A report of 22 unrecorded bacterial species in Korea, isolated from Namhangang

As part of a larger study of indigenous prokaryotic species diversity in South Korea, various samples from Namhangang were subjected to analyses. Fresh water, underwater sediment, and moss-inhabiting aerobic and anaerobic bacteria were isolated. 22 of the isolates were identified as unrecorded bacterial species in Korea that had ≥9 8.7% 16S rRNA gene sequence similarity with published species. The aerobic strains isolated were Kurthia gibsonii and Massilia plicata. Also identified were four facultative anaerobic strains: Bacillus hisashii, Enterococcus rotai, Paenibacillus vini, and Pediococcus pentosaceus. 16 strictly anaerobic strains were identified as Bacteroides xylanolyticus, Carnobacterium maltaromaticum, Clostridium argentinense, Clostridium beijerinckii, Clostridium butyricum, Clostridium cavendishii, Clostridium diolis, Clostridium frigidicarnis, Clostridium perfringens, Clostridium saccharoperbutylacetonicum, Clostridium sphenoides, Clostridium subterminale, Cutibacterium acnes, Paraclostridium bifermentans, Prevotella paludivivens, and Romboutsia lituseburensis. Based on the examination of morphological, cultural, physiological, and biochemical properties of the isolates, descriptive information of these previously unrecorded species is provided here.

Objectives : Ulcerative colitis is a chronic recurrent inflammatory disease of the gastrointestinal tract. However, there are some drawbacks to long-term drug therapy such as the risk of opportunistic infections. Recently, there was an increasing interest on the use of khorasan Kamut wheat because of their higher value of selenium and fiber than modern wheat. The present study was aimed to investigate the effect of Kamut brand wheat enzyme (Kamut WE) diet on colon health in dextran sulfate sodium (DSS)-induced colitis mice. Methods : Female C57BL/6J mice were divided into 6 groups. (1) normal (Water and AIN-93G diet), (2) control (1.25% DSS and AIN-93G diet), (3) Kamut WE (1.25% DSS and Kamut WE diet), (4) normal (Water and AIN-93G diet), (5) control (2.50% DSS and AIN-93G diet), (6) Kamut WE (2.50% DSS and Kamut WE diet). Dietary intake, body weight change, disease activity index (DAI), colon length and spleen weight were monitored. Results : Kamut WE group alleviated colitis symptom, including dietary intake loss, DAI (weight loss, loose stools, bleeding), colon length shortening and spleen swelling. Further, Kamut WE diets showed a significant effect against pathological damage by the increased colon length, decreased DAI and spleen weight in DSS 1.25% as well as DSS 2.50%. Conclusions : Our study provides evidence that Kamut WE diet increased colon length, decreased DAI and spleen weight in intestinal inflammation.

Chalcones, one class of polyphenolic compounds synthesized as secondary metabolites during flavonoid biosynthesis in plants, have anti-inflammatory and antitumorigenic effects. In this study, we investigated the roles of a novel chalcone derivative, (E)-3-(3,5-dimethoxyphenyl)- 1-(1-hydroxynaphthalen-2-yl)prop-2-en-1-one (DK-59), on rheumatoid arthritis (RA) fibroblast-like synoviocyte MH7A cells. DK-59 treatment reduced cell viability and induced caspase-mediated apoptosis in MH7A cells; activation of caspase-7 and caspase-3, as well as fragmentation of poly (ADP-ribose) polymerase (PARP-1), occurred in a time-dependent manner, and treatment with a pan-caspase inhibitor, Z-VAD, significantly reduced DK-59-mediated PARP-1 cleavage. DK-59-mediated apoptosis was caused by the production of intracellular reactive oxygen species (ROS); DK-59 treatment induced ROS production in a time-dependent manner, and treatment with the antioxidant N-acetyl-Lcysteine significantly inhibited DK-59-mediated fragmentation of PARP-1 and recovered cell viability. Many genes that function in the unfolded protein response (UPR), including inositol-requiring protein 1, X-box binding protein 1, and C/EBP homologous protein (CHOP), were upregulated by DK-59 treatment. CHOP knockdown by lentivirus infection reduced DK-59-mediated PARP-1 cleavage, suggesting that CHOP plays a proapoptotic role in DK-59-mediated apoptosis in MH7A cells. Taken together, the results suggest that DK-59-mediated induction of ROS and activation of the UPR can be used as a target for the therapeutic treatment of RA.
A report of 28 unrecorded bacterial species, phylum Bacteroidetes, in Korea

In order to investigate indigenous prokaryotic species diversity in Korea, various environmental samples from diverse ecosystems were examined. Isolated bacterial strains were identified based on 16S rRNA gene sequences, and those exhibiting at least 98.7% sequence similarity with known bacterial species, but not reported in Korea, were selected as unrecorded species. 28 unrecorded bacterial species belonging to the phylum Bacteroidetes were discovered from various habitats including wastewater, freshwater, freshwater sediment, wet land, reclaimed land, plant root, bird feces, seawater, sea sand, tidal flat sediment, a scallop, marine algae, and seaweed. The unrecorded species were assigned to 18 different genera in five families: Flavobacterium, Epilithonimonas, Dokdonia, Gillisia, Flavicella, Chryseobacterium, Algibacter, Aquimarina, Lacinutrix, Gaetbulibacter, Cellulophaga, Tenacibaculum, and Maribacter of Flavobacteriaceae, Dyadobacter of Cytophagaceae, Draconibacterium of Draconibacterium_f, Sunxiuqinia of Prolixibacteraceae, and Fulvivirga of Fulvivirga_f. The selected isolates were subjected to further taxonomic characterization including analysis of Gram reaction, cellular and colonial morphology, biochemical activities, and phylogenetic trees. Descriptive information of the 28 unrecorded species is provided.
