오령산이 HgCl<sub>2</sub>에 의한 흰쥐 신부전에 미치는 영향

오령산은 한방이뇨제의 대표적인 약물로서 이 작용이 신부전시에 나타나는 효과를 측정하였다. 오령산의 물엑기스와 알콜엑기스 각각 0.5 g/㎏ 1.0g/㎏을 정상흰쥐와 HgCl₂투여에 의해 신부전을 일으킨 흰쥐에 furosemide 25 ㎎/㎏ 비교 약물로하여 뇨량, 뇨중전해질 혈청중의 urea nitrogen, uric acid, creatinine치를 측정하였다. 뇨량에 있어 정상군, 신부전군에 모두 유의한 상승을 나타내었으며, 전해질에 있어서는 신부전시 정상군에 비해서는 감소하였으나, 대조군에 비해서는 유의한 증가를 나타내었으며 이러한 증가는 오령산에서는 별 영향이 없었으나 furosemide는 초기에는 억제경향을 나타내었다. 혈청중의 urea nitrogen, uric acid, creatinine치에 있어서는 대조군에 비해 유의한 차이를 나타내지 않았다.

Pharmacological studies have been carried out with the methanol extract of the whole plant of Pseudostellaria palibiniana Ohwi (Caryophyllaceae). The results showed that it had stimulation effect on heart, hypotensive actions depending on extract contents, stimulation effect on respiration, contraction on excised intestines, enhancement of tension on excised uterus, antineoplastic activity on Ehrlich carcinoma, liver protective activity against CCl₄ intoxication, writhing and diuretic activities.

From the BuOH extract of Pseudostellaria palibiniana Ohwi (Caryophyllaceae), isovitexin, m.p. 220∼222°, [α]<sup>20</sup>_D+16°(c=0.34, EtOH) was isolated and its structure elucidated on the basis of its spectral data.

Pseudostellaria palibiniana Ohwi (Caryophyilaceae) plant is perennial hero which is growing uprightly in the mountains, and characteristicaily bloom in closed flower. This plant has been used for foods as wild greens or the relief famining foods in the folk and medicinally used for galactogogue, emmenagogue and hemorrhoidal remedy. An attempt was made to identify the flavonoid compound and biological activity. Compound A was isolated by silicagel and sephadex LH-20 column chromatography. Compound A, C_(21)H_(22)O<sub>11</sub> mp 220∼222˚, [α]_(20)^0 -182 (in Methanol) flavone-C-glycoside was islated by silicagel and sephadex LH-20 column chromatography. It was identified to be 6-C-β-D-glucopyranose apigenin through physicochemical approaches compared with literated data and UV, IR, NMR spectra.

Phellinu linteus (Hymenocaetaceae) is a member of Basidiomycotinae, which has been used for the treatment of gastric cancer, Non-insulin-dependent diabetes mellitus, diarrhea, and menstrual irregularity: To investigate the diuretic action of Phellinu linteus, urinary volume, serum parameters and urinary electrolytes in HgCl2- induced acute renal failure rats in vivo were measured. The results showed that its MeOH extract produced significant increases on urinary volume, sodium and potassium excretion, and revealed lowering effects of the abnormally elevated BUN, creatinine and uric acid value in acute renal failure rats.

The effects of Gamdutang extract on the toxicity of lead acetate in mice were examined. Mice were given intraperitoneally daily doses of lead acetate 50㎎/㎏ with Gamdutang Ex. 250 ㎎/㎏, 500 ㎎/㎏ and 1,000 ㎎/㎏ for two weeks. The exposure of Pb(Ac)₂showed the toxicity at all experimental assay such as the gain of body weight, the ratio of organ weight to body weight, serum transaminase activity and cholesterol contents. This toxicity was restored significantly by Gamdutang Ex. administration.

The Youngkaechulgamtang(Y) is composed of four herbal drugs, including Hoelen(H), Cinnamomi Ramulus(C), Atractylodis Rhizoma Alba(A) and Glycyrrhizae Radix(G). In oriental medicine literatures, Youngkaechulgamtang is described to be effective in headache, inflammation, uremia, gastritis, diarrhea, hypertension. To estimate the clinical effectiveness of Youngkaechulgamtang, several pharmacological experiments were carried out. The results are summarized as follows: On $HgCl_2-induced$ renal failure rat, Youngkaechulgamtang showed the significant increase of $Na^+,\;K^+,\;Cl^-$ contents in urine and the significant decrease of creatinine and BUN concentrations in serum. Youngkaechulgamtang was more effective than mixture of 4 ingredients in the urine volume and excretion of electrolytes. In the diuretic effect, Hoelen and Atractylodis Rhizoma Alba were more effective than other drugs.

In oriental medicine literatures, Youngkaechulgamtang is described to be effective in headache, inflammation, uremia, gastritis, diarrhea and hypertension. To estimate the clinical effectiveness of Youngkaechulgamtang, several pharmacological experiments were carried out. The results are summerized as follows; On acetaminophen-induced hepatotoxicity, C+A, Y-G, Y-H, MIX and Y showed the significant elevation of glutathione-S-transferase. But, C+A, Y-G, Y-H, MIX and Y showed the significant suppression of serum aminotransferases. On ANIT-induced cholestasis, U (Ursodesoxycholic acid 50 mg/kg)+$Y_l$ (760 mg/kg) showed the significant increase of bile juice volume. $Y_l,\;Y_2$ (1520 mg/kg), U, $U+Y_l$ showed the remarkable increase of cholic acid. U and $U+Y_l$ showed the significant decrease of total bilirubin. From these results, it is suggest that Y shows liver protective effect against various hepatic injury. Especially, Youngkaechulgamtang was more effective than mixture of 4 ingredients in the elevation of glutathione-S-transferase in acetaminophen-induced hepatotoxicity.

Fruits of Arctium lappa L. (Compositae), which has been used as the antiinflammatory, detoxifying and diuretic agents in the folk remedies, was examined on the in vitro oxidation of human low density lipoprotein (LDL). Several lines of evidence indicate that oxidized LDL (Ox-LDL) may promote atherogenesis. Hence. The role of antioxidants in the prevention of LDL oxidation needs to be determined. The activity of fractions of Arctii Fructus treated with oxidized LDL which was incubated with $16\;{\mu}M$ of $Cu^{2+}$ for metal catalyzed oxidation was investigated. The BuOH fraction (4\;ppm) inhibited the oxidative modification of LDL as evidenced by a decrease in the lipid peroxide content (thiobarbituric acid reacting substances activity), the negative charge of LDL (electrophoretic mobility) and increase of the vitamine E content.
