        능성어, Epinephelus septemfasciatus의 바이러스성 신경괴사증 바이러스의 병원성 연구

        The pathogenicity of the fish nodavirus causing viral nervous necrosis (VNN) of sevenband grouper, Epinephelus septemfasciatus was examined in sevenband grouper and other marine fish by intramuscular injection. Sevenband groupers of 27~104 g in body weight were highly susceptible to the fish nodavirus, but yellowtail (537 g in body weight), red seabream (207 g), rock bream (43 g), flounder (41 g), tiger puffer (27 g) and rockfish (94 g) of the sizes used to this experiment were not. The pathogenicity of the viral agent to the sevenband grouper was high without regard to fish sizes at rearing water temperature over $20^{\circ}C$, but not at $15^{\circ}C$. Therefore, susceptibility of sevenband grouper to the viral agent causing VNN was thought to be water temperature-dependent rather than fish size (age-dependent). Compared to the infectivity of the viral agent to sevenband grouper with artificial infection methods, fish were successfully affected by intramuscular, intraperitoneal, oral, dipping and cohabitation administrations although there were slight differences in mortalities among infection methods. And survival sevenband grouper after infection with the fish nodavirus was resistant to the reinfection for a long time. 능성어의 바이러스성 신경괴사증 바이러스(fish nodavirus)의 병원성을 능성어 및 다른 해산어류를 대상으로 인위감염법에 의해 조사한 결과, 능성어 바이러스성 신경괴사증 바이러스는 능성어에서는 소형어(27 g)부터 대형어(104 g)까지 제사를 일으켜 강한 병원성을 나타냈지만, 실험에 사용한 크기의 방어(537 g), 참돔(207 g), 돌돔(43 g), 넙치(41 g), 자주복(27 g) 및 조피볼락(94 g)에는 폐사를 일으키지 않아 병원성을 나타내지 않았다. 그리고 사육수온별 능성어에 대한 바이러스의 병원성은 수온 $15^{\circ}C$에서는 병원성을 나타내지 않았고, 수온 $20\sim28^{\circ}C$에서는 수온이 높을 수록 병원성을 강하게 나타냈다. 그리고 바이러스는 근육주사, 복강 주사 및 경구감염뿐만 아니라 침지감염과 혼합사육감염에 의해서도 쉽게 능성어에 감염이 되지만, 감염 후 생존한 능성어에 대해서는 바이러스를 재감염시켜도 폐사가 일어나지 않았다.

        동자개 치어의 암모니아 급성 독성

        Juvenile banded catfish(Pseudobagurus fulvidraco, mean length 10.7 ± 0.42 cm and mean weight 15.0 ± 0.23 g) were exposed to varies TAN(total ammonia) concentrations at pH levels of 6.12 ± 0.51, 7.00 ± 0.26 and 8.04 ± 0.07 for 96hrs to check the level of acute toxicity on biofloc technology aquaculture system(BFT). The result showed that cumulative mortalities for juvenile banded catfish at TAN levels 48.95, 55.96, 66.47, and 78.88 mg/L at pH 6.12 ± 0.51 were 0, 30, 30, and 50%, respectively. At pH 7.00 ± 0.26, its mortalities to TAN 5.20, 11.68, 15.31, and 18.31 mg/L were 0, 10, 20, and 70%, respectively and at pH 8.04 ± 0.07, the mortalities to TAN 0.96, 1.49, 2.13, and 3.62 mg/L were 10, 20, 40, and 100%, respectively. Its 96h-LC<SUB>50</SUB> (median lethal concentration, LC<SUB>50</SUB>) at pH 6.12 ± 0.51, 7.00 ± 0.26, and 8.04 ± 0.07 were 78.12, 15.87, and 2.21 mg/L for TAN, and 0.05, 0.10, and 0.14 mg/L for NH₃, respectively, and the acute toxicity for ammonia to juvenile banded catfish increased exponentially with increase of pH.

