        Laxative Activity of the Hot-Water Extract Mixture of Hovenia dulcis Thunb. and Phyllostachys pubescens Mazel in Chronic Constipation Model SD Rats

        This study examined the laxative effects of hot-water extracts of Hovenia dulcis Thunb. (HD), Phyllostachys pubescens Mazel (PM), and a 2:8 mixture of both (HP) in two chronic constipation models. For the loperamide-induced constipation model, animals were divided into an untreated group, negative control group (loperamide 4 mg/kg), positive control group (bisacodyl 4 mg/kg) group, and six treatment groups (HP 100 or 400, HD 50 or 100, and PM 100 or 400 mg/kg). For the lowfiber diet-induced constipation model, animals were divided into an untreated group (normal diet), negative control group (low-fiber diet), positive control group (Agio granule, 620 mg/kg), and the same treatment groups. Fecal number, weight, fecal water content, and intestinal transit ratio were higher in the groups treated with HP, HD, and PM than in the groups treated with loperamide or lowfiber diet. Thickness of colon mucosa and muscle layers were increased in the treated groups. Colon tension increased in the HP groups, and [Ca2+]i measurements using fura-2 as an indicator showed that HP inhibits ATP-mediated Ca2+ influx in IEC-18 cells. These results showed that the HP mixture has laxative activity by increased mucin secretion and inducing contractile activity and relaxation. It may be a useful therapeutic strategy for ameliorating in chronic constipation.

        Estrogen Receptor alpha Regulates Dlx3-Mediated Osteoblast Differentiation

        Estrogen receptor  (ER-), which is involved in bone metabolism and breast cancer, has been shown to have transcriptional targets. Dlx3 is essential for the skeletal development and plays an important role in osteoblast differentiation. Various osteogenic stimulators and tran-scription factors can induce the protein expression of Dlx3. However, the regulatory function of ER- in the Dlx3 mediated osteogenic process remains unknown. Therefore, we investigated the regulation of Dlx3 and found that ER- is a positive regulator of Dlx3 transcription in BMP2-induced osteoblast differentiation. We also found that ER- interacts with Dlx3 and increases its transcriptional activity and DNA binding affinity. Furthermore, we demonstrated that the regulation of Dlx3 activity by ER- is independent of the ligand (estradiol) binding domain. These results indicate that Dlx3 is a novel target of ER-, and that ER- regu-lates the osteoblast differentiation through modulation of Dlx3 expression and/or interaction with Dlx3.

        삼채․강황 복합추출물의 기능성 원료 표준화를 위한 HPLC 분석법 검증

        This study was preformed to establish a simple and reliable HPLC/UV analytical method to determine p-coumaric acid contents for the standardization of water extracts of Allium hookeri and Curcuma longa complex as functional health food ingredients. The quantitative HPLC method was optimized using a reversed-phase C18 column at 35℃ with methanol and H2O (30:70, v/v) as the gradient mobile phase at a flow rate of 1.0 mL/min and along with detection wavelength of UV 300 nm. This HPLC/UV analytical method exhibited good specificity and high linearity in the tested range of 1.0–10.0 mg/mL with excellent coefficient of determination (R2) of 1.0. The limit of detection and limit of quantification were 0.08 and 0.24 μg/mL, respectively. In addition, the relative standard deviation values from the intra and inter-day precision measurements were 0.4 and 0.1%, respectively. These results indicate that the established HPLC/UV analytical method is simple, specific, precise, accurate, and reproducible and so could be employed in the quantitative analysis of p-coumaric acid as a functional compound in Allium hookeri Thwaites and Curcuma longa complex.

