        국립백두대간 수목원의 터핀(terpene)류의 발생특성

        Forests are valuable natural resources for people living around the mountains. In particular, the comfortable feeling or healing is one of the most important benefits obtained from forests. This healing can be possible by many aspects of forests, including the landscape, natural sounds, anions, and pleasant aromas. We focused on the volatile organics from forest causing pleasant aromas, phytoncides. Twenty phytoncides were monitored from February to September in a national tree garden (BaekDoDaeGan SooMokWon). Five sites were monitored two times per month and 20 phytoncides were detected. Borneol showed the highest annual average concentration and the order of concentration was borneol > mycene > sabinene > limonene > α-pinene. The average phytoncide concentration was relatively high in spring and summer season when the trees were physiologically active. Daily monitoring showed that the afternoon hours had higher concentrations of phytoncides than the morning hours, which may be due to the stabilized atmospheric conditions at the sites. Among the five sites, coniferous forests gave higher phytoncide emissions than broadleaf tree forests. The current study showed that forests produce several phytoncides that cause a healing effect and a forest bath may be beneficial to the health of visitors to forests.

        채취 시기 및 지역에 따른 봉독의 성분 분석

        서양종 꿀벌 독의 채집시기와 지역에 따른 봉독의 성분 변화 및 약리효과에 미치는 영향을 검토하였다. 채집시기는 5월부터 9월까지, 채집지역은 전국 35개 지역으로부터 채취한 봉독을 대상으로 2010년과 2011년 2년에 걸쳐 동일 지역에서 동일한 방법으로 봉독을 채취하였다. 채취한 봉독은 액체크로마토그래피를 통해 멜리틴과 아파민 그리고 포스포리파아제 A2의 성분 함량을 분석하였다. 그 결과 채집시기와 지역에 따른 성분에 유의한 차이는 확인되지 않았다(One way-ANOVA, Duncan's test (${\alpha}$=0.05)). 봉독의 성분은 채집시기와 지역에 관계없이 멜리틴 $55.2{\pm}2.07%$, 아파민 $2.57{\pm}0.103%$ 그리고 포스포리파아제 A2는 $12.51{\pm}0.37%$을 차지하였다. 이상의 결과로부터 봉독은 채취시기에 따른 주요 성분은 차이를 갖고 있지 않았으며, 이는 꿀벌의 먹이, 사육온도 등 외부 환경이 봉독 분비에 영향을 주지 않는 것으로 사료되었다. This study aims to investigate whether geographical variation affects the antibacterial component properties of honeybee (Apis mellifera L.) venom in Korea. Honeybee venom samples were collected from May to September, during 2010 and 2011, from 35 different sites, and were analyzed for major components, including melittin, apamin and phospholipase A2 were determined by a liquid chromatography using ammonium formate, acetonitrile, trifluoracetic acid. On average, melittin, apamin and phospholipase A2 were determined $55.2{\pm}2.07%$, $22.57{\pm}0.103%$, and $12.51{\pm}0.37%$, respectively. The ratio of the major components, including melittin, apamin and phospholipase A2 did not differ significantly according to flower or temperature during collections (One way-ANOVA, Duncan's test (${\alpha}$=0.05)).

        봉독채취가 로얄제리의 항산화 및 항당뇨 활성에 미치는 영향

        This study was conducted to identify whether honeybee venom collection by means of electrical stimulation, affected the antioxidant and anti-diabetes properties of royal jelly using through 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, hydroxyl radical (ㆍOH) scavenging activity, reducing power activity, α-amylase and α-glucosidase inhibitory activities. This experimental groups were divided from two beehive groups; R group was collected only royal jelly, RV was collected together royal jelly and bee venom at same beehive. IC?? values for DPPH radical scavenging activity of R and RV were 25.5±0.49㎍/㎖ and 24.9±0.79㎍/㎖, respectively. R and RV were not significantly different inㆍOH scavenging activity (IC?? R: 13.8±0.6㎍/㎖, R+V: 13.1±1.3㎍/㎖), reducing power, α-amylase (R: 20.5±0.49%, R+V: 20.9±0.79%), and α-glucosidase (R: 23.8±0.6%, R+V: 23.1±1.3%). These results demonstrated that venom collection using the method of coupled electrical stimulation unrelated to the antioxidant and anti-diabetes activity of royal jelly.