        동자개 치어의 아질산 급성 독성

        Juvenile banded catfish(Pseudobagurus fulvidraco, mean length 10.4 ± 0.37 cm and mean weight 14.5 ± 0.46 g) were exposed to several nitrite(NO₂-) concentrations for 96h at pH levels of 6.18 ± 0.54, 6.5 ±0.30 and 7.07 ± 0.22. The result showed that cumulative mortalities of fish to NO₂- levels of 12.4, 19.8, 33.9 and 53.6 mg/L at pH levels of 6.18 ± 0.54 were 20, 25, 40 and 85%, respectively. At pH 6.5 ± 0.54, mortalities to NO₂-22.4, 36.4, 45.3 and 63.2 mg/L were 25, 35, 50 and 100%, respectively. At pH 7.07 ± 0.22, mortalities to NO₂- 25.5, 45.7, 56.3 and 66.4 mg/L were 0, 55, 70 and 100%, respectively. The 96h-LC<SUB>50</SUB>(median lethal concentration, LC<SUB>50</SUB>) of fish to several NO₂- concentrations at pH levels of 6.18 ± 0.54, 6.5 ± 0.30 and 7.07 ± 0.22 were assessed in these experiments. 96h-LC<SUB>50</SUB> at pH levels of 6.18 ± 0.54, 6.5 ± 0.30 and 7.07 ± 0.22 were 32.68, 40.49 and 45.85 mg/L, respectively. It indicated that acute toxicity of NO₂- to juvenile banded catfish increased with low levels of pH and lengthening of exposure time to NO₂-. In particular, smaller fish(mean weight 14.5 ± 0.46 g) were more sensitive to NO₂- than larger fish(mean weight 81.7 ± 1.42 g; not published). The 96h-LC<SUB>50</SUB> of juvenile banded catfish to NO₂- would be primary guideline for water quality management in the intensive culture system such as RAS and BFT.

        이리도 바이러스 감염에 의한 양식 돌돔 , Oplegnathus fasciatus 의 대량폐사

        From August to October 1998, over 60% mortality of cultured striped beakperch Oplegnathus fasciatus was occurred in net cages along the southern coast of Korea. Moribund fish showed some clinical signs of lethargic behavior, dark coloration or decoloration, severe gill anemia and enlargement of spleen. Also enlarged basophilic cells showing Feulgen -positive reaction were observed in the tissue section of spleen, kidney, liver and heart of the diseased fish. GF cells inoculated with spleen homogenate of diseased fish produced cytopathic effect of enlarged and rounded cells, therefore the causative virus was isolated from diseased fish. Striped beakperch fingerlings intraperitoneally inoculated with the causative virus (10⁴ TCID_(50)/0.1 ㎖) revealed symptoms similar to those of naturally infected fish and died from 7 to 14 days post injection. Transmission electron microscopy revealed that the causative virus was enveloped icosahedral particle with 120-130 ㎚ in diameter. PCR products of the expected size (500 bp) were amplified with a primer set based on the ATPase gene of RSIV(red sea bream iridovirus) using template DNAs which were extracted from the spleen of diseased fish and GF cells inoculated with the causative virus. According to the analysis of nucleotide sequence of these PCR products, the sequence from ATPase cDNA gene of the causative virus showed 95% homology with that of RSIV. These results indicate that the mass mortality in the cultured striped beakperch was caused by the infection of iridovirus similar to RSIV.

        산천어의 바이러스성 질병에 관한 연구 (1) : 산천어 치어의 병리조직학적 연구

        In Feb., 1990 an epizootic disease to masu salmon fry, Oncorhynchus masou cultured at the hatchery of rainbow trout in Samchok, Kwangwon-do have broken out and induced high mortality over 70%. Externally, the diseased fish showed dark discoloration, abdominal distension with ascites, slight exophthalmus and fecal casts. Internally, the gill and the liver of diseased fish were edematous and pale, and the stomach of moribund fish contained the milkish fluid. Microscopically there was extensive necrosis of the hematopoietic tissue in kidney and spleen, and scattered necrosis of pancreas, liver and lateral muscle. Especially, the necrosis of lamina propria, muscle layer and tela submucosa in the digestive tract known as the typical signs associated with infectious hematopoietic necrosis virus was seen clearly.

