        한국재래산양의 임신 및 분만전후의 혈중 Steroid Hormone 농도에 관한 연구

        The peripheral blood was collected from the jugular veins of Korean native goats during gestation and peripartum period. Three goats were selected as samples at every examination designed through the experimental period. The blood samples were centrifuged and stored at -20℃ until hormone assay. The peripheral serum concentration of progesterone was 0.13.03 ng/㎖ at the day of pregnancy and increased greatly to 11.42.44 ng/㎖ at 7 days of pregnancy and thereafter between 9.43 and 14.92 ng/㎖ until 140 days of pregnancy in these 3 goats with an equal gestation period of 145 days, and 8.01±1.67 ng/㎖ at 2 days before parturition, and then declined sharply to 4.60±0.15 ng/㎖, 0.96±0.08 ng/㎖, and (1.37±0.05 ng/㎖ at 1 days and 8 to 0 hours before partiurition and just after parturition, respectively. And this base level of peripheral progesterone concentration was maintained until the Final examination at 6 days after parturition. The peripheral serum concentration of estradiol failed to increase until mid-stage gestation, and thereafter it increased significantly (P$lt;0.05) to 130±25 pg/㎖ at 2 days before parturition and reached a peak of 370±29 pg/㎖ and then declined sharply at 12 hours after parturition. And the base level was maintained until the final examination at 6 days after parturition. There was found a great variation in the peripheral serum level of estradiol between the individuals throughout the gestation period. These results show that the corpus luteum pregnant goats maintain its function normally until 1 to 2 days before parturition and the estradiol may be synthesized in the placenta at increased rate during the late gestation, and its serum level reach to peak just prior to parturition.

        돼지의 번식형질에 대한 유전 및 환경적 효과 상호작용에 관한 연구

        This study was conducted to determine the main effects and interactions of breed, furrowing season and parity on swine reproductive traits: litter size at birth and weaning, pig and litter weight at birth, 21 days and weaning on the basis of the data obtained from 1047 litters of Berkshire and Landrace born from 1976 to 1980 at Sacheon Branch, National Livestock Station. The data were analysized by the least square method and results obtained are summarized as follows: 1. Landrace was heavier by 10% in litter weight at weaning, 17% in litter weight at 21 days, 10% in litter weight at birth, 23% in pig weight at 21 days than Berkshire. 2. The effect of breed for each litter weight and pig weight at 21 days was highly significant at 1% level. 3. The effect of furrowing year was highly significant for all the traits studied without any consistent pattern as the year passed. 4. The reproductive performance of the pigs born in the spring was significantly higher (p$lt;.001) than that in the fall except for the pig weight and litter size at weaning. 5. The litter size at birth or weaning was significantly larger (.1%) in the groups of the 2nd to 6th parity than in the other groups. All the body weight traits studied were significantly (.1%) superior in the 2nd 4th, the 5th - 6th, the 7th and over, and the 1st parity in that order. 6. The pig weight at weaning in Berkshire was significantly heavier in the spring-born group. The litter size at weaning and litter weight at birth in Landrace was significantly higher in the spring-born groups than in the fall-born ones. 7. The effect of breed x parity interaction was nonsignificant for the traits studied.

        닭의 주령 및 정액주입 간격이 수정률과 부화율에 미치는 영향

        This experiment was carried out to find out the effects of age of hens and insemination intervals on the fertility and hatchability of hens. A total of 1,230 older hens of PS Arbor Acres aged 64 weeks at initial stage and 225 younger hens of PS Arbor Acres aged 32 weeks at intial stage were used for the experiment. The results obtained were summarized as follows: The fertilities over the experimental period of older hens inseminated in the intervals of 3.5 and 7 day were 86.6±0.2, 83.7±0.7 and 74.9±0.8, respectively and those of younger hens were 92.8±0.4, 87.1±0.5 and 80.7±0.6, respectively. Hatchabilities of eggs from the younger hens were also significantly(P$lt;0.001) higher in all the groups of insemination intervals, compared with those from the older hens. The fertility of the hens of both age groups was not affected by the summer season. The fertility of younger hens increased significantly(P$lt;0.01) in August, compared to the fertility in May. In the group of hens inseminated repeatedly in the interval of 7 days, the fertility of hens was found to be highest in 2 to 3 days after insemination and it decreased significantly as the time passed. There was no significant decrease in fertility of hens in both age groups when the hens were inseminated once a week consecutively for experimental period of 91 days.

