A study on detection rates of acid-fast bacteria in urine by three sampling methods was performed on 898 cases randomly sampled from peoples who visited Dr. Chung’s Health Lab. from January. to December, 1983. Positive rate by 5 times sampIing of morring urine was 7.0% and 12.5% higher than that of 24 hours sampIing. Therefore, 5 times sampling of morning urine method was considered to be desirable method for the test of ambulatory cases.

In classical titrimetric analyses, the major concern is the concentration of titrant, usually the aqueous solution of hydrochloric acid or sodium hydroxide, that could be changed as time goes by and it is accompanied with the inaccuracy of the resulting data. And the statistical approach, the nonlinear regression analysis, which is a well-known statistical method, was introduced to determine the accurate concentration of the titrant and the exact value of parameters, Ka, r, Ca, Cb, for 0.01 M aqueous solutions of analytes, sodium pyruvate, sodium acetate, sodium bicarbonate, ammonium hydroxide, ammonium chloride and acetic acid at 25℃. We used Gauss-Newton method for the linearlization of the nonlinear titration system and the two-parameter fitting showed appreciable convergent data for the parameters of the analytes set with the various range of Ka value.
Polyaniline, 1-Butylalcohol, Toluene을 함유하는 H₂O-CH₂Cl₂-H₂O Liquid Membrane을 이용한 아미노산의 선택적 분리

This paper studies amino acid separation using a H₂O-CH₂Cl₂-H₂O liquid membrane system. A CH₂Cl₂ layer is modified and 1-butylalcohol and toluene act as carriers. The solubility of amino acids to CH₂Cl₂. and Polyaniline is also introduced to examine the ability of 1-butlacohol and toluene as carriers. From an amino acid mixture containing GLy, LEu, ILe, VAl, CYs, PHe, HIs, ALa, GLn we successfully separated ALa, LEu, ILe, VAl. This result indicates that this optimized analytical condition obtained could be successfully applied to the qualitative analysis of the amino acids in human blood, urine, and other tissue samples using this liquid membrane system.
한국여성의 요중 아초산염과 백혈구 에스터라제의 활성 및 세균뇨에 대한 평가

The study evaluated the usefulness of the leukocyte esterase and nitrite reagent strip tests in a general hospital as a screening test to detect significant urinary tract Infections in urine of Korean women and correlated the fmdings with bacterial culture, leukocyte esterase and nitrite results. A total of 689 urine samples were reviewed. 254 positive cultures (36.8%) were found, of which 190 (74.8%) were detected by leukocyte esterase-nitrite test. In addition, 357 of 372 (96%) urine samples with negative bacterial cultures were negative in leukocyte esterase and nitrite tests. The results of studies was the sensitivity (74.8%), specificity (95.2%), positive predictive value (92.6%), and negative predictive value was still 84.7% and was comparable to that found in previous studies.

A bulk liquid membrane system was introduced for selective separation of an amino acid mixture. We confirmed p-diamethylaminobenzaldehyde (DAB), sulfosalicylic acid (SSA) and 1-naphtol were very useful carriers for selective separation of an amino acid mixture. As a result, Ala, Leu, Val, Phe and Ile were successfully separated by SSA, 1-naphtol in basic condition, 1-naphtol in weak acidic condition, DAB in strong acidic condition and DAB in strong basic condition. The separation mechanism was proposed by ion pair mechanism in the case of SSA and 1-naphtol and Imine bond formation mechanism was also introduced for DAB.

The detection of leukocyte levels played an important role in indication of such medical informations as urogenital tract infection or other dysfunction. The test strip made of 0.5% borate buffer pH8.0, 0.03% 2-pheny!-4(N-tosyl-alanyloxyI)-thiazole[ PTT] , 0.1 -0.8% PolyVinyl Pyrrolidone[PVP] , and 1% decanol was tested for not only reactivity, stability, variation of the number of leukocytes in urine. The detectable responses were shown at the different number of leukocytes and clinical usefulness.
요 중 백혈구를 측정하기 위한 새로운 진단 시험지 개발에 관한 연구

