Developing new substance for immunosuppressor or immunomodulator from natural products is extremely important in the present biomedicine. In this paper, we focused our efforts on the mitogenicity and immunochemical properties of the two lectins (LCC-I, LCC-II) obtained from marine animal Lunella coronata coreensis. Immunochemical techniques were employed to elucidate the structural and/or functional similarities between the LCC lectins. Molecular weight of the LCC lectins, LCC-I and LCC-II were estimated to be around 60 KD and 66-70 KD, respectively. LCC lectins were mitogens for murine splenic lymphocytes, and the optimum mitogenic doses were 31.25mcg/ml and 3.91mcg/ml, respectively. LCC-II lectin was a good mitogen toward human peripheral lymphocytes at a concentration about 31.25mcg/ml.

The whole body extract of Lunella coronata coreensis agglutinated nonspecifically human and other animal erythrocytes. A new lectin was purified by the following procedures: 0.15 M NaCl extraction, salt fractionation, gel filtration, anionic and cationic ion exchange column chromatographies. Through these purification procedures, specific activity of LCC-I was increased from 276 to 9714.3units/mg, And on polyacrylamide gel electrophoresis, LCC-I exhibited one major band. A molecular weight of LCC-I was assumed to be 20,000 by sodium dodesyl sulfate polyacrylamide gel electrophoresis. The purified lectin was relatively stable at various pH and heat. Among the tested sugars, lactose and lactulose inhibited lectin activity at a concentration of 6.25mM, respectively.

밤고둥의 생리식염수용액 추출물은 사랑 및 각종 동물 적혈구를 비특이적으로 응집시켰으며 림프구도 응집시켰다. 밤고둥의 새로운 렉틴 성분을 생리식염수용액추출, 황화암모늄 침전, 이온교환 크로마토그래피 등의 방법으로 분리 정제하였다. 이는 전기영동상에서 하나의 주된 띠와 다른 몇개의 띠를 나타내었으며 주된 띠에서만 렉틴활성을 나타냈다. 부분 정제된 밤고둥 렉틴은 pH변화에는 비교적 안정하였으나 열에는 불안정하였다. 적혈구 응집은 lactose와 D-galactose의 각각 10mM과 100mM농도에서 저해되었다. 또한 밤고둥 렉틴은 마우스의 림프구에 대한 분열자극효과를 나타냈다. In this paper, we focused our efforts on the mitogenicity and immunochemical properties of the NI lectins (NIA, NIB, NIC), obtained from Neptunea intersculpta. Immunochemical techniques were employed to elucidate the structural and/or functional similarities between the NI lectins. According to these results, NIB and NIC were almost identical and the NIC proved to be homogeneous on immunoelectrophoresis as well as crossed immunoelectrophoresis. But these lectins are turned out as different molecules through the study of carbohydrate specificity. Only NIC lectin was a potent mitogenic toward murine splenic lymphocytes at a concentration ca. 6.4 ㎍/㎖.

Inflammatory cytokine, abundantly produced in Hashimoto's thyroiditis, induced Fas expression in normal thyrocytes. We determined that susceptibility to Fas-activated apoptosis could be influenced by inflammatory cytokine and investigated a potential role of Gamgung-tang (GGT, Glycyrrhizae Radix, black beans, Angelicae Radix and Cnidii Rhizoma) in the thyroid follicular cells. $IL-1\beta$ was able to induce Fas expression in normal thyrocytes. GGT inhibited $IL-1\beta-induced$ Fas expression. Thyroid follicular cells were found to undergo DNA fragmentation with the inflammatory cytokines. GGT inhibited DNA fragmentation in a dose-dependent manner. These results suggest that GGT inhibit Fas-mediated apoptosis in thyroid follicular cells, therefore, may have therapeutic potential in the treatment of autoimmune chronic thyroiditis.
키토올리고당 함유 해양심층수의 포도당 흡수에 대한 효과

