Paraquat(PQ)에 의한 산화적손상에 대한 방어기전을 알아보기 위해, 흰쥐에서 PQ(20㎎/㎏)만 투여하거나 PQ투여후 reduced glutathione(50㎎/㎏)을 투여하고 신장의 superoxide dismutase(SOD)와 glutathione peroxidase(GPX)의 활성과 과산화지질양을 측정하였다. SOD의 활성은 PQ투여후 2일에 대조군에 비해 65% 증가된 치를 보였다가 이후 감소하여 4일에는 30% 증가된 치를 보였다가 이후 감소하여 4일에는 30% 증가된 치를 나타내었으며, GPX의 활성은 1일 후에 50% 감소된 치를 보인 후 40일까지 계속 회복되지 않았다. 그러나 과산화지질양은 전 실험기간중 변동을 보이지 않았다. 그리고 GSH의 투여는 의의있는 효과를 나타내지 않았다. 이 결과들로 미루어 보아 SOD는 PQ유발 상해에 대해 방어효과를 나타낸다고 할 수 있다. 또 과산화물분해에 관여하는 GPX활성이 낮은 때에도 과산화지질양의 변동이 없다는 것은 어떤 기전에 의해 지질의 과산화 반응이 잘 조절된다는 것을 시사하는 것으로 생각되어 이 기전에 대한 연구가 더 필요할 것으로 사료된다. In order to study the mechanisms of defense against paraquat(PQ) induced oxidative injury, activities of the superoxide dismutase(SOD) and the glutathione peroxidase(GPX) and contents of lipid peroxides were measured in rat kidney after injection of paraquat(20㎎/㎏) alone or followed by reduced glutathione(50㎎/㎏). SOD activity revealed 65%-increased level after 2 days of PQ injection and thereafter, was decreased to show 30%-increased level after 4 day as compared to non-treated control group, but GPX activity exhibited 50%-decreased level after 1 day and was not restored to control level. Lipid peroxides content, however, did not show any significant changes during the entire peoiod of the experiment. In addition, GSH treatment did not show any influence in SOD and GXP activity and content of lipid peroxides. These results are compatible with the view that SOD has some defensive roles aginst paraquatinduced toxicity. Futhermore, the fact that the lipid peroxides content did not show any changes in spite of the low GPX activity, which is important in peroxide removal, may suggest tht the peroxide generation might be well controlled by certain mechanism.
