      • cDNA Cloning and Expression of a Human Interferon-$\gamma$ in E. coli

        인간의 peripheralblood lymphocyte에서 분리된 poly (A)$^+mRNA$를 사용하여 ${\lambda}gt$ 10 cDNA library를 제조하였으며, 화학적으로 합성한 oligonucleotide를 이용하여, 이 cDNA library로부터 IFN-$\gamma$ 유전자를 갖는 DNA을 분리했다. 분리된 clone 가운데 하나인 pBK04는 IFN-$\gamma$의 전체 염기서열을 갖고 있었다. IFN-$\gamma$를 E. coli내에서 발현시키기 위해 lpp promotor를 함유하고 있 는 pINIIIA3 vector를 사용하여 IFN-$\gamma$를 발현시킬 수 있는 재조합 plasmid ($pKS{\gamma}3B$)를 제조하였다. $pKS{\gamma}3B$로써 형질 변환된 bacterial cell을 antiviral activity에 의해 역가 분석을 해본 결과, 높은 정도로 IFN-$\gamma$를 발현시켰으며 이 IFN-$\gamma$는 천연의 IFN-$\gamma$에 대하여 특이성이 있는 monoclonal antibody에 의해서도 인지되었다. High copy number를 갖는 expression vector를 제조 (pHK-3) 하였으며, 이 high copy number vector를 사용하므로써 IFN-$\gamma$의 생성이 증가되는 사실을 확인하였다. A ${\lambda}gt10$ cDNA library constructed with poly $A^+$ mRNA from human peripheral blood lymphocytes was screened by a synthetic oligonucleotide having IFN-$\gamma$ sequence. One of the clones (pBK04) which hybridized to the oligomer was confirmed to have the whole IFN-$\gamma$ coding sequence. The eDNA insert was used to express IFN-$\gamma$ in E. coli under the control of lpp promoter employing pINIIIA3 vector. Cells harboring the recombinant plasmid ($pKS{\gamma}3B$) expressed high level of IFN-$\gamma$ assayed by antiviral activity, The molecule was also recognized by monoclonal antibody specific to a natural IFN-$\gamma$. An expression vector with high copy number (pHK-3) was constructed and was successfully used for the improvement of the expression of IFN-$\gamma$.

        단부 파쇄 후 수직철근 보강공법이 적용된 2층 철근콘크리트 구조벽체의 휨 거동 특성

        The purpose of this study is to evaluate the flexural behavior of flexural retrofitted shear walls connected to slabs retrofitted with vertical rebar after wall end crushing. The main variable was the application of the flexural retrofit method. The evaluation of the effect of the shear studs when the retrofit method was applied was intentionally constructed with 60% of the actual required number. As a result of this experiment, it was possible to confirm the strength increase of about 56% with the flexural retrofit. As suggested in the previous study, the retrofitted wall and the existing wall exhibited partial composite behavior, which resulted in the deformation of the slab and the phenomenon of the concrete covering falling off. Double curvature appeared as a strain on the upper part of the wall when it was reversed. The part where the moment was reversed appeared to be the upper part of the wall and not the center. It was confirmed that the damage to the slab was greater when the joint section of the existing wall and the new wall were subjected to tension. Additionally, it was judged that this should be considered when designing the retrofit of the slab and the shear wall.

        건설현장의 작업성 향상을 위한 기초적 연구 - 이동식 비계의 연구개발 동향 분석을 중심으로 -

        건설업에서 비계는 다른 공사와 더불어 공사장 안팎의 고소작업에 널리 사용된다. 이동식 비계는 조립과 해체가 쉽고 이동이 간편하지만, ‘전도’, ‘추락’ 등의 안전사고가 발생한다는 결점이 있다. 본 연구는 작업성을 향상시킨 이동식 비계의 개발을 위한 기초적 연구로써, 현재까지 보고된 비계 관련 연구 동향을 정리하고 작업성 관점에서 비계발판에 대한 분석을 실시하였다. 비계를 사용함에 있어 반복적인 작업을 줄이고 세부적인 설치를 간편화하게 된다면, 작업성을 증대시킬 수 있다. 숙련도와 관계없이 안전사고가 발생할 가능성이 있기 때문에 안전성을 중심으로 분석하였다. 그러나 작업성과 안전성을 향상시키기 위한 기술개발을 위해서는 높은 비용이 측정되며, 이를 보완하기 위한 비계 자재의 재사용 또는 재활용을 통한 비용감소가 필요할 것으로 판단된다. In the construction industry, scaffolding, along with other construction works, is widely used for high-rise work inside and outside the construction site. Mobile scaffolding is easy to assemble and dismantle and move, but it has the disadvantage of safety accidents such as “conducting” and “falling.” This study is a basic study for the development of mobile scaffolding that improved workability, and the research trends related to scaffolding reported so far were summarized and the analysis of scaffolding was conducted from the perspective of workability. If repetitive tasks are reduced and detailed installation is simplified in using scaffolding, workability can be increased. Safety was analyzed mainly because there is a possibility of safety accidents regardless of proficiency. However, high costs are measured to develop technologies to improve workability and safety, and it is judged that costs should be reduced through reuse or recycling of scaffolding materials to supplement them.

