The fatty acid composition of muscle were investigated to compare muscle composition among the 9 domestic animals including cattle. In major domestic animals, analyzed the effects of age, part and sex of the animal on their fatty acid composition. The content of 4 types of major fatty acids of muscle was determined and calculated their ratio. Myristic acid and palmitic acid levels were high in chicken and sheep. Besides dog muscle contained a lot of stearic acid. Linoleic acid content showed evident difference in the content depending on the animal species. The ratios of linoleic acid/palmitic acid (L/P ratio) and linoleic acid/stearic acid(L/S ratio) were characteristically high in horse and pig, whereas the ratio of palmitic acid/stearic acid(P/S ratio) was $0.71{\pm}0.17$, showing very low level in dog. As for the content of stearic acid, in cattle and chicken it was higher in young animal than adults. In duck, the contents of all fatty acids and ratio were increased by the age. As for the content of fatty acids according to the part of chicken, high level was shown in thigh than in breast and wing, while there was no remarkable variation by the part in other animal. The differences in the content of myristic acid, palmitic acid and linoleic acid among some animal could be verified in muscle lipid composition. The L/P ratio which maintained certain level regardless of age, part, sex shown distinctive pattern between the species.

We analyzed the contents of 12 types of minerals such as calcium in muscle from various species. Thereafter we observed changes of the concentrations according to age, part and sex in major domestic animals. The concentrations of calcium, iron, sodium, potassium, magnesium and zinc were high respectively whilst the content of cobalt, chromium, copper, manganese, molybdenum and nickel were low respectively in the muscle. The concentration of calcium was high in duck and dog but low in pig and horse. Also high level of sodium content was detected in dog and the content of potassium, iron showed high level in horse compared with the content in other animals. In 6 types of microminerals as cobalt, the level of muscle were no more than 1 ppm showing very low content in all animal, but $2.99{\pm}0.85$ppm of copper in duck was an exceptional case. According to the age some species showed small range of variation centering on macrominerals and there was no remarkable change in microminerals. Distribution of minerals was different according to the part and the variation was very diverse compared with other factors such as age and sex. Additionally, the content of minerals in muscle was higher in female than in male chicken and duck. In conclusion, the difference of the content of minerals according to the species was mainly focused on macrominerals.
DNA hybridization을 이용한 축종특이성 구명

DNA hybridization assay using probes prepared from liver was carried out to identify species characterization of the domestic animals. Gel electrophoresis showed that the target DNA extracted from raw muscle were 1kb and uniform pattern while fragments size of heated muscle were irregular. Hybridization was performed by adding 200ng/ml probe in hybridization solution and incubating for 12 hours at $68^{\circ}C$. To obtain good discrimination, applied washing buffer and washing step differently depending on the species. The probes of pig, horse and dog formed the specific hybrids with each target DNA respectively. Although cross reaction was detected in cattle, goat and sheep but signal intensity among these species made the discrimination possible each other. Such pattern was the same in the cases of chicken, turkey and duck. The hybridization pattern of heated muscle was similar to that of raw muscle in general, but the signal intensity was inferior to that of raw muscle. Species identification between closely related animal species, hybridized using the target DNA of such closely related animal species as a blocking agent, remarkable increase of discrimination from the evident decrease of non specific reaction compared with the control group. In addition, in the admixture where certain meat was included in the beef, pork, chicken meat, we could find whether any unjust meat was admixed or not. In this case, detection limit of certain meat in admixture was 1%.

These studies were carried out to investigate the changes of blood pictures, hitological findings and testosterone concentrations after orchidectomy in ageing rat. The results obtained were summarized as follows: 1. The total W.B.C. counts were $9.27{\pm}12{\times}10^3/mm^3$ in the control group, $9.54{\pm}34{\times}10^3/mm^3$ in the sham-operation group and $12.08{\pm}28{\times}10^3/mm^3$ in the orchidectomy group when it was checked 50days after the operation. 2. The number of lymphocytes after orchidectomy began to increase gradually as time passes, thereafter markedly increased $10.12{\pm}03{\times}10^3/mm^3$ at 50days after orchidectomy. 3. The concentration of testosterone in serum began to decrease gradually after orchidectomy showed lowest level $1.68{\pm}0.61pg/ml$ at 50days after orchidectomy while it was $5.82{\pm}0.39pg/ml$ in control group. 4. The histological findings of thymus after orchidectomy were marked increase of thymic lymphocyte, disapearance of fat cells and appearance of mitotic figures.
초고압초음파분해법을 이용한 축산물내 미량금속 잔류분석을 위한 시료전처리 방법

Simple and rapid sample preparation method for trace elements in foods of animal origin using ultra high pressure microwave digestion system (UHP/MDS) and inductively coupled plasma atomic emission spectrometer (ICP/AES) were developed. 1. For the digestion of sample using UHP-MDS, 20% nitric acid (v/v) was the most suitable solvent for the determination of trace elements in foods of animal origin. 2. The optimal digestion conditions for UHP-MDS were as follows: final temperature $180^{\circ}C$, final pressure 400 PSI, and magnetic power 900 W in the solid sample. For the liquid sample final temperature $170^{\circ}C$, final pressure 300 PSI and magnetic power 700 W were optimal conditions. 3. As result of interlaboratory test, the average recovery rate of the for solid sample were 88.3~99.1% for As, 82.4~93.3% for Cd, 89.2~101.2% for Hg and 86.5~93.8% for Pb, respectively. In liquid sample, it were 87.0~96.8% for As, 80.9~96.6% for Cd, 87.5~91.2% for Hg and 91.4~95.5% for Pb, respectively. 4. The average coefficient variation rate were 3.3~15.9% for solid sample and 2.9~10.8% for liquid sample.

The present studies were conducted to monitor radioactive contamination in dairy products imported from 16 countries located in the East Europe which were affected by the Chernobyl nuclear accident. The 556 samples such as butter, cheese, ice cream, whey protein and hydrolysed milk protein products were collected randomly and determined from 1999 to 2002. All sample were below the Koeran and CODEX maximum tolerance level of radioactivity for $^{131}I$, $^{134}Cs$ and $^{137}Cs$.

The contents of histidine dipeptides, a metabolic products of muscle protein, were investigated to compare muscle composition among the 9 domestic animals including cattle. In major domestic animals, analyzed the effects of age, part and sex of the animals on their muscle composition. Large amounts of carnosine($68.63{\pm}17.41{\mu}mol/g$) were detected in cattle and it was higher than other animals. Whereas the content of anserine showed high level in order of turkey, chickens and duck. The ratio of carnosine and anserine(C/A ratio) was different depending on the animal species. Statistical data indicated that difference among species was significant(p < 0.05). The contents of histidine dipeptides in heated muscle by boiling for 40 minutes at $110^{\circ}C$ was similar to thoes of raw muscle. C/A ratio in heated muscle was not different from that of raw muscle, indicating that no change has been made after heating process. The contents of carnosine and anserine were different according to the parts of their muscle. Especially chuck of the mammalian showed extremely low level of histidine dipeptides compared with other parts, while C/A ratio maintained certain level regardless of age, part, sex. Therefore, based on the content of histidine dipeptides, could be found the difference of muscle composition among the species. Also C/A ratio of horse, pig, cattle, duck, sheep and turkey were characteristic respectively.
