        A Fluorescence-based cDNA-AFLP Method for Identification of Differentially Expressed Genes

        Identification of differently expressed genes under specific tissues and/or environments provides insights into the nature and underlying mechanisms of cellular processes. Although cDNA-AFLP (Amplified Fragment Length Polymorphism) is a powerful method for analyzing differentially expressed genes, its use has been limited to the requirement of radioactive isotope use and the difficulty of isolating the bands of interest from a gel. Here, we describe a modified method for cDNAAFLP that uses a fluorescence dye for detection and isolation of bands directly from a small size polyacrylamide gel. This method involves three steps: (i) preparation of cDNA templates, (ii) PCR amplification and differential display, and (iii) identification of differentially expressed genes. To demonstrate its utility and efficiency, differentially expressed genes during vegetative growth and appressorial development of Magnaporthe oryzae were analyzed. This method could be applied to compare gene expression profiles in a diverse array of organisms.

        벼도열병균의 비병원성 유전자 AVR-Pita1

        The rice blast fungus, Magnaporthe oryzae, is one of the most economically important crop diseases. In addition, rice-M. oryzae interaction is a classical gene-for-gene host-pathogen system. Race variation in pathogen groups was proposed as the main mechanism for rapid break-down of resistance in newly introduced rice cultivars. These new pathogen race variations may be caused by changes in an avirulence gene, such as (i) point mutations, (ii) insertion of transposons, and (iii) frame shifts. The avirulence gene AVR-Pita1 is representative avirulence gene in which all of these mutations are reported. In this review, we present a useful information for avirulence gene AVR-Pita1 and its homologous genes AVR-Pita2 and AVR-Pita3. We also review examples that cause mutations in these evolutionarily significant genes.

        Genetic Stability of Magnaporthe oryzae during Successive Passages through Rice Plants and on Artificial Medium

        Genetic instability of the rice blast fungus Magnaporthe oryzae has been suggested as a major factor underlying the rapid breakdown of host resistance in the field. However, little information is available on the mechanism of genetic instability. In this study, we assessed the stability of repetitive DNA elements and several key phenotypic traits important for pathogenesis after serially transferring two isolates though rice plants and an artificial medium. Using isolate 70-15, we obtained a total of 176 single-spore isolates from 10 successive rounds of culturing on artificial medium. Another 20isolates were obtained from germ tubes formed at the basal and apical cells of 10 three-celled conidia. Additionally,60 isolates were obtained from isolate KJ201after serial transfers through rice plants and an artificial medium. No apparent differences in phenotypes,including mycelial growth, conidial morphologies, conidiation,conidial germination, appressorium formation,and virulence, or in DNA fingerprints using MGR586,MAGGY, Pot2, LINE, MG-SINE and PWL2 as probes were observed among isolates from the same parent isolate. Southern hybridization and sequence analysis of two avirulence genes, AVR-Pita1 and AVR-Pikm, showed that both genes were also maintained stably during 10 successive generations on medium and plants. However,one reversible loss of restriction fragments was found in the telomere-linked helicase gene (TLH1) family,suggesting some telomere regions may be more unstable than the rest of the genome. Taken together, our results suggest that phenotype and genotype of M. oryzae isolates do not noticeably change, at least up to 10 successive generations on a cultural medium and in host plants.

      • 固腸丸 水性 엑기스의 家兎腸機能에 미치는 影響

        Effect of Gojanghwan(固腸丸) on intestinal function was investigated in rabbits, making use of its water extract. Gojanghwan water extract(GW) made the motility and tension of ileum strips isolated from rabbit decreased. While it did not affect on the acetylcholine action, GW exhibited the significant inhibition on the actions of serotonin and histamine to ileum strips. In the longitudinal muscle of colon, GW supressed the motility, but did not influenced significantly on the tension, and in the circular muscle of colon, GW showed the inhibitory effect on the motility and tension. GW inhibited markedly the actions (motility and tension)induced by acetylcholine serotonin and histamine in the longitudinal and circular muscles of colon. According to the above results suggest that GW inhibited the intestinal function in rabbit, this inhibition was related to the various mechanism being induced by responses on the receptor of muscarine, serotonin and histamine.

        마이욜의 조각에 나타난 모더니티의 구현 방식

        Maillol studied the form itself. He simplified the details and created the female figure that posses transcendent physiognomy through the non-active, symmetrical and frontal pose with simple, ample and smooth texture of the surface volume. This figure presents the primitivism found in Greek, Egypt, and Khmer culture. Maillol understood the female body as a pure object of visualizing the idea of the artist. He pursued the totality of the attitude in order to show the primitive spirit and the beauty of life. He concentrated the simple volume of the form into the geometric abstract space. Even with the figurative sculpture, Maillol opened the new era of modern sculpture with its simplified form as well as the contend approaching the sprite. Maillol made all monuments as a female no matter of its own sex. Besides, he destroyed the typical function of the monument by weakening the uniqueness of the character and causing the discordance between the form and the role via marcottage technique. He stressed that artist's emotion and originality come first by emphasizing on the form itself instead of instructive allegory and existence rather than external format. Maillol transferred the sculpture from its traditional function that is decorative, instructive and religious to the modern concept which contains inner mind or abstract thought of the artist. It proved the development of the sculptural art from the perceptual representation to the conceptual approach. Therefore, Maillol established the modernity by showing that the sculpture is a form of expressing the spirit and the philosophy of the artist within free visual language.

      • 자바프로그램의 보안 정보 흐름 분석

        자바 언어의 사용에 있어서 안전한 데이터의 관리는 중요한 요소가 된다. 자바 바이트코드 검증기 (Java bytecode Verifier)나 클래스 로더(class loader), 보안 관리자(Security Manager)를 통한 자바언어 자체의 보안장치로서는 플랫폼 독립을 지원하는 자바언어의 특성을 살리면서 동시에 민감한 정보의 유출을 방지하는 역할을 수행하기에는 부족한 점이 있다. 이러한 점을 고려하여 안전한 정보 흐름에 관한 연구들이 수행되어 왔다. 그중 자바 가상 기계의 바이트코드 검증기의 타입 검사기능을 이용한 방법이 연구되어 왔는데 본 논문에서는 구문분석정보를 그래프로 나타내어 보다 효율적인 방법으로 안전한 정보 흐름을 분석하고 그 결과를 시각화 하여 시스템 관리자가 보다 편리하게 사용할 수 있는 안전한 정보 흐름 분석기를 구현하였다.
