This experiment was conducted to find out the effects of L-phenylalanine on the saikosaponin content of callus induced from Bupleurum falcatum leaf segments. In the fresh and dry weight of callus, the addition of 2,4-D than L-phenylalanine was significantly effective. However, the L-phenylalanine treatment rather than 2,4-D was effective for high saikosaponin accumulation in the callus of Bupleurum falcatum.

In order to product the saikosaponin which is one of the secondary product from Bupleurum falcatum efficiently through the tissue culture, several levels of agar and gellan gum as the gelling agent, 2,4-D as the growth regulator, and L-phenylalanine as the precursor were used with single or combination treatment on MS basal medium. Gellan gum was more effective than agar as the gelling agent in fresh and dry weight increase of callus induced from Bupleurum falcatum leaf segment. Gellan gum medium supplemented with L-phenylalanine produced 1.6 times of fresh weight more than that of agar. The fresh weight was remarkably high in gellan gum when the calli was treated with the combination of 2,4-D and L-phenylalanine similar to the single treatment of 2,4-D or L-phenylalanine. However, the saikosaponin content in callus was high in gellan gum with the single treatment of L-phenylalanine. Especially, the saikosaponin content in gellan gum supplemented with 1.0mg/L L-phenylalanine was 2 times(2.4 mg/g) higher than that in agar medium supplemented with 0.1 mg/L 2,4-D(0.9 mg/g).

Lonicerae Flos, the flower of Lonicera japonica Thunb., has been used as a diuretic, stomachic, antipyretic, analgesic and anti-inflammatory agent in Korea. In order to evaluate the quality of Lonicerae Flos, the method of isolation and quantitative determination of luteolin $7-O-{\beta}-D-glucoside$ as a reference standard compound has been developed. Different specimens of Lonicerae Flos were collected from twenty Korean markets and were analyzed with HPLC using the mobile phase of MeOH-4.5% acetic acid solution (16.5:83.5). The average content of luteolin $7-O-{\beta}-D-glucoside$ from Lonicerae Flos in Korean markets was $0.43{\pm}0.34%$.

Polygoni Multiflori Radix, the root of Polygonum multiflorum Thunb., has been used as a tonic, and in the inflammation and constipation in Korea. In order to evaluate the quality of Polygoni Multiflori Radix, the method of isolation and quantitative determination of emodin as a reference standard compound has been developed. Different specimens of Polygoni Multiflori Radix were collected from twenty Korean markets and were analyzed with HPLC using the mobile phase of MeOH-water (64:36). The average content of emodin from Polygoni Multiflori Radix in Korean markets was 0.021%.
고성능 액체크로마토그래피를 이용한 당귀 중 Azocyclotin 및 Cyhexatin 잔류분석법

A High-performance liquid chromatographic (HPLC) method to determine residues of azocyclotin and cyhexatin in dry root of Angelica gigas, a medicinal herb was developed for the official inspection. Azocyclotin and cyhexatin were extracted from the moistened sample with acetone/hydrobromic acid mixture. Following to dilution of the extract with saline, the analyte was partitioned into the hexane phase. During extraction and partition steps, azocyclotin was completely converted to cyhexatin. Ion-suppressed silica gel column chromatography was employed for further purification of the extract prior to HPLC determination. In reversed-phase HPLC using an octadecylsilyl column, cyhexatin could be eluted only when hydrochloric acid and sodium chloride was added to the mobile phase and quantitated by ultraviolet absorption at 214 nm. No interference was found in the sample extract and, as a result, quantitation limit of the proposed methods ranged 0.04∼0.05 mg/kg. Mean recoveries from samples fortified with azocyclotin and cyhexatin were in the range of 80.7∼82.0% and 81.2∼83.2%, respectively. Relative standard deviations of the analytical method were less than 10%. Even though the LC/mass spectrometry was failed due to poor ionization of cyhexatin, an alternative HPLC parameter could be applied for confirmation in situ as the dramatic change in retention behavior between co-extractives and the analyte was observed.
