        HPLC에 의한 목초액 중의 유기산, 페놀 및 벤조피렌의 분석법 개발

        The organic acids (formic acid, acetic acid and propionic acid), phenol and benzopyrene in wood vinegar were determine by HPLC. An $Atlantis^{TM}dC_{18}$ column with a acetonitrile: 0.1% phosphoric acid (5: 95, organic acids), acetonitrile: water (10 : 90, phenol), 100% acetonitrile (benzopyrene) as a mobile phase was used. Retention time of acetic acid, formic acid, propionic acid, phenol and benzopyrene was 4.77, 3.73, 9.08, 30.97 and 6.10 min, respectively. The calibration curves of organic acids, phenol, benzopyrene were linear over the concentration range of $30{\sim}500,\;60{\sim}1000$, and $3{\sim}50\;{\mu}g/ml$ with correlation coefficient of above 0.999. The limit of detection (LOD) and limit of quantitation (LOQ) of acetic acid, formic acid, propionic acid, phenol, and benzopyrene was 1.71 and 5.19, 1.11 and 3.35, 4.87 and 14.74, 6.45 and 19.55, 0.08 and $0.24\;{\mu}g/ml$, respectively. The coefficients of variation for intra- and inter-day assay were $0.21{\sim}4.14$ and $0.07{\sim}1.19%$, respectively and the precision was $95{\sim}115%$. From this study, we suggest that HPLC is suitable method for the determination of organics in wood vinegar and could be applied to quality control of wood vinegar.

        가래나무 추출물의 미백효과

        본 연구에서는 가래나무 추출물의 항산화 효과와 티로시나제 활성 저해, 멜라닌 생성 및 티로시나제 합성 억제 효과를 측정하고 이 추출물을 함유하는 제품의 임상시험을 실시하여 미백 화장품 소재로서의 개발 가능성을 관찰하였다. 가래나무 추출물은 농도 의존적으로 DPPH radical 과 초산소 음이온 라디칼을 소거하였고, SC50 값은 각각 20 μg/mL 및 25 μg/mL 이었다. B16/BL6 멜라노마 세포를 이용하여 세포내 티로시나제 활성, 멜라닌 생성 및 티로시나제 합성 억제 효과를 측정한 결과 농도 의존적으로 티로시나제의 활성 및 멜라닌생성을 억제하였으며, 티로시나제 단백질의 합성도 감소시켰다. 또한, 임상시험을 통하여 가래나무 추출물을 함유한 화장품이 대조 제품에 비해 통계적으로 유의한 피부 미백 효과가 있음을 확인하였다. 이상의 결과를 종합해 볼 때 가래나무 추출물은 색소침착 방지와 개선에 효과있는 새로운 미백 원료로 개발할 수 있을 것으로 사료된다. Juglans mandshurica belongs to the family Juglandaceae is known to contain a wide range of pharmacological activities including anti-cancer, anti-inflammation, astringent, and anti-human immunodeficiency virus-type 1 (HIV-1). Melanogenesis refers to the biosynthesis of melanin pigment in melanocytes. In this study, to investigate the whitening activity of the extracts from Juglans mandshurica, we measured effects on a tyrosinase activity, a melanogenesis, and a tyrosinase synthesis in the B16/BL6 melanoma cells and an antioxidant activity. The extracts significantly scavenged a 1,1-diphenyl-2-picrylhydrazyl (DPPH) and a superoxide anion radicals in a dose-dependent manner with a SC50 value of 20 μg/mL and 25 μg/mL, respectively. Also, the tyrosinase activity and melanogenesis were significantly inhibited by the extracts. Furthermore, the synthesis of tyrosinase protein was ignificantly decreased by the extracts in enzyme-linked immunosorbent assay. Double blind study on the clinical efficacy of a cream containing 2% of the extracts showed that the extracts have a significant skin whitening effect. Therefore, this study demonstrates that the extracts from Juglans mandshurica may be useful as a potential agent for skin whitening.

        경피제제로서 수종의 플루비프로펜 Vehicle 과 O/W 마이크로에멀젼의 평가

        N/A In order to reduce systemic side effects following administration, flurbiprofen was formulated as O/W microemulsion consisting of the surfactant, oil phase and aqueous phase. Particle size distribution, apparent viscosity, solubility and skin permeation of flurbiprofen in various vehicles and microemulsion were evaluated. The domain of O/W microemulsion`s phase diagram had difference between oil types and the area of O/W microemulsion was wide distributed by adding to PG and cosurfactant than that of water alone. As increasing 10, 15 and 20% of Brij 97 content and 1, 2.5, 5% of oil content, the solubility of flurbiprofen in O/W microemulsions and various vehicles was 400∼1,000 and 10∼500 times higher than that of control. Also, apparent viscosity of soybean oil microemulsions was higher than that of IPM microemulsions and that of vehicle were increased as increasing vehicle content. Since skin permeation of flurbiprofen decreased as increasing viscosity, in each vehicle, it was not affected 2% β-CD and decreased as increasing PG content and to 2, 5 and 10% of HP-β-CD. In O/W microemulsion, 5% soybean oil, 20% Brij 97 and 75% water(A-1) with high viscosity showed low skin penetration.

