A New Phenolic Aldehyde from the Seeds of Phytolacca americana

Caffeic aldehyde was isolated from the seeds of Phytolacca americana. This is the first reported occurrence of the compound in nature.

Ten saponins named phytolaccoside A∼I and O were separated and shown to contain only D-glucose as sugar except phytolaccoside A which contains only D-xylose. Phytolaccagenic acid was found to be a genin for phytolaccosides A and D, phytolaccagenin for B, E and F, jaligonic acid for G, and esculentic acid for C, H, I and O, respectively. The total saponin fraction exhibited strong anti-inflammatory activity.

Steroid and terpenoid constituents of 3 species of genus Phytolacca have been examined by TLC. All contained α-spinasterol, its glucoside and acylated glucoside, jaligonic acid and its methylester, esculentic acid and its methylester, and 10 phytolaccosides. Jaligonic acid was the major free triterpenoid and phytolaccoside E was the major saponin in all plants. Bayogenin, oleanolic acid, spergulagenic acid, acinosolic acid, hederogenin, and their derivatives contained in other Phytolacca spp. were not found in these plants.

In the course of continuing study on sedative constituents of seeds of Zizyphus vulgaris Lamarck var. spinous (Zizyphus jujuba, Rhamnaceae), new acylated flavone C-glycoside mixture, p-coumaroyl-, feruloyl- and sinapoyl spinosin, was isolated from the ethylacetate soluble fraction of the methanol extract. Column chromatography gave pure 6```-O-feruloyl spinosin, mp 194-197°, [α]<sup>20</sup>_D-45.2, whose structure was determined on the basis of chemical and spectral data. The partial structure of sinapoyl spinosin, mp 198∼204°, [α]<sup>20</sup>_D-40.5°, will also be discussed.

In previous communications, we reported on the isolation and structure elucidation of new lignans, Americanin A and B, from the seeds of Phytolacca americana. In the course of a continuing study of the lignans, we isolated a new compound, Americanin D, C<sub>18</sub>H_(18)O_6 (M^+328), mp 197∼200˚; its structure was determined spectroscopically as follows;

Americanin A is a main lignan component of the seeds of Phytolacca americana(Phytolaccacae). Through the chemical transformation to (-)-silandrin, a well established flavanolignan from Sihybum marianum, the structure of americanin A was settled to be trans-2-(3`, 4`-dihydroxyphenyl)-3-hydroxymethyl-6-(3-oxo-prop-1-enyl-benz-1, 4-dioxane. A new compound, mp 242˚ isolated from mother liquor after separation of americanin A was elucidated as acetonylidene americanin A. This compound is, however, not a genuine con stituent but formed in the plant tissue on extracting with acetone.

When pathogens invade fish, MHC class II and CD74 bind to the Golgi apparatus of antigen-presenting cells and are transcribed, causing an immune response. Through NGS analysis, the molecular characteristics and tissue-specific expression of PmCD74a identified in red sea bream were confirmed. As a result, PmCD74a showed high expression in the gills and intestines, and showed downward expression after pathogen infection, and then upward expression. The results of this study suggest that CD74a will play an important role in the immune process of red sea bream.
