        In order to investigate an endocrinological effect on metamorphosis of the midgut in silkworm, Bombyx mori, 20-hydroxyecdysone and crude extract of brain (PTTH) were injected into pupal isolated abdomens, devoid of prothoracic glands and also in the brainless diapausing pupae with the prothoracic glands (so-called dauer pupae). When ecdysone was assayed abdomens, drastic morphological changes occurred in the midgut. The wet weights of midguts were found decrease as like the normal pupae at metamorphosis. Proteineous changes in epithelial midgut were also observed; pupal specific proteins (PSP) vanished while the adult proteins were newly synthesized. Similar results were also found in the changes of protein in the midgut of dauer pupae by injecting ecdysone and pTTH.

        Treatment of juvenile hormone analogue(JHA) at the 5th instar larvae prolonged the duration of adult development one to one and half days as well as the elongation of feeding time with the increasing of larval body weight. Morphological observation and protein analysis in hemolymphs, integuements, alimentary canals, fat bodies and ovaries also revealed that the development of these tissues and organs for adultation are affected by the JHA treatment.

      • 동위효소 다형특성에 의한 누에 품종의 유연관계

        Isozyme was used to characterize general protein patterns of genetic relationships among 303silkworm stockf preserved in National 誇ericultural and Entomology Research Institute, RDA.Six isozymes (esterase, acid phosphatase, alkaline phosphatase, amylase, glucose-6-phosphatedehydrogenase and sucrase) from hemolymph, midgut, and digestive juice were employed toconstruct dendograms(UPGMA method) using a polyacrylamide Eel electrophoresis. A clusteranalysis revealed fnur major groups out of 303 strains. The current data showed that the firstand the second group, which were divided into sneral subgroups within each group, rontainedassemblages of Japanese and Chinese races. Especially, genetic differentiation in the nrst andsecond group was greatest rather than within Japanese and Chinese races repectively and wasconcordant with the hypothesis of phyletic sorting of initial variability in China many yearsago. Hypothesized recent introgression between groups was also plausible, but the evidencesuggested bidirectional gene flow between the the thinese and the Japanese lineages.Interpreting the results in light of evidence from the current study, the genetic diversity andrelationship showed in Korean silkworm race, Hansammyun reflected early and independentevolution from the Chinese ancestor, limited addition of new variability and phyletic sortingwithin Korean penin$ula more than 4,000 years.

      • 누에로부터 Polyhydroxylated Alkaloids의 분리와 효소저해능 조사

        It is well known that the isolated polyhydroxylated alkaloids from the mulberry trees and silkworms inhibit the activity of α-glucosidase, β-glucosidase, β-galactosidase. From the 5th larvae of silkworms, we isolated seven polyhydroxylated alkaloids, from which three kinds of alkaloids such as DNJ, HS-58 and HS-74 were purified through a series of column chromatography. 1-Deoxynojirimycin supressed α-glucosidase actively, showing comparatively weak supression to β-glucosidase. In contrast, HS-58 inhibited β-glucosidase strongly, though α-glucosidase was not almost affected by this compound. HS-58 as well as other isolated alkaloids showed in relative high inhibition to the maltase which extracted from the porcine intestine.

        Twenty seven out of ca. 5,000 actinomycete strains, which were isolated from soil collected throughout the country, showed anti-microbial effects against fungus, Rhizopus stronifer (ATCC6227a) and yeast, Candida albicans (ATCC10231). From these anti-fungal microorganisms, we further selected seven strains which seems to produce insecticidal substances with in vivo test, using silkworm, Bombyx mori. Morphological and biochemical experiments revealed that at least three strains out of seven were Streptomyces. Further investigations on the physical and chemical properties of these antifungal and insecticidal substances are now in progress.