        호박벌 일벌독의 성분 분석 및 생리활성 탐색

        호박벌 일벌독의 성분과 생리활성을 규명하기 위하여 단백질 성분분석과 암세포 생육 저해 효능, 항균력을 검토하였다. 이차원단백질 분석을 통해 호박별의 일벌독은 63개의 단백질이 존재하는 것으로 확인되었으며, 가장 많은 함량을 보이는 3개의 단백질을 염기서열 분석하였다. 그러나 이들 성분은 아직 밝혀지지 않은 성분으로 판단되었다. 호박벌 일벌독의 암세포 (간암; Hep3B, 폐암; A549, 유방암; BT-20, 위암; AGS) 에 대한 생육 저해능은 시료 농도가 증가함에 따라 증가하는 경향을 보이며 100ng/ml에서 간암세포 (Hep3B) 에 대한 생육 저해능이 55%로 가장 높았다. 항균활성에 E. faecium 과 S. sonnei에 대하여 최소발육억제농도와 최소살균농도 모두 각각 0.256ug/ml 로 강한 항균활성을 나타내었으며, 그 외의 피검균에 대해서도 비교적 높은 활성을 보였다. 이상의 결과로부터 호박벌 일벌독의 성분은 다른 벌의 독성분과는 차이를 보이며, 그 생리활성에 있어 약리학적 이용이 가능할 것으로 판단되었다. Pure Bombus ignitus venom samples were submitted to two-dimensional gel electrophoresis. A total of 64 excised spots were analyzed by mass spectrometry. Three main proteins resulted in the identification have not been described in other bee venoms before. Dose-dependence against human carcinoma (Hep3B, BT-20, A549 and AGS) were observed from 1ng/ml to 100ng/ml. Expecially, the treatment of 100ng/ml B. ignitus venoms showed the highest cytotoxicity with 55% against hepatocellular carcinoma (Hep3B). The B. ignitus venoms showed strong antimicrobial activities against Enterococcus faecium and Shigella sonnei, and practically antimicrobial activity against the other microorganisms tested. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of E. faecium and S. sonnei, were 0.256ug/ml, respectively.

        국내산 봉독의 주요성분 분석

        The aim of the study was to the determination quantitatively of the major constituents of honeybee (Apis mellifera L.) venom from Korea by liquid chromatography. Samples of honeybee venom were purchased from Chungjin, Ibee and Sigma over two consecutive beekeeping season (2007~2008). The sephadex TM75 and source 15RPC ST column was applied. Several honeybee venom constituents were separated and the content of three of them (melittin, apamine and phospholipase A₂) were determined. The chemical composition of honeybee venom from Korea were evaluated. Melittin content varied from 48.5 to 67.2 and averaged 50.7%. Apamin content camewithin a range of 2.0 to 3.2, and averaged 2.8%, and phospholipase A₂ content was between 10.8 to 14.5, averaging 12.8%. There is no statistically significant differences for the content of the main component compared standard honeybee venom to domestic honeybee venom. The quantitative composition of melittin, apamine and phospholipase A₂ may provide a basis for the development of the domestic standard for honeybee venom.

