Acetaminophen 유도 간독성에 대한 백삼과 홍삼 추출물의 간보호 효과

Acetaminophen(APAP) is one of the most extensively used analgesics and antipyreics worldwide. In order to investigate preventive effects of white and red ginseng extracts, male ICR mice pretreated with white or red ginseng extracts(50 or 250 ㎎/㎏/day, for 5 days, orally) before treatment with acetaminophen(800 ㎎/㎏, i.p, single dose). In an attempt to elucidate the possible mechanism of hepatoprotective effect, superoxide dismutase(SOD), catalase(CAT), hydroperoxide, malondialdehyde(MDA) contents were studied. In pretreatment with red ginseng extract(250 ㎎/㎏), the activities of SOD, CAT were generally highest and the hydrogen peroxide content was lowest. The levels of MDA were significantly lower in white and red ginseng extract groups than those in the APAP groups. By treatment with ginseng extract, high content of hydrogen peroxide and increased lipid peroxidation level caused by APAP could be lowered. Also, ginseng extracts were found to increase antioxidative enzyme activity. Finally, the results suggest that the antioxidant effects of (white and red) ginseng extracts prevent oxidative damage by direct antioxidant effects involving SOD, CAT and increasing the ability to synthesize endogenous antioxidants. It was concluded that ginseng can protect against APAP intoxication through its antioxidant properties.

The purpose of this study is to investigate the pollution characteristics of groundwaters in Kansan city. Volatile organic compounds such as TCE(tri-choroethylene), PCE(tetruchloroethylene), 1,1,1-trichloroethane, THM(trihalo-methane), dichloromethane, pesticides such as diazinon, parathion, malathion, toxic inoganic matters such as As, Hg, Se, Cd, Al, Cr6+, oder, taste and phenol were not detected in the groundwaters. Mean values of groundwater pollution indicators were bellow drinking-water standards, but Bacteria, Total Coliforms, Pb, Mn, NO3-N and NH3-N, total solid and pH exceeded a little in some samples.

To study on antioxidant effects in the liver of 40-week-old mouse, the sample were orally pretreated 5mg/kg/day for 5 days with red ginseng saponin components(total saponin, protopanaxadiol saponin, protopanaxatriol saponin, ginsenoside-Rd, ginsenoside-Re, compound-K) for 5 days. The ability of saponin to protect the mouse liver from oxidative damage was examined by determining the activity of superoxide dismutase(SOD), glutathione peroxidase(GPx) and the contents of glutathione, the level of malondialdehyde. The only protopanaxadiol among the ginseng saponin fractions was significantly increased the hepatic SOD activity(p<0.01). The red ginseng saponin induced a slight increase of GPx activity, especially ginsenoside Rd, compound K and protopanaxatriol treatments significantly increased its activity. The content of glutathione was significantly increased by total saponin, protopanaxadiol and ginsenoside Rd(p<0.01), but the oxidized glutathione level was lowered in all the red ginseng saponin. Finally, the level of malondialdehyde was significantly decreased by ginsenoside Rd and protopanaxadiol. In conclusion, protopanaxadiol and ginsenoside Rd among the saponin fraction were especially increased in the activity of hepatic antioxidative enzyme and decreased the lipid peroxidation that was expressed in term of MDA formation. This comprehensive antioxidant effects of red ginseng saponin seems to be by a certain action of saponin other than a direct antioxidant action.
만경강 동진강 유역의 토양, 현미, 저질토중의 중금속 함량 및 분포

This study was performed to investigate the heavy metal contents and distribution in soil, sediment, and rice from the downstream area of ManKyeong and DongJin River. Of the sites on Mankyeong river area, site M-1(Mokchon bridge) showed the highest average contents of Cd, Cr and Pb in paddy soil. In DongJin river area, site D-3(Munpo) and D-4(Gerjeonri) showed relatively high level of average contents of Cr, Pb and Zn in paddy soil. The average contents of heavy metals in brown rice from ManKyeong river area were 0.10㎎/㎏ for Cd, 0.99㎎/㎏ for Cr, 2.07㎎/㎏ for Pb, 4.44㎎/㎏ for Cu and 32.03㎎/㎏ for Zn while those in brown rice from DongJin river area were 0.14㎎/㎏ for Cd, 0.74㎎/㎏ for Cr, 1.78㎎/㎏ for Pb, 4.57㎎/㎏ for Cu and 33.60㎎/㎏ for Zn. Zn showed the highest transportation-rate from paddy soil to brown rice while Pb showed the lowest. From the results of heavy metal analysis in sediments, the average contents of Cd, Cr, Pb and Cu were generally high in site M-5(Euonri) and D-4(Gerjeonri), the most downstream sites in Mankyeong river and DongJin river, respectively.
Paraquat를 투여한 생쥐 간에서 홍삼 사포닌의 항상화 효소 활성과 전기영동에 관한 연구

ICR계 주령 생쥐에 PQ를 투여한 후 TS, PD, PT를 경구 투여하여 생쥐 간에 미치는 항산화 효소(SOD, CAT)활성 증대 효과를 알아보기 위하여 효소 활성도와 전기영동상밴드의 변화를 조사하였다. SOD의 활성은 PQ+PD 투여군, CAT의 활성은 PQ+PD 투여군이 활성화되었으며, SOD isozyme은 전기영동상에서 3개의 밴드를 나타내었는데 이중 한 개의 밴드는 음극에 가까이 위치하고 다른 2개의 밴드는 양극 쪽에 위치하였고, KCN 처리한 결과 Cu, Zn-SOD는 음극 쪽에 위치한 반면 Mn-SOD는 양극쪽에 있었다. 이러한 결과를 종합하여 볼때 홍삼 사포닌의 항산화 효과는 항산화 효소인 SOD, CAT의 직접적인 작용으로 인하여 산화적 손상에 대한 보호 능력을 증대시큰 것으로 생각된다. This study examined effects of the active ingredients from ginseng on paraquat(PQ) toxitity. Mice were given PQ(25mg/kg, ip) and then they were given total saponins (TS; 5mg/kg, orally), protopanaxadiol (PD; 5mg/kg, orally) and protopanaxatriol(PT; 5mg/kg, orally) per day for periods of 1,3 & 7 days. We measured the activities of superoxide dismutase (SOD), electrophoretic isozyme band, catalase (CAT) were compared in the liver of mouse that dose with PQ and/or TS, PD and PT. The activities of SOD, CAT were generally higher in PQ+PD group than others groups. Especially the activity of SOD was the highest in PQ+PD group than others groups. SOD isozyme separated into three bands by electrophoresis. One band was located to near the anode side and two bands were cathode side. As the results of treated with KCN, we were confiremed that the Cu, Zn-SOD was located to near the anode side but the Mn-SOD were cathode side. Our results suggested that an antioxidant effect of ginseng saponins elevated a protection ability to an oxidative damage by direct action of SOD, CAT and reinforced the synthetic ability of endogenous antioxidant material in living organism. Particularly, PD was a effective antioxidant compared with others.
