고구마 괴근을 MeOH 수용액으로 추출하고, 얻어진 추출물을 EtOAc, n-BuOH 및 물로 용매분획하였다. EtOAc 분획에 대하여 column chromatography를 반복하여 4종의 화합물을 분리하였다. 각 화합물의 화학구조를 NMR, MS 등 스펙트럼 데이터의 해석 및 가수분해, 메칠화 및 아세칠화반응을 적용하여, 수지 배당체, simonin I, ${\beta}-sitosterol$ 및 2종의 불포화지방산 화합물로 구명하였다. The MeOH extracts obtained from the tuber of Ipomoea batatas Lam. were solvent-fractionated with EtOAc, n-BuOH, and $H_2O$, respectively. From EtOAc fraction four different compounds were isolated through the repeated silica gel column chromatographies. From not only the results of NMR and MS data but also the adaptation of hydrolysis, methylation, and acetylation, the chemical structures of compounds were elucidated as resin glycoside, simonin I, ${\beta}-sitosterol$, and two kinds of unsaturated fatty acids.
두릅 추출물의 기능성 원료 표준화를 위한 HPLC 분석법 검증

개별인정형 건강기능식품 기능성 원료로 개발한 두릅추출물의 표준화를 위해 항고혈압활성 효과를 가지고 있는 기능성분 HE를 지표성분으로 설정하고, HPLC/UV 방법을 이용하여 기능성분 분석법을 확립하며 그에 따른 유효성 검증을 실시하고자 하였다. 유효성 검증 결과, 본 시험법에서 표준용액의 피크유지시간과 두릅추출물의 피크유지시간이 일치하였다. 검량선의 결정 계수(R2)는 0.9999 이상의 높은 직선성을 보여 분석에 적합함을 알 수 있었으며, 검출 한계는 $12.0{\mu}g/mL$, 정량한계는 $36.5{\mu}g/mL$로 두릅 추출물 중의 HE 함량분석을 위한 충분한 한계 수준으로 확인되었다. 정확성에서 $98.8{\pm}0.865-104.7{\pm}2.536%$로 높은 수준의 HE 회수율을 보였으며, 평균 분석오차는 0.013%로서 기준인 10% 이내를 만족하였다. 또한 반복성에서는 평균 $101.5{\pm}0.4%$로 RSD 0.4%를 나타내었고, 실험실내 정밀성에서 RSD 0.2%, 완건성에서 RSD 0.2%를 나타내어 모두 적합함을 확인하였다. 한편, 제주산 두릅추출물 중에는 기능성 지표성분인 HE가 4.88% 함유되어 있는 것으로 확인되었다. 이상의 실험 결과, 두릅추출물의 항고혈압활성 기능성분인 HE의 HPLC/UV 분석방법은 특이성, 직선성, 정확성, 정밀성, 반복성, 완건성 등의 모든 항목에서 validation 기준에 적합한 시험 방법으로 확인되었다. Aralia elata Seemann (AE) has long been used as a folk medicine for the treatment of various diseases including diabetes mellitus, anti-arthritic, and anti-gastric ulcer agent in Korea, Japan, and China. This study was performed to establish a simple and reliable HPLC/UV analytical method for determination of most active anti-hypertensive compound, a 3-O-${\alpha}$-L-rhamnopyranosyl($1{\rightarrow}$2)-${\alpha}$-L-arabinopyranosyl hederagenin 28-O-${\beta}$-D-xylopyranosyl($1{\rightarrow}6$)-${\beta}$-D-glucopyranosylester (HE) for the standardization of the shoot extract of AE as a health functional food ingredient. The quantitative analytical method of HE was optimized by HPLC analysis using reverse-phase C18 column at $40^{\circ}C$ with $H_2O$ and acetonitrile (70:30, v/v) as an isocratic mobile phase at a flow rate of 1.0 mL/min and detection wavelength of UV 205 nm. This HPLC/UV analytical method showed good specificity and high linearity in the tested range of 0.03125-2.0mg/ml with excellent coefficient of determination ($R^2$) of 0.9999. The limit of detection and limit of quantification were $12.0{\mu}g/mL$ and $36.5{\mu}g/mL$, respectively. Relative standard deviation (RSD) values of data from intra- and inter-day precision were less than 0.2% and 0.1%, respectively. These results indicate that the established HPLC/UV analytical method is very simple, specific, precise, accurate, and reproducible and thus can be useful for the quantitative analysis of HE as a functional anti-hypertensive compound in AE extract.

The effect of seventy kinds of medicinal plants on the activities of GABA-metabolizing enzymes as glutamate dehydrogenase I (GDH I), glutamate dehydrogenase II (GDH II), GABA transaminase (GABA-T), succinic semialdehyde dehydrogenase (SSADH) and succinic semialdehyde reductase (SSAR) were estimated. The following plants extracts from Acori graminei Rhizoma, Longnae Arillus, Gastrodiae Herba, Lycii Fructus, Ligusticum officinale, Ferula assafoetida, Corydalis Tuber, Eucommiae Cortex, Zizyphi spinosi Semen activated the activity of GDH I to more than 35%, and the following ones from Visci Ramulus, Ligusticum officinale, Myristicae Semen, Ferulae Resina, Scolopendrae Corpus, Corydalis Tuber, Eucommiae Cortex, Zizyphi spinosi Semen did that of GDH II. The plant extracts from Cynanchi Radix, Astragali Semen, Angelicae dahuricae Radix, Biotae orientalis Folium, Uncariae Ramulus et Uncus, Polygalae Radix, Cynomorii Herba inhibited that of GABA-T to 35% and over, and the following ones from Hyoscyamus niger, Cynanchi Radix, Acori graminei, Caesalpiniae Lignum, Cannabis Semen, Sedum aizoon, Sedum kamtschaticum, Schisandrae Fructus, Lilii Bulbus, Biotae orientalis Folium, Uncariae Ramulus et Uncus, Myristicae Semen, Akebiae Fructus, Cynomorii Herba, Buddleiae Flos, Mucunae Caulis, Zizyphi Fructus, Paeoniae Radix rubra did that of SSADH to 70% and over; the following ones from, Caesalpiniae Lignum, Sedum kamtschaticum, Schisandrae Fructus, Astragali Semen, Angelicae dahuricae Radix, Dioscorea nipponica, Myristicae Semen, Akebiae Fructus, Cynomorii Herba, Scutellariae Radix did that of SSAR.
한국인 다소비 채소의 에탄올 추출물이 LPS 처리된 대식세포에서 NO, TNF-α와 MCP-1 분비에 미치는 영향