골격근, 지방조직과 같은 말초 조직의 세포내 포도당 흡수의 조절단계는 GLUT 4, IRAP, STX 6, STX 16에 의한 세포내 포도당의 유입과정이며 이 과정은 인슐린 신호전달계에 의해 주로 조절된다. 인슐린 수용체에 인슐린이 결합한 후 IRS 1의 인산화가 증가되고 GLUT 4가 세포막으로 이동을 촉진하여 혈중 포도당 농도를 낮춘다. 그리하여 본 연구에서는 3T3-L1 성숙지방세포에서의 포도당 흡수, 이 과정에 관련된 주요 전사인자인 GLUT 4, IRAP, STX 6, STX 16의 유전자 발현과 IRS 단백질의 인산화 반응에 대해 해양심층수 단독 또는 해양심층수와 키토올리고당 함께 처리 후 유전자 발현과 활성을 확인하였으며 해양심층수와 키토올리고당을 함께 처리하였을 때 포도당 흡수에 대한 상승효과를 증명하였다. 포도당 흡수에서는 대조군과 비교하였을 때 해양심층수 단독으로 처리하였을 때와 해양심층수와 키토올리고당을 함께 처리하였을 때 경도 의존적으로 흡수가 증가하였으며, 포도당 수송 관련 인자인 GLUT4, IRAP, STX 6, STX 16의 유전자 발현과 IRS 1 단백질의 인산화 발현이 증가하는 것을 확인하였다. 결론적으로, 해양심층수와 키토올리고당을 함께 처리한 경우 해양심층수의 비교적 낮은 경도에서도 우수한 포도당흡수 촉진효과를 나타내어 키토올리고당 함유 해양심층수는 효과적인 항당뇨 기능성 소재으로서의 활용가치가 기대된다. To investigate the effect of deep sea water (DSW) containing chitosan oligosaccharides (COS) on glucose uptake, we performed glucose uptake assay, RT-PCR and western blot in mature 3T3-L1 adipocytes with the treatment of DSW or DSW containing COS. Both DSW and DSW containing COS treatment increased glucose uptake in mature 3T3-L1 adipocytes. RT-PCR results revealed that the expressions of genes related to glucose uptake such as GLUT 4 for glucose transporter, IRAP for an abundant cargo protein of GLUT 4 storage vesicles (GSVs), Syntaxin 6, and Syntaxin16 for vesicle trafficking were increased. Moreover, the phosphorylation of IRS 1, which is an important signal molecule for insulin-dependent glucose uptake, was increased by DSW and DSW containing COS treatment in mature 3T3-L1 adipocytes. In conclusion, these results suggest that DSW containing COS is more effective than DSW alone in stimulatory effect on glucose uptake.
저분자 키토산 함유 해양심층수의 알코올대사에 미치는 효과

본 연구에서는 저분자 키토산과 해양심층수가 알코올 대사에 미치는 영향을 알아보기 위하여 해양심층수와 저분자 키토산을 함유한 해양심층수를 실험동물에 3주 동안 투여한 후 알코올 대사효소인 ADH, MEOS 및 ALDH의 활성, 혈중 알코올 농도를 측정하였다. 그 결과 간에서 알코올을 일차적으로 아세트알데히드로 산화시키는 효소인 ADH의 활성은 알코올만 투여한 군과 해양심층수 투여군에 비해 저분자 키토산을 함유한 해양심층수를 투여한 군이 27.8~29.9 unit로 증가하였다. 그리고 MEOS 활성도 저분자 키토산 함유한 해양심층수 투여군이 해양심층수에 비해 유의적인 증가를 나타내었다. 숙취 및 세포상해의 원인물질인 아세트알데히드를 분해하는 ALDH도 저분자 키토산을 함유한 해양심층수 (경도 1,500)를 투여한 군에서 13.0 unit로 유의적인 높은 활성을 보였다. 혈중 알코올 함량에서도 해양심층수 단독 투여군보다 저분자 키토산 함유한 해양심층수를 마우스에 투여한 군에서 유의적으로 낮게 나타났다. 이상의 결과에서 보는 바와 같이 저분자 키토산 함유한 해양심층수 (경도 1,500)는 아급성 알코올 중독시 간세포내의 알코올 대사에 관여하는 효소인 ADH, MEOS 및 ALDH의 활성을 증가 시킴으로서 알코올 분해대사를 촉진시켜 혈중 알코올 함량을 낮게 해 주는데 효과가 있음을 알 수 있었다. This study was done to investigate the effect of deep sea water (DSW) containing low molecular weight chitosan (LMC) on alcohol-metabolizing enzymes activities in ICR mice. The mice were orally administered 25% ethanol (alcohol group), 25% ethanol and DSW (hardness 0 or 1,500) (DSW group), 25% ethanol and DSW (hardness 0 or 1,500) containing LMC (2.5 mg/ 0.3 mL/day) (DSW containing LMC group) for 3 weeks. Alcohol dehydrogenase (ADH) activity was increased in the DSW (hardness 1,500) containing LMC group compared to the alcohol and DSW group. Also, in the DSW (hardness 1,500) containing LMC group, Microsomal ethanol oxidizing system (MEOS) and aldehyde dehydrogenase (ALDH) activities showed significantly increasing tendency compared to the other groups. The blood alcohol content was lower due to the increased activities of alcohol-metabolizing enzymes (ADH, MEOS, and ALDH) in the DSW (hardness 1,500) containing LMC group. These results suggest that the DSW (hardness 1,500) containing LMC may alleviate ethanol-induced hepatotoxicity by altering alcoholmetabolizing enzymes activities.
