병 마우스에 미치는 영향

        Morus alba extract(MAE) was tested for its ability to inhibit alloxan induced lipidperoxidatin. Lipid peroxide contents in serum, liver, kidney and heart were measured by the TBA method. ICR mice receiving alloxan at a dose of 60mg/kg intraperitoneally after a 24hrs starvation showed significantly increased lipid peroxide contents as compared to untreated control. Lipid peroxide contents in serum, liver, kidney of alloxan-diabetic mice were decreased by the treatment of MAE at the dose of 50mg/kg, 100mg/kg for 7 days.
