Rebamipide, a novel antipeptic ulcer drug, 2-(4-chlorobenzoylamino)-3-[2(1H)-quinolinone-4-y1]-propionic acid, was studied for its inhibitory effect on gastric xanthine oxidase activity and type conversion of the enzyme that has a profound role in free radical generation. Intraperitoneal administration of rebamipide at 60mg/kg body weight reduced gastric mucosal hemorrhagic lesions and lipid peroxidation, which was proportional to the inhibitory effect of rebamipide on alcohol-induced xanthine oxidase-type conversion and enzyme activity. It was also observed that the activity of xanthine oxidase was significantly inhibited by administration of rebamipide at 60 mg/kg body weight, leading to a significant reduction of lipid peroxide content in alochol-treated rats. The results suggest that alcohol-induced gastric mucosal lesions might be, in part, due to the increased activity of xanthine oxidase and type conversion rate of the enzyme and the protective effect of rebamipide on gastric mucosal lesions would result from its ability to protect against oxidative stress on gastric mucosal lesions of alcohol-treated rats.z
The Effect of Hydrogen Peroxide-Treated Metallothionein on the Hepatic Xanthine Oxidase Activity

We investigated the effect of hydrogen peroxide-treated metallothionein on the hepatic xanthine oxidase activity in vitro. When the metallothionein was preincubated with 1mM of hydrogen peroxide, the activity of xanthine oxidase and type conversion were elevated dose-dependently by the addition of metallothionein into the reaction mixture. While increasing the treatment of hydrogen peroxide to the 50μg of metallothionein, the xanthine oxidase activity and type conversion ratio were remarkably elevated dose dependently compared to the control. When cadmium ion was added to the reaction mixture, the increasing pattern of the enzyme activity was similar to the effect of hydrogen peroxide-treated metaliothionein. DTT or penicillamine restored the increasing activity and type conversion of xanthine oxidase by the cadmium ion to the control level.

In order to determine the structure-activity relationships for antioxidative effects of gagaminine, a steroidal alkaloid isolated from the roots of Cynanchum wilfordi (Asclepiadaceae), two derivatives identified as sarcostin and penupogenin were prepared from gagaminine by hydrolysis and reduction. These compounds were evaluated for the inhibitory effects on the aldehyde oxidase activity and on lipid peroxidation in vitro. Furthermore, their effects were compared with those of gagaminine and the related compounds, cinnamic acid and nicotinic acid. The results of this study prove that the cinnamoyl group in the structure of gagaminine is critical in inhibition of the aldehyde oxidase activity while the nicotinoyl group may be necessary for anti-lipid peroxidation of the compound.

This study was done to determine the effect of lead acetate on the activities of the hepatic cytosolic xanthine oxidase and aldehyde oxidase which were well known as oxygen free radical generating enzyme in vitro. Lead ion accelerated the formation of lipid peroxide and the increment of xanthine oxidase(type O) activity and the type conversion ration from xanthine dehydrogenase to xanthine oxidase dose-dependently. But xanthine dehydrogenase(type D) activity was decreased. Aldehyde oxidase activity was not changed by lead ion. These data suggested that lead-induced cellular toxicity may be concerned partially with xanthine oxidase mediated lipid peroxidation.

The effects of metallothionein and cadmium ton on the hepatic aldehyde oxidase activity and brain lipid peroxidation were tested in vitro. The content of brain lipid peroxide at the condition of normal or 300μM Fe(Ⅱ)-induced was remarkably reduced by the addition of metallothionein in the incubation mixture. The induced content of lipid peroxide by cadmium (30μg/ml) lon was reduced by metallothionein (100μg/ml). The activity of aldehyde oxidase was not affected by metallothionein, but cadmium lon (8.38μg/ml) in creased the activity of aldehyde oxidase about 80% compared to the control. The cadmium-induced activity of aldehyde oxidase was restored to the control level by metallothionein or peniclilamine.

Gagaminine, a steroidal alkalold, was isolated for the first time from the root of Cynanchum wilfordi Hemsley (Asclepiadaceae), and lts effects on lipid peroxidation and the activity of aldehyde oxidase (EC. 1.2.3.1) were investigated in vitro. This alkaloid suppressed significantly the formation of lipid peroxides in rat liver tissues and potently inhibited the hepatic aldehyde oxidase activity in a dose-dependent manner, with IC_(50) value of 0.8㎛ (0.5μg/ml). These results indicated that gagaminine is a potent natural antioxidant and may be useful for clinical tests.
Effect of Heat-treated Ceruloplasmin on the Hepatic Xanthine Oxidase Activity and Type Conversion

The effect of ceruloplasmin or copper ion on hepatic xanthine oxidase activity and type conversion was investigated using rat liver in vitro. It was observed that ceruloplasmin increased xanthine oxidase type conversion depending on duration of is storage. Xanthine oxidase (type O) activity and type conversion in incubation mixture was increased by the addition of heated celuroplasmin in a temperature dependent manner. The type conversion of xanthine oxidase induced by heated ceruloplasmin was returned to normal by the treatment with DTT or penicillamine. The effect of copper ion on type conversion of xanthine oxidase was similar to that of heated ceruloplasmin.

Effect of Sex Hormones on Lipid Peroxidation in Rat Liver

The role of sex hormones in hepatic lipid peroxidation, and in hepatic aldehyde oxidase and xanthine oxidase activities were investigated using rat liver homogenates. It was observed that male rat had a significantly greater content of malondialdehyde in liver than female. Among the sex hormones tested, estradiol, one of female hormones, markedly inhibited the formation of lipid peroxides in liver tissues in vitro. Especially, the inhibitory effect of estradiol appeared more remarkably in Fe^(-2)-induced lipid peroxidation. The hepatic xanthine oxidase activity was decreased about 15% by 10^(-6) M estradiol, whereas, the aldehyde oxidase activity was almost completely disappeared at the same concentration of estradiol. It implies that sex differences in lipid peroxidation is attributed to the suppression of free radical generating system by estradiol.

Sex hormones not only regulate external sexual characteristics but several internal biochemical processes. It is well accepted that life-span of female is longer than that of male. Life-span is closely related with aging process in which free radicals are known to be involved. We investigated the effect of testosterone on free radical generating systems and lipid peroxidation based on the sexual difference. Lipid peroxide levels of male and female mouse were increased proportionately with age, especially in male mouse. Increase in enzyme activity of aldehyde oxidase with age was observed in male mouse, while no significant change in enzyme activity was found in female mouse. Enzyme activity of xanthine oxidase also showed similar results. It, however, was not significant statistically. Lipid peroxide level and xanthine oxidase type conversion ratio of male and female mouse liver homogenate incubated at 37℃, increased remarkably in proportion to incubation time, especially in male mouse. Lipid peroxide level and aldehyde oxidase activity were measured in normal male mouse, castrated mouse and testosterone treated-castrated mouse. Castrated mouse group showed decrease in lipid peroxide level and aldehyde oxidase activity compared with normal group. Treatment of castrated mouse with testosterone, however turned the level of lipid peroxide and aldehyde oxidase activity back to normal. From the above results, it might be concluded that testosteron could increase the activities of free radical generating enzymes which would result in the formation of lipid peroxide, consequently leading to aging.