        The purpose of this study is to increase the economic feasibility and workability of metal windows, which are materials of curtain wall construction methods used for high-rise buildings in large cities, by utilizing carbon fibers. Based on structural and economic aspects, the carbon fiber reinforcing windows and steel reinforcing windows were evaluated after comparative analysis. Carbon fiber reinforcement windows were assessed to be more stable than steel reinforcement windows by 16%, cost reduction by 15%, and weight reduction by 23%. Therefore, it was evaluated that carbon fiber reinforcement windows are superior to steel reinforcement windows in economic feasibility and construction.

        사례분석을 활용한 시공단계 BIM 인력 투입 성과 분석

        BIM is effective to improve the labor productivity of construction participants . From this point of view, it is important to analyze the outcome related with BIM Labor which covers most of the BIM investment costs. This research focuses on BIM RFI which is one of the major task of the BIM labor and analyze the outcomes. In addition, this research was quantitatively analyzed by the standby time and related cost caused by BIM labor, which affect the results of the project participants. To this end, analytical standby queue model was utilized to analyze the labor focusing on micros TASK. 11 projects were selected to analyze the results of BIM labor and RFI that the project participants requested to the BIM labor was collected. Through this, it collected variables for analyzing results, and Finally, we pulled out 4 projects for analysis. In this study, the basic results analysis of RFI processing of the BIM labor, the probabilistic analysis of BIM labor service status, and the economic analysis of BIM labor optimal inputs were performed by using the research model presented. The results of this study can be utilized to formulate the optimal strategy for BIM labor inputs(e.g. number of employees, level, time point, etc.) of the construction phase. Moreover, it can contribute to ensuring the credibility of the BIM ROI results by presenting the cost of BIM services in BIM ROI analysis and the standby cost of project participants.

        소규모 건축공사의 BIM 정보 활용을 위한 프로세스 제안 - 철골공사 중심으로 -

        The current study focused on the utilization of building information modeling (BIM) data in steel-frame structures, which help to reduce project durations because they employ prefabricated structural members that are assembled on-site. In addition, a business process model was proposed using BIM data collected during the preconstruction, structural steel fabrication, and on-site construction phases of an actual steel-frame project. The ultimate expectation is that BIM data support at each phase, as well as the increased understanding among project participants, will result in an increase in project management productivity. The results from the current study are summarized as follows: To implement a BIM capable of application to steel-frame projects and data utilization, existing theories were studied to develop the construction project steps, both generally into the preconstruction (A1), steel fabrication (A2), and on-site construction phases, (A3) and specifically into 19 BIM-applicable phases. Based on the derived BIM-applicable phases, the model elements of the BIM object were identified, and the shortcomings of existing steel-frame projects were ameliorated, resulting in an improved data flow model. Moreover, for the proposed BIM data flow to progress efficiently, the BIM specialist needs to be well-acquainted with the phase-specific three-dimensional (3D) model output, and the infrastructure to construct an error-free 3D model must be provided. Based on the actual construction example, the BIM data utilized steel-frame projects - via production reports, clash checks, two-dimensional (2D) drawings, four-dimensional (4D) simulations, and 3D scanning - to make cooperation and communication among participants easier.