        양식 넙치 치어에서 분리한 birnavirus 의 특성

        During 1993 and 1994, some mortalities of flounder(Paralichthy olivaceus) fry were recorded in several fish farms and viruses were isolated from 3 of the farms. Electron microscopic examination revealed that the virus particles were hexagonal and unenveloped with an average diameter of 50 to 55nm. Serological and molecular properties of these isolates were examined. The viral RNA and polypeptides patterns on electrophoresis, as well as neutralization test results, showed that these isolates were birnaviruses and two were closely related to infectious pancreatic necrosis virus(IPNV) serotype AB and one was to IPNV serotype SP. This is the first isolation of birnaviruses from marine fish in Korea

        대하새우로부터 분리한 WSBV 의 게놈서열 분석

        Baculovirus associated with white spot syndrome (WSBV) is the causative agent of a disease with high mortalities and causes severe damage to shrimp cultures. In this study, we analyzed a recombinant clone (E3) obtained from a viral genomic library to characterize the causative agent in diseased shrimp Penaeus chinensis with white spot syndrome. According to the analysis of nucleotide sequence of E3, this clone did not showed considerable sequence homology with those of other known viruses, including baculovirus Autographa californica nuclear polyhedrosis virus (AcNPV), indicating that WSBV is a novel virus causing a serious disease in P. chinensis. Based on the sequence of E3 clone, a pair of PCR primers was designed. After 30 cycles of amplification, a specific product of the expected size was detected only if the total nucleic acids extracted from the diseased shrimp was used as a template DNA, suggesting that this method can be used to diagnose the virus infection in diseased shrimp.

        산천어의 바이러스성 질병에 관한 연구 (2) - 산천어 치어에서 1HNV 분리 -

        In February of 1990, an epizootic disease to masu salmon. Onchorynchus masou cultured at the hatchery of trout in Samchuk. Kwangwondo have broken out and induced heavy mortality. An infectious hematopoietic necrosis virus(IHNV) was isolated from diseased masu salmon fry by the use of fish cell line. CHSE-214. This IHNV isolated from masu salmon was compared with USA isolates of IHNV. SRCV and RB-76 by analysis of virion proteins in sodium dodecyl sulfate poly-acrylamide gel electrophoresis (SDS-PAGE) and neutralization tests with two monoclonal antibodies raised against SRCV(MAb SRCV/A4) and RB-76(MAb RB/B5). In the antigenicity and the size of structural proteins. this IHNV. SCS atrain was smilar to RB-76 belonged to the electropherotype I proposed by Hsu et al.(1986).

        White Spot Baculovirus 의 모하와 중간숙주 감염 및 해수생존

        Infection rates with white spot baculovirus(WSBV) in wild-caught shrimp spawners appeared to be 52% in Penaeus chinensis and 20% in Penaeus japonicus when diagnosed by polymerase chain reaction (PCR). Infection rates of WSBV from inhabitant organisms in shrimp farm showed 37% in Upogebia major and 29% in Calliamassa japonica. The viruses maintained their survivorship in seawater at 4℃, but lost it at 25℃.

        한국 양식 넙치에성 분리된 Iridovirus 의 면역학적 특성 비교에 관한 연구

        In order to determine whether the tumor-inducing iridoviruses and the mortality-associated iridoviruses from cultured marine fish in Korea belong to same type, we compared the immunological characteristics of these viruses. The electrophoretical pattern of structural proteins of the tumor-inducing was different from that of the mortality-associated iridoviruses. The antigenicity of structural proteins of these viruses were identified by Western blotting using two monoclonal antibodies against tumor-inducing iridovirus. Two monoclonal antibodies recognized a 150 kDa structural protein of tumor-inducing iridoviruses showed. However, the structural proteins of the mortality-associated iridoviruses did not react with these monoclonal antibodies. These results demonstrate that the antigenicity of the structural proteins of tumor-inducing iridovirus is different from that of mortality-associated iridovirus, indicating that these two iridoviruses belong to different types.