        맹종죽 열수추출물의 기능성 원료 표준화를 위한 HPLC 분석법 검증

        This study was performed to establish a simple and reliable HPLC/UV analytical method for determination of isoorientin for the standardization of the water extract of Phyllostachys heterocyacla as a health functional food ingredient. The quantitative analytical method of isoorientin was optimized by HPLC analytical using reverse-phase C<SUB>-18</SUB> column at 35℃ with methanol and H₂O (30:70, v/v) as a gradient mobile phase at a flow rate of 1.0 mL/min and detection wavelength of UV 254 nm. This HPLC/UV analytical method showed good specificity and high linearity in the tested range of 0.010625–0.17 mg/ mL with excellent coefficient of determination (R²) of 0.999 to 1.0. The limit of detection (LOD) and limit of quantification (LOQ) were 0.0011 μg/mL, 0.0039 μg/mL, respectively. Relative standard deviation (RSD) values of data from intra and interday precision were less than 0.4% and 0.1%, respectively. These results indicate that the established HPLC/UV analytical method is very simple, specific, precise, accurate, and reproducible and thus can be useful for the quantitative analysis of isoorientin as a functional compound in P. heterocyacla water extract.

        이고들빼기 열수 추출물의 면역증진 개선 효과

        본 연구는 최근 COVID-19 팬데믹으로 인해 면역력 향상에 관심도가 증가함에 따라, 비교적 부작용이 적은 천연물에서 면역증진 소재를 탐색하고자 하였다. 먼저, 이고들빼기 열수추출물(CDW)의 비장세포에서 증식능을 확인한 결과, 농도의존적으로 세포증식이 증가하였으며, 생체 내에서 면역반응을 촉진하는 NO 생성능을 확인한 결과 CDW에 의해 농도의존적으로 NO 생성능이 증가하는 것을 확인하였다. T림프구 중에서 보조 T림프구(Th cell)의 활성을 확인하기 위하여 비장세포에 CDW의 사이토카인 생성량을 확인한 결과 CDW 처리군에서 농도별로 IL-2, IL-4 및 IFN-γ 사이토카인 생성량이 증가하는 것을 확인하였다. 그러나 IL-10 생성량에는 유의적 변화가 없었다. 또한 NF-κB의 신호 활성을 간접적으로 알아보기 위하여 세포질의 IκB 농도를 측정한 결과 CDW 처리에 의해 세포질의 IκB는 급격하게 감소하였다. 또 다른 면역 활성을 확인하기 위하여 MAP kinases 발현량을 확인한 결과 CDW에서 p38, ERK, JNK의 인산화를 각각 증가시킴을 알 수 있었다. Balb/c 마우스에 CDW를 농도별로 경구투여한 후 흉선, 비장 무게 측정, 혈청에서의 사이토카인 생성량 측정 및 면역세포 혈액 분석 결과, CDW에 의해 흉선 및 비장의 무게가 증가하였고 사이토카인 생성능이 증가하였으며, 림프구 및 단핵구가 증가하는 것을 확인하였다. 이 결과로부터 CDW가 Th1 세포의 활성화에 관여 함으로써 대식세포 활성을 증가시키고 IL-2 및 IFN-γ 등의 분비를 자극시키며, 비장세포 면역력 증가 활성은 IκB의 분해를 촉진함으로써 NF-κB의 신호 활성을 증가시킬 뿐만아니라 MAP kinases인 ERK, JNK, p38의 신호를 보다 강하게 함으로써 면역증진 효과가 발현됨을 알 수 있었다. 마지막으로 면역 동물모델인 blab/c 마우스에서 CDW는 탐식작용을 하는 단핵구와 대식세포의 활성을 돕는 T림프구를 활성화함으로써 면역증진 효능을 나타내는 것으로 사료되며, 부작용이 적은 천연물 면역증진 소재 개발에 도움이 될 것으로 기대된다. Crepidiastrum denticulatum (Houtt.) Pak & Kawano (also called Crepidiastrum sonchifolium ginseng) is cultivated as a traditional food in South Korea. This study demonstrates that the water extract of Crepidiastrum denticulatum (CDW) induces immune-stimulating responses in splenocytes. Cell viabilities were determined using a WST-8 assay, and CDW at doses of 0.5, 1, or 5 μg/mL significantly increased splenocyte viability. In addition, CDW increased levels of nitric oxide, and pro-inflammatory cytokines (IL-2, IL-4, IL-10, IFN-<SUB>γ</SUB>). These immunostimulatory effects of CDW confirmed that it indirectly participates in the activity of NF-κB by reducing the expression of IκB, and increasing the phosphorylation of MAP kinases (p38, ERK and JNK). Notably, CDW also had an immune-enhancing effect in balb/c mice by activating T lymphocytes, which help activate phagocytic monocytes and macrophages.