      • 동위효소 다형특성에 의한 누에 품종의 유연관계

        Isoenzyme was used to characterize patterns of genetic relationships among 141 silkworms, Bombix mori, races in National Sericultural and Entomology Research Institute, RDA. Six isoenzymes (esterase, succinate dehydrogenase, glucose-6-phosphate dehydrogenase, acid phosphatase, α-amylase) from blood and midgut were employed to construct dendrograms (UPGMA method) using a polyacrylamide gel electrophoresis. A cluster analysis revealed four major groups out of 141 races : 1) a Chinese group (group 2), 2) two Japanese group (group 1 and 3), and 3) the most distinct group 4. This current data showed that the Chinese and Japanese assemblage contained races originated from China and Japan respectively. Therefore, these distinctive groupings supported that the four major group associations were strongly originated geographically. Frequencies of correlated presence and absence of alleles with geography suggested that marked differences exist at esterase (Est1, Est2) and general protein (from blood) for groups 2 and 3, and a α-amylase allele present at a high/low frequency in groups 1, 2 and 3. Characteristics of group-specific enzymes were able to resolve the relationships between genotype and geographic origin as markers effectively.

        Adult specific proteins, referred to as ASP-I (adult specific protein of slow mobility) aud ASP-II (adult specific protein of fast mobility) at the pharate adult stage of the silkworm, Bombyx mori, were detected by polyacrylamide gel electrophoresis. The adult specific proteins(ASP-I and ASP-II) were defined as a kind of adult hemolymph proteins without sex specificity, and there was no variation in the respective electrophoretic mobility of ASP-I and of ASP-II among the forty-one silkworm varieties. tested.

        PBS soluble-and insoluble-extracts from silkworm eggs were analyzed by native-PAGE and SDS-PAGE. During the embryonic development, soluble proteins which are mainly composed of ESP, vitellin and 30K proteins showed similar degradation pattern in both electrophoretic analyses. Several peptides which seemed to be intermediated forms of yolk proteins were detected in latter part of embryogenesis, while a protein band newly appeared in one day elapsed after acid treatment. When insoluble extracts from silkworm eggs were analysed with SDS-PAGE, several peptides were detected at the later stages of the embryonic development and newly hatched larvae. These peptides are considered to be structure proteins for embryonic morphogenesis.

        동위효소에 의한 누에품종의 계통분류를 위해 누에 계통간 다형성을 나타내는 동위효소를 탐색하였다. 누에의 체액, 중장소화관 및 소화액으로부터 추출한 효소액을 대상으로 PAGE로 분석한 결과 acid phosphatase, alkaline phosphatase, amylase, esterase, proteinase, sucrase 및 malic dehydrogenase를 비롯한 수종의 탈수소효소에서 효소다형성을 나타내고 있음이 밝혀졌다. 이들 효소의 다형성은 단백질의 구조, 유전자좌위 및 대립유전자의 수 등에 의한 것으로 추정된다. The relationships between phenotypic characters and genetic distances based on isozyme markers were poorly understood. The primary objective of this study was to assess genetic relationships and clonal structures of Korean silkworm races using several isozymes. Among fourteen isozymes extracted from silkworm blood, midgut, digestive juice, ten isozymes (i.e., acid phosphatase, alkaline phosphatase, amylase, esterase, proteinase, sucrase and some dehydrogenases) were found to be polymorphic loci by polyacrylamide gel electrophoresis(PAGE). Results suggested that polymorphic loci are characterized by protein structures(structural gene), number of loci and alternative forms and number of alleles.

      • 누에와 천잠의 α-glucosidase 활성 비교연구

        α-Glucosidase activities in the hemolymph and midgut of the mulberry silkworm, Bombyx mori, and Japanese oak silkmoth, .Antheraea yamamai were determined using various substrates such as maltose, sucrose, trehalose, and ρ-nitrophenyl α -D-glucopyranoside. The a-glucosidase activities in the hemolymph of B. mori were in general higher than those in the hemolymph of A. yamamai, however, the enzyme activities in the midgut of A. yamamai were higher than those in the midgut of B. mori for all or the substrates tested. In addition, the effect or pH and temperature on a-glucosidase activities in the midgut were similar between B. mori and A. yamamai for all or the substrates tested. The migration patterns or the a -glucosidases detected by native PAGE were different between B. mori and A. yamamai, suggesting that the molecular structure or a-glucosidases or both insects may be different in spite or the similarity or Physiological function.
