참돔(Pagrus major)에서 동정된 Lumican의 분자적 특성과 유전자 발현분석

Lumican is a member of the small leucine-rich proteoglycan (SLRP) proteoglycan Class II family. In particular, it plays the role of a regulator of extracellular matrix (ECM) tissue and biological function. It also has a function for tissue repair by regulating collagen fibrin formation in interstitial tissue. In this study, after analyzing the characteristics and expression of lumican in red sea bream, the highest expression was found in the skin, and expression was also confirmed after infection with Edwardsiella piscicida (E. piscicida), Streptococcus iniae (S. iniae), and red sea bream iridovirus (RSIV). As a result, it could be confirmed that lumican has a function of repairing tissue damage and immune response in red sea bream.

Given the increasing importance of Platichthys stellatus in the aquaculture industry, basic research on the immune response is needed. Therefore, we analyzed the molecular biology and expression of the Nuclear factor interleukin 3 regulated protein (NFIL3) gene, which plays an important role in innate immunity. Tissue-specific expression analysis of healthy P. stellatus showed that NFIL3 mRNA had the highest expression in gill, followed by skin and trunk kidney. In the Viral Hemorrhagic Septicemia Virus (VHSV) infection group, the tissues that were significantly expressed after infection were the trunk kidney, spleen, gills, and heart, and the strongest expression was observed in the trunk kidney. In the Streptococcus parauberis PH0710 infection group, high expression was observed in the liver on day 1, and in the intestine and brain on day 7.

Zinc finger protein (ZNF) is widely expressed in eukaryotes and is involved in a number of important biological processes such as cell differentiation, proliferation and apoptosis. ZNF703 is one of the transcription factors that regulates the expression of multiple genes involved in transcription modulation, cell proliferation, cell cycle regulation, and signaling pathway regulation. However, studies on the immune response of ZNF703 after pathogen infection in fish are lacking. We analysed the coding sequence (CDS), genomic structure, and temporal variations in the mRNA transcription of ZNF703 after challenge with viral hemorrhagic septicemia virus (VHSV) and Streptococcus parauberis PH0710 in starry flounder (Platichthys stellatus) (Ps). PsZNF703 was confirmed to have a NLZ1 domain and ZNF C2H2 domain. The strongest expression of PsZNF703 mRNA in healthy starry flounder was observed in the gills. The expression pattern of PsZNF703 mRNA after infection with pathogens was significantly up-regulated in the spleen, liver, and intestine 12 h post-VHSV injection (***p < 0.001). After infection with S. parauberis PH0710, the PsZNF703 mRNA expression pattern was up-regulated in almost all major tissues, whereas it was significantly down-regulated in the gills and heart. Our results indicate that PsZNF703 may be involved in the host immune response to starry flounders.

Red Sea Bream Iridoviral Disease (RSIVD) is a prevalent infectious disease in South Korea fish farms. Detecting the disease early and preventing its spread through continuous surveillance activities are crucial for disease prevention and management. To achieve this, we utilized a virus concentration method in seawater, providing a non-invasive means to indirectly monitor disease progression. We investigated the viral copy and viral shedding ratio in Oplegnathus fasciatus, known for their high susceptibility to red sea bream iridovirus (RSIV), under flow-through conditions. Horizontal transmission was confirmed in RSIV-infected O. fasciatus and uninfected Lateolabrax japonicus and Acanthopagrus schlegelii. In a flow-through conditions, the mortality rate of O. fasciatus increased with rising water temperature, and it was observed that the maximum viral copy was higher under high-density conditions compared to other density conditions. The study also confirmed horizontal transmission between RSIV-infected O. fasciatus and uninfected L. japonicus and A. schlegelii.

Recently, as the demand for ornamental fish has increased due to the expansion of the ornamental fish market, various species are being imported, but the possibility of new species or species that have not been reported is increasing. In particular, there are concerns about problems such as the introduction of diseases and ecosystem disturbance, so the need for systematic quarantine of ornamental fish is increasing. Currently, ornamental fish, as designated quarantine items, are under quarantine in accordance with the Aquatic Organism Disease Control Act. However, due to a diverse range of breeds and dispersed packaging, quarantine takes a long time, and there are concerns that this may lead to death. To solve these problems, it is important to establish a list of ornamental fish breeds and basic research. In this study, we aim to improve the speed and accuracy of quarantine by visually observing and genetically confirming the relationship between the phenotype and genotype of ornamental fish.

The major histocompatibility complex class II-associated invariant chain is a molecule involved in antigen presentation, specifically in the context of T cells, which are effector cells of adaptive immunity. CD74 is a protein that reacts with MHC class II molecules and functions to transmit signals into cells by interacting with macrophage migration inhibitory factor. Red sea bream is one of the major marine aquaculture fish in Korea. Many studies on breeding but molecular studies are markedly lacking. In this study, CD74b was isolated from red sea bream (Pagrus major) and phylogenetic analysis, tissue expression and expression analysis through pathogen artificial infection were performed. As a result, it showed high expression in gills, brain and intestine. In addition, when analyzing Edwardsiella piscicida, Streptococcus iniae, and red sea bream iridovirus, it was confirmed that CD74b has an immune response function. This study characterizes CD74b may play an important role in the immune response of red sea bream following pathogenic infections.

With the increasing fishing population, there is a growing demand for bait, leading to an expansion in the use of baitfish. However, there is a concern that baitfish carrying diseases may introduce infections into domestic water systems. In this study, various samples of baitfish, crustaceans, annelids, and mollusks were analyzed for the presence of 21 diseases registered in the World Organization for Animal Health (WOAH) Aquatic Animal Health Code. The disease analysis results revealed the detection of White spot syndrome viruses (WSSV) and Acute hepatopancreatic necrosis disease (AHPND) in certain shrimp samples. WSSV and VpAHPND were detected 5 out of 10 analysis, with detection rates ranging from 3% to 100%. The study also examined the detection rates of these diseases in case of positive reactions. To assess the infectivity of WSSV, artificial infection experiments were conducted. The results of this research can serve as foundational data for the establishment of preventive measures against diseases associated with baitfish in aquaculture.

Offshore wind power generation is an energy generation field that is rapidly developing owing to the increasing demand for clean energy. However, the physiological response of fish to the underwater noise generated during construction or operation of wind turbines is unclear. We confirmed the effects of sound pressures of 125, 135, 145, and 155 dB/μPa, including 140 dB/μPa (the standard sound pressure for noise damage recognition in South Korea), through serum analysis in rock bream (Oplegnathus fasciatus). High mortality induced by reduced immunity through artificial infection after stimulation was confirmed. These results suggest that rock bream is negatively affected by the noise generated during the construction of offshore wind power plants.