        Thanks to the recent technical development of embryo transfer, it has become possible to produce many good offsprings from the genetically highly productive dairy cattle in a short period, With the help of this technical development this study aims to analysis genetic change by use of multiple evulation and embryo transfer (MOST), Under the condition that the heritability of milk yield is 0.25, the number of donors per male is 20, and the number of embryo transfers per donor is 20, comparing their respective selection effects of dam I, dam II, dam I + dam II, and the females and males selected according to the genetic selection index, 1 have found that the annual genetic change of the group selected by the genetic selection index is the highest. And the genetic change is the highest when one male per half-sib is selected.

        생쥐 수정란의 분할조작후 생존성 향상에 관한 연구

        Demi-embryos were successfully produced by bisection of ICR mouse embryos at preimplantation stages. They were microsurgically bisected using a microsurgical blade attached to a micromanipulator after pretreatment with 0.5% pronase in PBS for two minutes or not. Embryos with softened zona pellucida were more easily bisected and less damaged than intact embryos. The highest success rate in bisection has been achieved by selecting blastocysts(94.1% in success rate with intact blastocysts and 100% in success rate with zona softened blastocysts). Demi-embryos without zona pellucida were cultured in D-PBS or M-16 medium at $37^{\circ}C$, 5% $CO_2$ in air for 72 hours for 2-cell stage embryos, 48 hours for 4-to 8-cell stage embryos, 24 hours for morula stage embryos and 6~12 hours for blastocyst stage embryos. For the in vitro culture of 2-cell stage embryos, $100{\mu}M$ 2Na-EDTA was added to the media. M-16 medium was better for the in vitro development of mouse embryos than PBS, and PBS is not considered to be suitable for long-term culture of embryos, especially at early stage of cleavage. In M-16 medium, developing rate of demi-embryos of which pair underwent development to form eublastocysts was 15.8% at 2-cell stage, 16.8% at 4-cell stage, 38% at 8-cell stage, 89.6% at morula stage and 94.4% at blastocyst stage, respectively. The more rapid and efficient production of demi-embryos and higher viability after bisection can be expected by softening zona pellucida with pronase and by selecting morulae or blastocysts rather than embryos at early stage of cleavage.

        한우의 조정기능발달에 관한 연구 1 . 한우와 교잡종의 전반기 고환발육과 조정기능

        In order to study on the development of the spermatogenic function and the sexual maturity between the Korean purebred and three-way crossbred ((Angus × Korean purebred) × Holstein), this experiment was conducted to investigate the testicular development and the spermatogenetic function by means of repeating biopsy at one month intervals from one to 9 months of age with 6 Korean purebreds and 3 three-way crossbreds. 1. In the testicular development, the scrotum circumference of the Korean purebred and three-way crossbred increased steadily with increasing age. The scrotum circumferences of the Korean purebred and three-way crossbred were 19.4 ㎝ and 28.9 ㎝ at 9 months of age, respectively, and doubled to one month of age. The testicular width and length increased rapidly from 6-7 months of age, but the differences between two groups were not statistically significant. 2. The seminiferous tubules in the testis of two groups, solid at 3 months of age, increased appearently in diameter at 5 to 6 months of age, but were not reached mature size. 3. Only gonocytes and indifferent supporting cells were present in the tubules at 3 months of age and spermatogonia appeared from 5 months of age, spermatocytes increased rapidly from 6 months of age and spermatid were seen for the first time in the 7 months old Korean purebred and three-way crossbred. On the basis of the above results it is concluded that sexual maturity of three-way cross bulls was somewhat earlier than the Korean bulls, but the difference of two groups was not significant.