      • Pharmacokinetic Study of Recombinant Human Interferon Alpha A in Rats

        Recombinant human interferon-${\alpha}A$ (인터페론)를 rat에 정맥주사한 다음의 혈청 및 뇨중 인터페론 역가를 정량하여 약물체내속도론을 이용하여 해석하였다. 정맥주사시 2가지 $9.0{\times}10^6\;IU/ml$ 및 $3.6{\times}10^7\;IU/ml$)로 다른 용량을 투여하여 보았으나 용량에 따른 속도론 파라메타의 유의성있는 차이 (p>0.05)가 나타나지 않았다. 이로부터 이정도의 용량에서는 체내동태에 용량의존성 (포화과정)이 나타나지 않는 것으로 생각되었다. 이 인터페론의 체내동태는 저자 등이 이미 보고한 바 있는 Namalva 인터페론의 체내동태와 유사하였으나 문헌에 있는 베타인터페론과는 상이하였다. 이 인터페론의 뇨중 배설속도와 혈중농도의 상관관계가 직선성을 보이는 점으로부터 비록 인터페론이 신장에서 여파, 분비, 재흡수 및 분해, 대사라는 복잡한 과정을 거쳐 뇨중에 배설된다. 하지만 이 연구에서와 같은 혈중농도 범위내에서는 각 과정에 포화현상이 나타나지 않았다는 (또는 나타나도 무시할 수 있는 정도라는) 사실을 알 수 있었다. 신클리어란스($CL_r$)가 전신클리어란스($CL_s$)의 0.1%에 불과한 것은 인터페론이 신장에서 상당량 catabolism을 받기 때문이라고 생각된다. Pharmacokinetic behavior of recombinant human interferon alpha rIFN-${\alpha}A$ was studied in the rat. Serum concentrations of rIFN-${\alpha}A$ after intravenous administration could be explained by two - compartment open model. Pharmacokinetic parameters of f recombinant human interferon alpha administered intravenously at two different doses $9.0{\times}10^6$ and $3.6{\times}10^7\;IU/kg$) were calculated by fitting the observed data to the nonlinear least square program. There were no significant differences in pharmacokinetic parameters such as systemic clearance (CLs), apparent distribution volume at steady state ($Vd_{ss}$), biological half life ($t_{1/2}$) etc. between the two doses. There were also no significant differences between pharmacokinetic parameters of this rIFN-${\alpha}A$ and those of Namalva rIFN-$\alpha$ which were reported previously by us. rIFN-${\alpha}A$ aministered intraveneously had a small distribution volume, which indicates its poor distribution to peripheral or poorly-perfused organs. Renal excretion which might be a sum of filtration, secretion, reabsorption or catabolism in the kidney didn't show any saturation phenomena in the serum concentration range studied $1{\times}10^3-5{\times}10^4\;IU/ml$). Renal clearance accounted less than 0.1% of systemic clearance of rIFN-${\alpha}A$.

        Visual Color Deterioration of the Extract of Lithospermi radix

        자초(紫草)추출물의 외관상 변색에 영향을 주는 온도와 pH에 대해 조사하였다. 변색은 Hunter치(値)의 차이로 표시 할 수 있으며 자초추출물의 Hunter치(値)는 $50^{\circ}C$ 이상에서 변화하기 시작하고 $60^{\circ}C$ 이상에서는 온도와 상관관계를 이루었다. 온도에 따른 Hunter L, a, b, 치(値)의 변이상관계수는 열처리 시간이 증가함에 따라 감소하였으며, 전체변색의 상관계수는 증가하였다. 66.7% 에탄올로 추출한 추출물을 1시간 열처리 할때 Hunter치(値) 변이 상관계수는 L치(値)의 경우 -0.3696, a치(値)의 경우 -0.4124, b치(値)의 경우 -0.2279, 전체 변색에 있어선 0.5983이었다. 이 때 전체변색의 상관계수는 Hunter L, a, b 각각의 상관계수로부터 직접 계산할 수 있었으며 0.5988로서 거의 비슷한 값을 보여주었다. 한편 pH를 낮추면 Hunter a치(値)는 감도하고 L, b치(値)는 약간 증가하였으나 pH를 증가시키면 pH6.5를 전후로 해서 Hunter L, a, b 치(値) 모두 다 급격히 감소하여 외관상 색깔이 좋지 않았다. 자초(紫草)추출물은 pH가 4.0-6.5일 때 외관상 가장 좋은 '선명한 붉은색'을 나타내었다. The effects of temperature on the visual color deterioration of Lithospermi radix were investigated under steady conditions of various pH and temperature. The changes of Hunter L, a, b, and ${\Delta}E$ values related to the color deterioration were sharply increased above $50^{\circ}C$ and the deteriorations were expressed linear relationships with the temperature above $60^{\circ}C$. Linear regression coefficients of Hunter L, a, b values decreased with an increase of heat treatment time, whereas the color difference was increased. The linear regression coefficients of Hunter L, a, b, ${\Delta}E$ values in 66.7% ethanol extract solution of Lithospermi radix for 1 hr were -0.3696, -0.4124, -0.2279, and 0.5983, respectively. Inear regression coefficients in color difference (${\Delta}E$) could be calculated from the coefficients in Hunter L, a, b values. The low pH treatment of extract from pH 6.07 to pH 1.35 led to decrease of Hunter a value and a little increase of Hunter L, b values, but the high pH treatment above pH 6.07 decreased all Hunter values. Particularly, the visual color of the extract of Lithospermiradix was appeared to be faborable at pH 4.0-6.5, which was a bright red color.
