      • 만성 골수성 백혈병 세포주에서 As2O3가 세포주기 및 세포고사에 미치는 영향

        Leukemia arises in hematopoietic progenitor cells and is characterized by impaired or blocked differentiation, uncontrolled proliferation and resistance to apoptosis. Molecular mechanisms underlying cellular functions by As2O3, however, have been poorly investigated. The consensus of several reports is that As2O3 induces apoptosis in leukemia cells by activating genes for apoptosis. The present study aimed to investigate the effects of As2O3 on the cell cycle and its morphological change and a relationship between the caspase-3 and As2O3-induced apoptosis. Caspase-3 is involved in As2O3-induced apoptosis in K562 cells. In this study, to address whether As2O3-induced apoptosis is mediated by caspase-3 activity, the same samples were probed with a specific antibody. The pretreatment of 25 μM Z-VAD-fmk, a specific inhibitor of caspase, decreased As2O3-induced cytotoxicity. And As2O3 significantly increased the percentages of the cells accumulated in the G2/M phase of the cell cycle in a time- and dose-dependent manner. Chromatin condensational changes were observed with Hoechst 33258 staining after treatment of As2O3. It was shown that As2O3-induced apoptosis is controlled through caspase-3 activation. These results may provide a useful rationale for CML treatment.

        Mice에서 Calcein 표지를 이용한 골 변화 관찰

        골은 일생에 걸쳐 지속적인 재형성과장을 거치면서 유지되고 이러한 기전에 대한 연구는 골다공증을 비롯한 골대사 질환의 병태생리와 치료에 있어 큰 진전을 이루고 있다. 특히 생체 내 골형성 및 재생과정을 연구하는데 있어 형광표지자를 이용하는 방법이 널리 알려져 있는데, 그 중 calcein은 칼슘 킬레이터로 골이 새롭게 형성하는 부위에 녹색을 띔으로써 유용한 마커로 사용된다. 그러나 대부분의 골형성 연구에서 실험동물의 경우 표본제작을 할 때 크기가 작고 뼈가 부숴지기 쉬워 rat이나 rabbit을 이용하였으며, mice의 femur를 cross-section해서 관찰한 연구는 거의 없는 실정이다. 그래서 본 연구에서는 어린 mice를 실험동물로 이용하였으며, 생체 내 calcein을 4주간, 8주간 투여한 후 골 형성 변화를 형광현미경으로 관찰한 결과 8주차 쥐에서 4주차보다 진하고 골 형성 간격도 넓게 관찰된 것을 확인 할 수 있었다. Mice는 빠른 시일 내에 결과를 얻을 수 있고 부작용이 적은 장점이 있어서 앞으로 knock-out mice를 이용한 생체 내 실험에 활용하기 적합하다고 생각되며, 골형성 속도 평가 등 다양한 분야에서의 골 형성과 재생연구에 있어 기초 정보를 제공할 것으로 기대한다. In vivo labeling of bone with fluorochromes is a widely used method for assessment of bone formation and remodeling processes. In particular, calcein is used as a marker for identification of bone growth, which is indicated by a green color. Calcein green is a calcium chelator that adheres to regions of mineralizing bone thereby allowing localization of new bone. Bone formation and remodeling in vivo can be assessed by calcium-binding calcein labeling. In this study, changes in the femoral bone of a normal mouse model at both 4 and 8 weeks were evaluated using calcein labeling. Intense deposition of calcium in the bone was observed after application for 8 weeks. A mouse model is suitable for application in in vivo experiments using genetically modified mice, such as knock-out mice, however data regarding femoral cross sectional bone in young mice are limited. The current study confirmed calcein as a useful marker for identification of bone growth, which was indicated by a green color on photomicrographs. This methodological process may provide basic information for interpreting bone formation and regeneration to pharmacologic or genetic manipulation in mice.