        위임신 중 토끼 황체에서의 Estradiol 수용체 농도와 이에 대한 Tamoxifen 과 Estradiol -17β 의 영향

        The changes of cytoplasmic and nuclear concentration of estradiol receptor of corpus luteum were investigated on day 6, 9, 10, 12 and 13 of pseudopregnancy. And in order to determine the effects of tamoxifen and estradiol-17β, rabbits were injected subcutaneously with 500㎍ tamoxifen on day 9, and with 50㎍ of estradiol 17βon day 12 after priming of 3㎎ of indomethacin twice a day from day 10 to 11 of pseudopregnancy, respectively. The concentration of cytoplasmic estradiol receptor was significantly (P $lt;0.01) increased from 118.94 ± 7.19fM/㎎ of protein on day 6 to 140. 60 ± 8.14, 143.64 ± 11.15 and 144.26 ± 10.20fM/㎎ of protein on day 9, 10 and 12 of pseudopregnancy, respectively. But it was significantly (P $lt;0.01) decreased to 93.34 ± 4.49 fM/mg of protein on day 13 of pseudopregnancy. The translocation of estradiol receptor from cytoplasm to nucleus was significantly (P $lt; 0.01) accelerated from lhr. after injection of tamoxifen or estradiol-17β, but the rate of translocation by tamoxifen was significantly (P $lt;0.01) lower than that by estradiol-170. These results suggest that tamoxifen binds competetively with estradiol-17β to estradiol receptor of corpus luteum, but the affinity of tamoxifen to estradiol receptor is lower than that of estradiol-17β. The concentration of nuclear estradiol receptor was significantly (P $lt;0.01) increased from 0.35 ± 0.02 fM/㎍ of DNA on day 6 to 0.46 ± 0.02, 0.60 ± 0.02 and 0.62 ± 0.03fM/㎍ of DNA on day 9, 10 and 12 of pseudopregnancy, respectively. But it was significantly (P $lt;0.01) decreased to 0.20 ± 0.02 fM/㎍ of DNA on day 13 of pseudopregnancy. Significant (P $lt; 0.01) increase of nuclear estradiol receptor was induced by the translocation from cytoplasm to nucleus after injection of tamoxifen or estradiol-17β, and the replenishment of estradiol receptor from nucleus to cytoplasm was inhibited over 24hr. by tamoxifen injection.

        The change of the concentration of serum progesterone was investigated on day 6, 9, 10, 12 and 13 of pseudopregnancy. And in order to determine the effects of tamoxifen and estradiol-17β rabbits were injected subcutaneously with 500㎍ of tamoxifen on day 9, and with 50㎍ of estradiol-17β on day 12 after priming of 3㎎ of indomethacin twice a day from day 10 to 11 of pseudopregnancy, respectively. The concentration of serum progesterone was significantly(P$lt;0.01) increased from 6.55±0.63ng/ml on day 6 to 12.11±0.93, 17.20±0.98 and 16.62±1.19ng/ml on day 9, 10 and 12 of pseudopregnancy, respectively. But it was significantly(P$lt;0.01) decreased to 13.39±1.12ng/ml on day 13 of pseudopregnancy. Significant(P$lt;0.01) decrease of the concentration of serum progesterone was induced from 6hr. after tamoxifen injection in contrast to significant(P$lt;0.01) increase from 6hr. after estradiol-17β injection.

        Royal Jelly 가 생쥐의 발육에 미치는 효과

        This experiment was conducted to determine the effect of royal jelly on growth in mouse. Five or 10 ㎎ of royal jelly was injected to 8 male or female mice every two other day for 18 days. Body weigt gain, feed consumption and feed efficiency were measured in the mice for 4 weeks. Body weight gain of royal jelly treated lots was 10-20 percent more than than that of control lot, but it was not significant statistically. The difference in feed consumption way found insignificant between treatments, although male mice fed slightly more feed than female mice. No appreciable difference was found in feed efficiency between treatments.
