Non-Viral Transgenesis via Direct In Ovo Lipofection in Quail

Transgenic animals have been widely used for developmental biology studies, as disease models, and even in industry such as transgenic bioreactor animals. For transgenic birds, quail has the great advantages of small body size, short generation time, and frequent egg production. To date, retroviral or lentiviral transduction has been used to generate transgenic quail for various purposes. However, the efficiency of transgenic offspring production with these methods is relatively low and viral vector usage has safety issues. Unfortunately, non-viral transgenesis has not been established in quail due to a deficiency of stem cell and germ cell culture systems. In this study, we established a direct in ovo lipofection method that could be used to create transgenic quail without germline-competent cells or viruses. To optimize the injection stage during embryo development, the liposome complex (containing piggyBacCMV-GFP and transposase plasmids) was introduced into an embryonic blood vessel at 50 hr, 55 hr or 60 hr. GFP expression was detected in various tissues (heart, kidney, liver and stomach) on day 12 of incubation under a fluorescence microscope. Additionally, GFP-positive cells were detected in the recipient embryonic gonads. In conclusion, the direct in ovo lipofection method with the piggyBac transposon could be an efficient and useful tool for generating transgenic quail.
노인의 근골격계 특성에 적합한 기공운동 개발을 위한 기초연구

Objective : The objective of this study is to develop a Qigong exercise program to reinforce musculoskeletal system of seniors appropriate for physical strength and conditions of seniors aged 65 years or above based on health Qigong exercise of oriental medicine. Method : Qigong exercise motions that can improve strength, muscular endurance, flexibility, and cardiovascular endurance of seniors were developed by primarily performing literature review of Qigong experts on the fields like oriental medicine, Qigong exercise, physical education and health science and secondarily using motion training for increased validity of motions. Results : The Qigong exercise program for musculoskeletal system of seniors was designed with 12 motions repeated 8 times and 20 minutes per session, including shoulder exercises (alternate turning of left and right shoulders, turning both arms back and forth, holding and lifting elbow to put it aside), waist exercises (wrapping the head with hands to bow, twisting waist while looking at the tip of hand, large spinning of ball, putting hands together to pull back), and knee exercises (going up a down while lifting a rock, balancing the body while lifting a knee, lifting and spreading knees while drawing circle with arms, raising both arms to the side while lifting heels, breathing). Conclusion : Once the effects of Qigong exercise for musculoskeletal system of seniors developed in this study are tested, the program is expected to contribute to development of Qigong exercise, a core part of oriental medicine health improvement project.

The purposes of this study were to investigate the patient satisfaction with physical therapy services and find the related factors to patient satisfaction. In this study, three hospitals were selected at large, middle, small city. Ninety subjects (53 males, 37 females) who had received physical therapy service were participated in this survey. Survey data were collected by a written questionnaire. The patient satisfaction scale had a good reliability (Cronbachs alpha=.9134). The collected data were analyzed by t-test, ANOVA, and Kruskal-Wallis. The scores of patient satisfaction with physical therapy services at large, middle, and small city were 79.67, 76.23, and 86.33 respectively. The score of patient satisfaction at small city was significantly higher than middle and large city(p<.01). There was no significant difference in patient satisfaction with physical therapy services according to gender, age, education years, occupation, marital status, religion and average monthly income. Further studies are needed to identify which specific factors are related to patient satisfaction with physical services. This information will be useful in improving the patient satisfaction.
생식반월 및 혈액 원시생식세포를 이용한 생식선 카이메라 닭 생산

This study was conducted to produce germline chimeric chicken using primordial germ cells (PGCs) collected from germinal crescent (GC) and blood vessel. Korean Ogol Chicken (KOC) PGCs were collected from GC (H&H stage 6-7) and blood vessel which contains circulating PGCs (H&H stage 1617), and were injected into 2.5-day-old recipient White Leghorn (WL) embryonic blood vessel (H&H stage 16-17). The hatchabilities of manipulated embryos which were injected GC and blood PGCs were 10.7% and 23.2% respectively. Fifty one among 192 chicks arrived at sexual maturity and were used for progeny test. The progeny test for WL(KOC) by mating KOC revealed that three black feathered chicks were hatched in the group injected with germinal crescent PGCs and one black feathered chick was produced in the group transferred with blood PGCs. The black feathered chicks must have originated from the injected KOC PGCs. This result indicated that PGCs of donor KOC embryos might migrate into embryonic gonads of recipient WL and differentiate into germ cells.