        콜라겐 합성과 MMP-1 발현에 대한 생물전환 지실 추출물의 효과

        Ponciri fructus, the unripe fruits of Poncirus trifoliata, are widely used in oriental traditional medicine as a remedy for inflammation, gastritis, emesis, digestive ulcers, allergy, and dysentery. To study the anti-wrinkle effects of Ponciri fructus extract (PFE) containing flavanone glycosides, PFE was fermented with Ganoderma lucidum mycelia and its biological activities were investigated. In Ponciri fructus extracts fermented with G. lucidum (G-PFE), polyphenol content was 1,021.00±0.50 μg/mL and flavonoid content was 589.41±0.21 μg/mL. G-PFE was found to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and superoxide anion radical by a dose dependent manner, respectively. G-PFE showed higher antioxidant activity than that of PFE. In addition, the photoprotective properties of G-PFE was tested in human dermal fibroblasts (HDF) exposed to UVA radiation. G-PFE inhibited the activity of matrix metalloproteinase-1 (MMP-1) and showed a dose dependent decrease in the expression level of MMP-1. G-PFE also increased collagen biosynthesis in HDF. These results demonstrate that G-PFE could be useful as a potential cosmetic ingredient for anti-wrinkle.

        젠트로 $F^{(R)}$의 복부지방에 미치는 영향

        The participants were recruited 77 healthy adult persons aged between 20 and 50 who have BMI above $25kg/m^2$ in this study. All subjects were randomly assigned to the Gentro $F^{(R)}$ drinking group and non-drinking group. We were investigated about abdomen fat decreasing effect of Gentro $F^{(R)}$, distillate of pepper. Total fat area (TFA), subcutaeneous fat area (SFA) and visceral fat area (VFA) and subcutaneous fat area/visceral fat area ratio (SVR) has been assessed by obesity index (BMI, body fat percent, waist circumference), CT scan taken on the $L4{\sim}5$ position, umbilicus level and blood analysis evaluated during 3 month. The obesity indexes were a little decreased in two groups. However, the waist circumference (WC) was decreased about 5% in drinking group after 3 month and there was significant difference in the change 2 and 3 between two group. TFA and VFA were significantly decreased in the drinking group compare to the non-drinking group (p<0.05) and in umbilicus and $L4{\sim}5$ position, were $370.33{\pm}92.30,\;380.35{\pm}97.64\;and\;114.90{\pm}44.91,\;101.99{\pm}41.24$, respectively. These result means that Gentro $F^{(R)}$ is effective on abdomen fat decreasing. Total cholesterol were decreased without difference in both groups. Lipid factors (TG, HDL- and LDL-cholesterol) were decreased without significance. The mea surement of SVR taken on the $L4{\sim}5$ position were more significantly correlated with obesity index that BMI was 0.787, body fat percentage was 0.754 than on the umbilicus level. The TFA and VSA was correlated with ALP, ASP, Albumin, Insulin and the SVR was correlated with the indexes of liver function and lipid factor found in the blood. Therefore, it is conclude that Gentro $F^{(R)}$ drinking leads to a decrease in abdominal obesity by reducing waist circumference and visceral fat area.

      • 분포형 모델을 이용한 유역내 이동강우(MOVING STORM)의 유출해석(1) -모델의 개발-

        유역내 공간적 및 시간적 분포특성을 가진 이동강우를 해석하기 위하여 분포형 모델을 개발하였으며 이 유역모델은 지표면 흐름과 하천망 흐름으로 나누어 유출을 해석하였다. 지표면의 흐름은 2차원의 연속방정식과 운동량 방정식을 이요하였는데 kinematic 이론을 도입하여 운동량 방정식을 간략화 하였으며 하천망 흐름은 하천망을 일체로 하여 1차원의 연속방정식과 운동량방정식들을 이용하였다. 기본방정식들의 수치해석법으로 지표면의 흐름은 유한요소법을 이용하였으며 하천망에 대한 해석은 음해법의 유한차분법을 이용하였다. 모델은 특히 이동강우에 있어서 중요한 특색인 공간적 및 시간적 특성을 효과적으로 해석할 수 있도록 개발되었다. 또한 모델은 구성된 행렬의 특징을 이용하였는데, 지표면 유출모델은 Gauss 소거법을 이용하여 그해를 구하였으며 하천망 해석은 double sweep 방법을 적용하기 위한 여러 종류의 순환계수방정식을 제안하였고 이를 이용하여 그 해를 구하였다. In this paper for simulating spatially and temporally varied moving storm in a watershed a distributed model was developed. The model is conducted by two major flow simulations which overland flow simulation and channel network flow simulation. Two dimensional continuity equation and momentum equation of kinematic approximation are used in the overland flow simulation. On the other hand, in the channel networks simulation two types of governing equations which are one dimensional continuity and momentum equations between two adjacent sections in a channel, and continuity and energy equations at a channel junction are applied. The finite element formulations were used in the overland flow simulation and the implicit finite difference formulations were used in the channel network simulation. The finite element formulations for the overland flow are analyzed by the Gauss elimination method and the finite difference formulations for the channel network flow are analyzed by the double sweep method having advantages of computational speed and reduced computer storages. Several recurrent coefficient equations for channel network simulation are suggested in the paper.