        Gamipaidok-san Possesses Antiallergic and Anti-inflammatory Activities

        Gamipaidok-san(GPDS) is the Hyungbangpaidok-san prescription fortified with the additional ingredients known to be effective for dermatitis. So it has been used for atopic dermatitis in the clinic work actually. In this study, we investigated the effects of GPDS on in vitro and in vivo anti-allergic effect on RBL-2H3 rat basophilic leukemia cells and on IgE-induced passive cutaneous anaphylaxis (PCA) in mice. The in vitro anti-inflammatory activity of GPDS in RAW 264.7 cells was investigated. GPDS potently inhibited β-hexosaminidase release from RBL-2H3 and the lgE-mediated PCA reaction in mice. GPDS inhibited LPS-induced NO and PGE2 production in a dose-dependent manner. Furthermore, It also inhibited inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) protein expression in RAW 264.7 cells, and the activation of the transcription factor, NF-κB, in nuclear fraction. The antiallergic action of GPDS may originate from anti-inflammatory activities, and can improve the inflammation caused by allergies.

        Anti-inflammatory Activities of Cheongpyehwadam-tang

        In oriental medicine, Cheongpyehwadam-tang (CHT) has long been used for the cure of inflammatory diseases in the lung and bronchus such as bronchitis, bronchial asthma, pneumonia and tuberculosis. It's use is currently further extended for the treatment of allergic asthma.To investigate the anti-inflammatory effects of CHT, we investigated the effects of CHT on the lipopolysaccharide (LPS)-induced nitric oxide (NO) and pro-inflammatory cytokines (TNF-α, IL-6, and IL-1β) production, and on the level of inducible nitric oxide synthase (iNOS) and proinflammatory cytokines expression in murine macrophage RAW 264.7 cells. CHT alone did not affect NO or pro-inflammatory cytokines production. In contrast, CHT inhibited LPS-induced NO and proinflammatory cytokines and the levels of LPS-induced iNOS and proinflammatory cytokine mRNA in a dose-dependent manner. CHT also inhibited the nuclear factor-kappa B (NF-κB) activation. Taken together, these results suggested that CHT inhibits the production of NO and pro-inflammatory cytokines in RAW 264.7 cells through blockade of NF- κB activation.

        Anti-stress and Sleep-enhancing Effects of Ptecticus tenebrifer Water Extract Through the Regulation of Corticosterone and Melatonin Levels

        P. tenebrifer (PT) belongs to the Diptera order and Stratiomyidae family. Recently, insect industry have been focused as food, animal feed and environmental advantages. γ-aminobutyric acid (GABA) and melatonin have been associated with regulating sleep and depression. GABA is the primary inhibitory neurotransmitter and is synthesized via biotransformation of monosodium glutamate (MSG) to GABA by lactic acid bacteria. In this study, we first used a GABA-enhanced PT extract, wherein GABA was enhanced by feeding MSG to PT. The underlying mechanisms preventing stress and insomnia were investigated in a corticosterone (CORT)-induced endoplasmic reticulum (ER) stress and chronic restraint stress (CRS)-exposed mouse model, as well as in pentobarbital (45 mg/kg)-induced sleep behaviors in mice. In the present study, the GABA peak was detected in high-performance liquid chromatography-evaporative light scattering detector (HPLC-ELSD) analysis and showed in Ptecticus tenebrifer water extract (PTW) but not in non-PTW extract. The results showed that PTW and Ptecticus tenebrifer with 70% ethanol extract (PTE) exerted neuroprotective effects by protecting against CORT-induced downregulation of phosphorylated extracellular signal-regulated kinase 1/2 (ERK1/2) and cAMP-response element binding protein (CREB) expression. In addition, PTW (300 mg/kg) significantly reduced CORT levels in CRS-exposed mice. Furthermore, PTW (100 and 300 mg/kg) significantly reduced sleep latency and increased total sleep duration in pentobarbital (45 mg/kg)-induced sleeping behaviors, which was related to serum melatonin levels. In conclusion, our results suggest that PTW exerts anti-stress and sleep-enhancing effects by regulating serum CORT and melatonin levels.