Korean dishes, Hansik is characterized by healthful vegetable intake. The purpose of this study was to evaluate the inhibitory effect of commonly consumed vegetables by Koreans on obesity/metabolic disease-related inflammation. Through statistical analysis of the KNHANES database (1st 1998, 5th 2010, 2011) and a literature review, we selected vegetables for study. Among the vegetables, main or sub ingredients of Kimchi were excluded. Samples were prepared using only edible portions and freezedried. After grinding, samples were extracted with ethanol, evaporated and finally lyophilized. The cytotoxicity of samples was determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, at various concentrations that do not affect cell viability. Raw 264.7 macrophages were treated with lipopolysaccharide (LPS) and 11 kinds of samples or positive control (troglitazone) dissolved in dimethyl sulfoxide (DMSO). After 24 hours, nitric oxide (NO), tumor necrosis factor α (TNF-α) and monocyte chemoattractant protein-1 (MCP-1) production were determined. Excepts for young pumpkin and bracken, nine samples effectively reduced NO production compared with control treated with LPS and DMSO. NO levels of five samples (bean sprouts, leeks, eggplant, mugwort, and pumpkin) were similar to that of the positive control. These five samples showed significantly decreased TNF-α or MCP-1 compared to the control group. Our results suggest that consumption of commonly consumed vegetables contributes to partial prevention of obesity and related metabolic syndrome through reduction of NO, TNF-α, and MCP-1 production.
한국인 다소비 채소의 에탄올 추출물이 3T3-L1 지방세포와 HepG2 간장세포의 지질축적, MCP-1과 Adiponectin의 분비에 미치는 영향

The purpose of this study was to evaluate the inhibitory effects of commonly consumed vegetables in Korea on lipid accumulation and production of pro-inflammatory cytokines related to obesity/metabolic syndrome. Using KNHANES (Korea National Health and Nutrition Examination Survey) raw data (1<SUP>st</SUP>; 1998, 5<SUP>th</SUP>; 2010, 2011) and a literature search, we selected vegetables for study. Edible portions of samples were prepared, ethanol-extracted, and then freeze-dried. 3T3-L1 adipocytes and HepG2 hepatocytes cells were used as in vitro models. Lipid accumulation determined by Oil-red O staining showed that all samples except bracken had inhibitory effects on lipid accumulation in 3T3-L1 adipocytes. Especially, crown daisy and mugwort effectively reduced accumulation of lipids, and their inhibition rates were more than 60% of the control group. Young pumpkin, leeks, crown daisy, and mugwort showed significantly decreased MCP-1 levels compared to the control group. However, adiponectin protein level did not increase in the vegetables experimental group. In HepG2 hepatocytes, all samples showed inhibitory effects on lipid accumulation at one of the two concentrations. Although adiponectin protein levels did not increase, MCP-1 protein levels decreased in adipocytes. Further, lipid accumulation in adipocytes and hepatocytes decreased. In conclusion, all samples showed one or more improved obesity/metabolic syndrome indicators. Among them, young pumpkin, leeks, crown daisy, and mugwort were selected as the most effective portions of vegetables based on improvement of obesity/metabolic syndrome-related indicators.

전자 상거래가 활성화되기 위해서 판매자나 구매자에게 더욱 안전하고 편리한 구매 및 판매 수단을 제공해 주어야 한다. 디지털 상품을 위한 판매 시스템으로 미국의 Open Market에서 개발한 시스템이 있다. 이 시스템은 인증 기능은 있으나 지불 뒤 상품의 암호화된 다운로드는 지원하지 않고 있어 해킹의 위험성을 안고 있다. 본 논문에서는 이를 개선하여 지불 뒤 안전한 다운로드 방법을 제안함으로써 상품 브라우징, 주문, 지불, 전송까지 모두 온라인으로 처리할 수 있는 안전한 프로토콜을 설계하고자 한다.

The stochastic variations were analyzed periodicity by autocorrelation, 1ariance spectrum and Fourier series. These time series inc1uded hourly and hour1y mean observations on DO, water temperature and air temperature which measured by automatic recording instrument at Guü from 1, Jan., 1986 to 23 , Feb., 1986, The results of study were as fo11ows: 1. Autocorrelation coef. (1ag time 120) OO( 에= 0.9705), WT( 까 = 0.9890), and AT( 까 = 0.9874) were deeply related. 00 and AT c1early showedr 24-hour periodicities w뻐e WT showed 23-26 hour periodicity. 2. Spectra1 density showed high at 24 hour in e h item and a11 of them showed weak peak at 12 hour. 3. The explained variance, which was a measure of the contribution of periodic function to the original time series, varied high 90.8 - 94,7%. This results showed that water qualities at Guii were affected deterministic components.