        정밀여과막 및 입상활성탄을 이용한 수처리 공정에 따른 박테리아 거동 평가

        경제성장으로 생활수준이 향상됨에 따라 국민건강을 직접적으로 위협하는 수질오염에 대해 세계적으로 관심을 갖게 되었으며, 최근 맛ㆍ냄새 유발물질 발생의 문제가 대두되기 시작했다. 본 연구에서는 G정수장 내의 모형플랜트에서 사용된 원수 및 각 공정에대한 공정수를 물 시료로 채취하여 정밀여과막 공정 및 입상활성탄공정에서 박테리아의 거동에 대해 확인하였고, 또한 입상활성탄의 지속적 사용을 위한 역세척 가동 시 박테리아의 탈리 여부와 종 동정을 실시하였다. 분석 결과, 정밀막여과 공정을 거치는 경우 수계내 존재하는 박테리아가 제거가 되는 것을 확인하였으며, 역세척가동시 β-proteobacterium species, Porphyrobacter dong-haensis, Polaromonas rhizophaerae, Hydrogenophaga species, Pseudonocardia species 등 총 5종의 우점종 박테리아가 나타나는 것을 확인할 수 있었으며, 정밀여과막 공정 이후 UV 공정을 추가처리한 공정 처리수를 활성탄 공정에 유입한 2종의 박테리아는 나타나지 않는 것을 확인함에 따라 생물활성탄 공정에 의한 오염물질제거에서 박테리아 군집의 UV에 대한 민감도가 고려되어야 함을 알 수 있었다. 이를 바탕으로 수처리 공정의 설계에 유용한 지표를 제공하고자 하였다. Economic growth has increased the living standards around the world. Water pollution, in particular, is a public relations issue because it poses a direct threat to everyone…s lives. As of recently, the production of taste and odor (T&O) compounds has been a common problem in the water industry. The adsorption process using granular activated carbon (GAC) has been the most widely used process. The objectives of this study were to evaluate the microorganisms before and after the backwashing of GAC and to identify the species of the microorganisms found. Five dominants microorganisms were confirmed after the microfiltration process from backwashing of GAC, and the dominant bacterial species were found to be β-proteobacterium species, Porphyrobacter donghaensis, Polaromonas rhizophaerae, Hydrogenophaga species, and Pseudonocardia species. However, when UV treatment after microfiltration was performed, Hydrogenophaga species and Psedonocardia species were eliminated. Herein, I conclude that the UV treatment post microfiltration process is more efficient than microfiltration process alone. The findings of this study may provide useful information regarding the management of microfiltration process.

      • 진단검사의학과 검사실의 공간 환경만족도 조사 연구 - 근무자 중심으로 -

        The purpose of this study is to provide the basic guidelines of spatial and physical environment for the planning and design of clinical laboratory by analysing the extent of satisfaction of clinical laboratory scientists. The data for this analysis was gathered from March to May 2005 by the questionnaires of 208 clinical laboratory scientists who work in the 13 hospitals. All the collected data was analyzed by the SPSSWIN program. In this study, the satisfaction measurement tool was composed with 8 items and 5 score scale. The mean score of satisfaction for spatial and physical environment was 2.89 out of 5.0, "noise" was the lowest 2.40, "temperature and moisture of the lab." was 2.72, "lab. area, service area and administration area" was 2.77, "passageway space" was 2.94, circulation of workers was 2.94, "color of finish" was 3.19, "lighting of lab." was the highest 3.39. In conclusion, various factors, noise, temperature and moisture, clinical lab area, were evaluated to moderate dissatisfaction. Noise was especially the first serious problem in clinical lab. Considering the high growth of the number of tests, the planning of the clinical laboratory size should be considered not only to provide the optimal size but how it will correspond to the growth of the number of tests. Therefore the arrangement of each section need to be flexible in arrangement which is inevitable in expansion and reconstruction in the future.

      • 임상병리과 학생들의 임상실습에 관한 의식조사 연구

        This study was conducted to investigate an attitude related to clinical laboratory science students`` clinical practice. Data for this analysis was gathered from March to May, 2004 by the questionnaires of 348 students who are in their third year in 5 colleges and have experienced clinical practice. The students were satisfied with the term of clinical practice (70.4%) and the first factor influencing them in clinical practice was sincerity (45.1%), and the second was basic knowledge (19.8%). In the contents of clinical practice, 53.2% were satisfied but 43.3% of them showed negative responses because their practice was limited and they were simply assistants. 74.1% were satisfied after experiencing clinical practice and the first factor was improvement of understanding and interest in their major (65.9%) and the second was the gain of confidence (20.1%). Among anxieties in the clinical practice, a shortage of self-confidence (47.7%) was the highest. 81.3% of them wanted to maintain medical technologist``s life continuously. It appeared that the more satisfied with their major, the more satisfied with the clinical practice (p<0.05). As a result, the students were satisfied with the clinical practice and fresh experience generally. But they were relatively unsatisfied with the practicality, and connection with the lecture. As a part of efforts to formulate an effective system for clinical practice, it is necessary to establish concrete goals and detail check lists to guide these students among professors, trainers and students.

