        고등어에서 분리된 부패성 효모

        Microbiological spoilage of marine fish is complex process occurring by bacteria, yeasts and molds. There hove been rare study for saprophytic yeasts although haying enormous numbers of bacteriological studies on the spoilage of marine fish. The 14 genera of yeasts isolated from mackerel (Scomber japonicus) with high frequency of occurrence were Candida sp., Rhodotorula sp., Torulopsis sp., Cryptococcus sp. and Tricosporon sp.. Among these ones Candida lipolytica was identified as the strongest proteolytic yeast, then named Candida lipolytica FM5 (C. lipolytica FM5). C. lipolytica FM5 showed optimum growth at 25℃, pH 7.0 and could grow at 5℃ and in medium containing 10% sodium chloride. To evaluate the saprophytic activity of the selected strain, C. lipolytica FM5 and Pseudomonas fluorescens ATCC 17571 which is one of representative spoilage bacteria were individually inoculated into the sterilized fish muscle homogenates, and then pH changes and volatile basic nitrogen (VBN) values were checked during the storage at various temperatures. According to the experimental results, the productions of VBN by C, lipolytica FMS in the fish muscle homogenates were 50 ㎎-N/100 g at 5℃, 152 ㎎-N/100 g at 15℃ and 379 ㎎-N/100 g at 25℃ for 1 week storage, respectively. Above results were nearly same as in case of Ps. fluorescens ATCC 17571 inoculation. It suggest that saprophytic yeasts also have important role in spoilage of marine fish.

        Candida lipolytica FM5 가 생산하는 균체외 단백분해효소의 특성

        Candida lipolytica (C. lipolytica) FM5 was selected as one of the strong saprophytic yeasts isolated from mackerel (Scomber joponicus). The selected strain could produce extracellular proteolytic enzyme. The effective medium for production of proteolytic enzyme by C. lipolytica FM5 was TPPY broth containing Bacto-tryptone 0.5%, proteose peptone 0.5%, yeast extract 0.25%, NaCl 0.5% and CaCl₂ 0.2%. The pattern of proteolytic enzyme production by C. lipolytica FM5 was the almost same as that of growth curve of the strain. Namely, the enzyme production was begun from the early stage of exponential phase and it was reached the highest at the begining of the stationary phase of the yeast growth. The optimum temperature of the produced proteolytic enzyme was 35℃ and its activity was not significantly changed by pH between 6.5∼9.0 and also it was not significantly affected by several kinds of cations such as Ca^(2+), Cu^(2+) Fe^(2+) and Mg^(2+) but it was affected negatively by some cations such as Zn^(2+), Mn^(2+) and K^+.

        경남 거제만 및 자란만 굴( Crassostrea gigas )의 일반성분, pH, 글리코겐 함량의 월별 변화

        The seasonal variation in the proximate composition, pH, and glycogen contents of oystersCrassostrea gigas collected in Geoje and Jaran Bays on the southern coast of Korea was studied between March 2012 and February 2013. In the Geoje Bay oysters, the moisture content was 77.49-81.50 g/100 g, lipids ranged between 1.22 and 2.47 g/100 g, proteins between 9.46 and 13.11 g/100 g, and ash between 1.88 and 2.58 g/100 g. In the Jaran Bay oysters, the moisture content was 74.22-82.05 g/100 g, lipids comprised 1.32-2.37 g/100 g, proteins 9.19-13.35 g/100 g, and ash 1.96-2.45 g/100 g. The moisture content was highest in October and January in Geoje and Jaran Bay, respectively, and tended to increase from July until September. The highest protein levels occurred in August in both bays, which coincided with the timing of oocyte maturation, and then decreased at the beginning of total spawning. The highest lipid levels occurred in April in Geoje Bay, and February in Jaran Bay. The glycogen content was 0.40-2.28 g/100 g in Geoje Bay, and 0.61-3.53 g/100 g in Jaran Bay, and was highest in February and decreased from March onwards. The lowest glycogen content occurred in September and then increased from October onwards. The pH ranged between 6.29 and 6.48, and 6.32 and 6.59, for Geoje and Jaran Bay, respectively, and was highest in February.

        남해안 패류양식장에서 분리된 대장균(Escherichia coli)의 항균제 내성

        This study evaluated the impact of antimicrobial resistant bacteria produced by inland pollution sources on coastal areas, and investigates antimicrobial resistance profiles of bacteria isolated from marine filter feeders. A total of 107shellfish (short-necked clams Ruditapes philippinarum, ark shells Scapharca broughtonii, and oysters Crassostrea gigas) were collected from the southern coast of Korea, from which 204 Escherichia coli strains were isolated. The antimicrobial susceptibility patterns of isolated strains were analyzed for 15 antimicrobial agents used in Korea for clinical or veterinary therapy. Antimicrobial resistance was present in 44.6% of E. coli isolates against at least one antimicrobial agent. The rate of antimicrobial resistance in the Narodo area was higher than isolates from the Gangjinman area and Kamak Bay. E. coli isolates had a higher rate of resistance against: tetracycline (29.9%), streptomycin (25.5%), and trimethoprim (14.2%). Of 204 isolates, 29 (14.2%) were resistant to multiple antimicrobial agents.