백혈구는 인체가 세균이나 바이러스에 감염되면 그 숫자가 증가하게되고 특히 요 중에 백혈구가 검출되면 비뇨기계나 생식기계의 감염을 의심하게 되어 이 계통의 질병을 진단하고 예후를 판단하는데 중요한 지표가 된다. 이에 본 연구에서는 백혈구내의 에스터라제의 활성을 측정함으로써 간접적으로 백혈구의 수를 측정하기 위하여 pentyl-3-thiophene-carboxlyate(PTC), pentyl-8-quinolinecarboxylate(PQC), 2-phenyl-4(N-tosylalanyloxyl)-thiazole(PTT)의 에스테르 기질을 합성하였고 각 기질을 이용하여 백혈구 검출용 시험지를 제조하여 요 중의 백혈구를 측정하였다. 이 세 가지 기질을 사용하여 백혈구 측정을 한 결과 0.5% borate 완충용액 pH 8.0, 기질 0.03%, 0.1∼0.8% PVP 안정제, 1% decanol의 구성비로 만들어진 PTT 시험지의 반응 색상이 다른 기질에 비교하여 안정하며 표준 비색표에 의한 육안적인 감별은 물론 요 화학 자동분석기에서의 응용성이 좋았다. A number of leukocytes increases when infected by a germ or virus. Detection of leukocyte levels can indicate of such medical informations as urogenital tract infection or other dysfunction. In this study, pentyl-3-thiophene-carboxlyate (PTC), pentyl-8-quinolinecarboxylate (PQC), and 2-Phenyl-4(N-tosyl-alanyloxyl)-thiazole (PTT) were synthesized, and the test strips were prepared with these substrates for quantifying leukocytes in urine. Among these substrates, the PTT test strip prepared in 0.5% borate buffer pH 8.0, 0.03% PTT, 0.1-0.8% PVP, and 1% decanol showed not only better color reaction but also an excellent application possibility to be used in automatic urine analyzer.
통상 요 분석 에서 Clini-Tek 200을 이 용한 비교연구

Routine urinalysis is screening test, many parts of laboratory duty. Therefore many cases of urine test needed very speedy and exactive method. The most of urinalysis performed by strip test, recently S-G strip test and WBC esterase strip test benefit of performing urine S-G and microscopic examination. Urine microscopic examination needed to trained observer and S-G test performed by r~fractometer, therefore routine urinalysis was uneasy. Clini-tek 200 operation performed with together other tests, increased objectivity in reading-time and results. The obtained the following results : 1. Cases of S-G test, clini-tek 200 was easy method, because performed together other tests, highest value cases ~ l, 030 and lowest value cases ~ l, 005 recheked rate 20.7% (74/357). 2. Ciini-tek 200 blood strip specifity was 82.9% (244/294) and sensitivity was 87.3% (55/63) . 3. Clini-tek 200 WBC strip specifity was 92.9% (275/296) and sensitivity was 73.7% (45/61). 4. The urine colored medicine or concentrated urine effected all strip tests, therefore we recheked strip test with chemical test. 5. When microscopic exam, changed to clini -tek 200 results, excepted microscopic exam. rate was about 34%. Clini-tek 200 results were negative, in microscopic exam. RBC positive case was 0. 3% (1/357), WBC positive cases were 1. 7% (6/357).

Basal metabolism is the basal or minimal amount of energy produced by an individual measured 12 to 16 hours after eating when the individual is as nearly as possible at complete mental and physical rest, but not asleep. It is the energy expended under basal conditions to maintain respiration, circulation, peristalsis, muscle tonus, body temperature, glandular activity and other vegetative functions. The basal metabolism is proportional to the surface area of the body and varies with the age and sex of the individual. The basal metabolic rate (BMR) is the percentage variation from the normal for an individual of the same surface area, age and sex. Mean values obtained .according to sex, age, body weight and height. The results obtained from 456 cases of male and female were as follows: 1) The mean value of HMR are 298 male was -8.9~+8.8 and 158 female ``was -5.5~+9.5 2) Below 29 years male was -8.3~+8.6 and female was -6.0~ + 7.1, 30 to 39 years male was -8.4~+8.1 and female was --5.0~+9.7, 40 to 49 years male was -8.2~ + 7.2 and female was -5.9~+10.4, 50 to 59 years male was -8.1~+9.2 and female was -3.5~ +8.8, above 60 years male was -6.2~+8.9 and female was -6.0~+7.1 by the age. 3) Below 49kg male was -6.5~+9.8 and female was -6.2~+9.5, 50 to 59kg male was -7.2~+8.6 and female was -4.1~+9.7, 60 to 69kg male was -8.7~+7.9 and female was -6.0~+10.2, 70 to 79kg male was -8.4~+6.9 and female was -6.0~+9.0, above 80kg male was -9.3~+ 10.0 by the body weight. 4) Below 149cm female was -1.3~+11. 4, 150 to 159cm male was -8.4~+7.4 and female was -5.7~+9.2, 160 to 169cm male was -8.1~+8.8 and female was -7.2~+6.5 by the height.