        봉독채취가 로얄제리 생산과 생산비에 미치는 영향

        This study was conducted to identify whether honeybee venom collection by means of electrical stimulation, affected royal jelly production and costs in honeybee colonies. This experimental groups were divided from three beehive groups; R group was collected only royal jelly, RV was collected together royal jelly and bee venom at same beehive, and V was collected only bee venom. There was no difference in the yield of royal jelly at R and RV group. Physical peculiarity of royal jelly such as color, taste, flavor and form was not difference at R and RV group. The content of moisture, total protein and 10-HDA was not significantly at R and RV group. The value of production less total costs to beekeepers increased at R or V group. The results obtained demonstrated that the mean yield of royal jelly, obtained only from colonies in which venom was collected using the method of coupled electrical stimulation, was not significantly different than the yield of royal jelly recorded in control colonies.

        경상북도지역의 오염된 토양에서의 중금속 함량에 관한 곤충병원성 선충에 관한 연구

        경상북도내의 금속광산, 공장 및 공업, 일반폐기물, 공단주거지역 및 특수 용수사용지역 등 360곳의 토양을 2001년 3월부터 10월까지 채취하여 토양 오염도를 조사하였다. 토양 오염도는 중금속인 카드뮴, 구리, 비소, 수은, 납 6가 크롬 및 시안의 7개 항목을 대상으로 하였으며, 그 중 각 항목에서 가장 오염도가 심한 5개 지점을 대상으로 토양내 곤충병원성 선충의 분포를 조사하였다. 토양내 선충은 누에를 이용하여 곤충병원성, 사물 기생성 선충을 대상으로 하였으며, white trap을 설치하여 선충을 분리하였다. 카드뮴 오염이 0.870ppm 이하인 지역에서 Rhabditida 목 선충 2과가 확인되었으며, 비소에 오염된 토양에서는 0.75ppm 이하에서 Rhabditidae 과 선충이 확인되었다. 6가 크롬은 0.05ppm이하인 토양에서 Cylindrocorpidae과 선충이 확인되었고, 납과 구리, 시안 오염 지역에서는 오염농도와 관계없이 Rhabditida 목 선충 2과가 검출되었다. 그러나, 시안에 오염된 토양과 토양오염우려기준을 초과한 토양에서는 선충이 검출되지 않았다. 이러한 결과는 오염지역내 선충의 분포 밀도는 매우 낮거나 존재하지 않는 것으로 확인되며 토양의 오염은 선충의 생존에 불리한 환경으로 작용하는 것으로 보인다. The variety and density of entomopathogenic nematodes from the polluted soils of heavy metals were examined. In order to investigate the pollution of heavy metals in soil, 360 sites in kyungsangbuk-do were collected from March to October in 2001. We measured the contents of seven heavy metal elements (Cd, Cu, As, Hg, Pb, Cr6+, CN) and than choosed soil of 25 sites with high concentration of heavy metals. The seven strains of nematodes were isolated from seven samples by silkworm host (Bonbyx mori mon`) and white trap. Isolated nematodes are composed of two families, one order. The members of Rhabditida were isolated in the soil with mean Cd content of 0.870 ppm. And they were isolated in the soil samples with As content less than 0.745 ppm. However they were isolated regardless of concentration of Cu and Pb. The members of Cylindrocorpidae were isolated in the soil samples with Cre+ content less than 0.05 ppm. Any entomopathogenic nematode was not detected in the CN polluted soil. Isolated nematodes successfully cultured on the silkworm host and were confirmed the pathogenicity, multiplicity, and tolerance against various condition of preservation. Which proved its potential usefulness as biological agent.

        벌꿀을 이용한 매실추출물의 이화학적 특성

        We study that their optimal extraction condition honey maesil (Prunus mume) extracts to health functional food were extracted with the honey. The extraction ration was in the condition of 1 maesil and 1.5 honey weight for 2 months. As standard ingredient, citric acid were determined 10.2mg/g in honey maesil extracts by HPLC. Cyan compound was not detected by the picrate method. Also Escherichia coli was not detected by 3M paper method. The honey maesil extracts contained free sugars as fructose and glucose (43.5%), and sucrose (1.5%). These results suggest that honey is the effective extracts method for the health functional food in maesil extracts.
