편평세포성 폐암의 c-myc 암유전자 발현에 관한 in situ Hybridization 연구

By means of in situ hybridization emplying a biotinylated cDNA probe and the immunohistochemical reaction of avidin-biotin complex-alkaline phosphatase with substrate, expression of c-myc oncogene was investigated in the formalin-fixed, paraffin-embedded target tissue sections from six patients with squamous cell carcinoma of primary lung origin. Nuclear localization of c-myc expression as clumpy grains was assessed. One fifth of the tumor cell populations of the six cases studied revealed an increased c-myc expression with individual and cell-to-cell variation. In contrast, all of the specimens incubated with deoxyribonuclease after the pretreatment of ribonuclease were nil in the expression of c-myc oncogene. This in situ hybridization study permits estimation of a heterogeneous amplification of c-myc oncogene that may be related to secondary alterations occurring during the progression of the lung cancer.