        임상병리사 업무 범위와 법률적 고찰

        최근 과학과 의료기술의 발전으로 의료행위에 있어 많은 변화를 가져왔으며 환자의 치료에 있어 그 역할이 강조되고 있다. 특히 임상병리사의 경우, 역할과 중요성이 의료과학 분야가 발달함에 따라 증가되고 있으며, 이에 따른 책임감 역시 증가하고 있다. 임상병리사의 전문성과 독립성의 증가로 더 이상 의사의 보조로만 여기지 않고, 의사의 진료와 의료기사 등의 의료 보조행위에 기초하여 현대 의료서비스는 보다 조직화되고 복잡화, 전문화 되고 있다. 그러나 “의료기사 등에 관한 법률”에서는 의료기사는 의사 또는 치과의사의 지도하에서만 면허범위 내의 행위가 가능하도록 규정하고 있다. 그래서 본 연구에서는 각종의료행위에 있어서 임상병리사를 중심으로 하여 “의료기사 등에 관한 법률”에서 나타나는 문제점과 그 중에서도 의료업무, 의료지도에 관한 부분 등에 관해 살펴보았다. 결론으로 “의료기사 등에 관한 법률”에 명시된 의사의 지도에 대한 명확한 해석이 필요하며, 국가면허시험의 자격과 역할을 “의료기사 등에 관한 법률”에 명시하고, 임상병리사의 역할에 대해 보건의료인들과의 소통 또한 필요하며, 독립된 법제정으로 임상병리사들의 영역 확대와 전문성을 확대 강화하는 것이 필요할 것이라고 생각한다. In recent years, medical practice has seen a drastic change due to the rapid, exponential expansion of scientific and medical technologies. Specially, the role of medical technologists (also known as medical laboratory scientists and/or clinical laboratory scientists) are increasing in the development of science of medical technology. As such, their responsibility has also been increasing. Therefore, given their highly specialized knowledge and skills, they are not regarded solely as doctor`s assistants. Their independence and deeper specialization have been increasing, as they perform medial practices under the guidance of doctors or dentists pursuant to the “Act on Medical Service Technologists.” From a legal point of view, medical guidance and scope of work were examined. As a conclusion, the definition of doctor`s superintendency on the “Act on Medical Service Technologists” is required, and the qualification for the Korean license examination and their roles should be stated clearly. Moreover, communications among health professions regarding the roles of medical technologists are necessary to further facilitate clarification of their role. There is a need for independent legislation to expand the field of medical technologists and to strengthen their professionalism.

        임상병리사 교육과정 및 임상실습에 대한 연구

        본 연구는 임상병리사의 3년제 교육과정, 4년제 교육과정, 임상실습현황, 해외 교육과정 및 실습을 조사 분석하여 급변하는 우리나라의 임상병리분야에 대처할 수 있는 정책의 기초 자료를 제공하고자 하였다. 교육커리큘럼의 분석은 3년제 25개 대학 및 4년제 25개 대학의 2017학년도 기준 교육과정을 요청하여 전수 조사하였다. 임상실습은 설문조사를 실시하여 대학 76건, 산업체 1142건을 분석하였다. 교육과정의 분석결과 3년제 대학의 교육과정은 임상병리사 국가시험위주의 교육과정의 편성을 파악할 수 있었으며, 4년제 교육과정은 미래변화를 반영한 과목이 다양하게 개설되어 있었지만 일부 대학에 불과했다. 또한, 자율적인 수강신청으로 전공과목 중 학생들이 어렵게 여기는 과목에 대해서는 수강하지 않고서도 국가시험을 볼 수 있는 문제점이 발견되었다. 임상실습의 경우 다양한 실습기관과 실습시기, 실습학점으로 현실적으로 표준화가 어려운 것이 실정이었다. 실무중심의 교육을 위하여 교내실습교육도 강화되어야 하겠지만, 현장에서의 실무교육 또한 중요하므로 임상실습의 기간과 교육시간에 따른 학점의 편성이 표준화되고, 적절하게 산출되어야 할 필요가 있을 것이다. 임상병리사 직무분석 및 임상현장을 고려하여 현장실무형 교육과정 개정이 필요하며, 이는 임상병리사평가원을 통해 교육과정과 임상실습모델의 표준화 작업이 필요할 것으로 생각된다. This study analyzed the curriculum and clinical practicum of the department of clinical laboratory science in Korea. The 2017 educational curricula of all 3-year and 4-year universities were analyzed. Clinical practice was conducted by a questionnaire. As a result of curriculum analysis, the 3-year curriculum was able to grasp the curriculum that focused on the national examinations of medical technologists, and the 4-year curriculum was open to a variety of subjects reflecting the changes in the future, but only at a few universities. In addition, the autonomous enrollment application made it possible to take a national examination without enrolling in courses that students find difficult in major courses. In the case of clinical practice, it was difficult to standardize in various practical institutes, practice periods, and practice credits. Therefore, it will be necessary to standardize the composition of the credits according to the duration of clinical practice and the duration of education. Moreover, is necessary to revise the curriculum in consideration of medical technologist job analysis and clinical field, and it will be necessary to standardize the curriculum and clinical practice model through the Medical Technologist Evaluation Center.
