        에틸렌이 유도한 강낭콩 잎 키틴분해효소의 정제 및 특성

        Chitinase (EC 3.2.1.14), a potential pathogensis-related protein, was induced in leaves of 30-day-old bean (Phaseolus vulgaris) plants with treatment of 10 nl/ml ethylene for 30 hr. The enzyme was extracted in sodium citrate buffer (pH 5.0), and then purified by heat treatment, (NH₄)₂SO₄ precipitation, chitin-affinity and Sephadex G-75 chromatographies. Chitinase was enriched 22.5-fold with a yield of 29%. Its apparent molecular weight was estimated on SDS-PAGE to be 30 kD. Chitinase showed optimum pH 5.0 and broad pH stability, and showed thermal stability below 50℃. K_m and V_(max) values of the purified chitinase were 0.36 ㎎/㎖ and 0.7 nmole G1cNAc/min, respectively, for swollen chitin as a substrate. Chitinase was significantly inhibited by 10 mM Cu^(++), Hg^(++), S₂O₃^(--), S₂O₄^(--) or SO₃^(--) in reaction mixtures. Antibody was raised in rabbits against the chitinase and used for specific immunodetection of the enzyme.

        중국 현대시와 음악의 상관성 연구

        This study is about the Relationship between Poetry and Music in the modern Chinese poetry. Using a comparative literary study method, this research is based on the interrelationship between literature and other art, music. Many of the modern Chinese poems are sung as music. After the new culture movement during 1910s and 1920s, for instance, the representative works of colloquial Chinese poets such as Hushi(胡適), Liubannong(劉半農), Liudabai(劉大白), Xuzhimo(徐志摩), Daiwangshu(戴望舒) were written music by composers, Xiaoyoumei(蕭友梅), Zhaoyuanren(趙元任) and others. In 1930s, the various marches, military songs, anti-Japan revolutionary songs and ect. were created by the left revolutionary poets and musicians, and Maozedong(毛澤東)`s works were sung as hymns. In this paper, I found the examples that poetry was sung as a music and analyzed the characteristics. In the process that the modern Chinese poetry was changed into music, I searched to distinguish the types of poems dealing with love for men and women, criticism for reality, revolution, anti-Japan and etc. Through this analysis, I identify that poetry are related with music closely and the musical component is the essence of it.

        In situ hybridization에 의한 돼지 유행성 설사증의 국내발생 역추적 진단

        In this presented study, we established a method for diagnosis of porcine epidemic diarrhea(PED) by in situ hybridization(ISH), which made distinct progress in diagnostic pathology. We also carried out the retrospective diagnosis through ISH to assume the exact time of the first outbreak and incidence of PED in Korea. The outbreak of PED in Korea reported in 1992. However, since the end of 1980's, some researches of pig-industry have already suspected the outbreak of PED, not transmissible gastroenteritis(TGE). In this experiment, we performed the ISH using 80 formalin-fixed and paraffin-embedded tissues of porcine intestine which were requests for pathological diagnosis from 63 farms whose primary sign was diarrhea from 1984 to 1991. We prepared biotinylated cDNA probe(492base pairs) for ISH by nick translation method and carried out the ISH, using $Microprobe^{TM}$ capillary action system(Fisher $Biotech^R$). We detected PED virus in intestinal mucosa of 2 cases in 1992, 1 case in 1988, and 1 case in 1987. As a result, we assume that the outbreak of PED in Korea have already started since 1987.

        In situ RT-PCR 및 In situ hybridization 기법에 의한 닭 뉴캣슬병의 진단법 개발

        Newcastle disease (ND) is a highly contagious infection of poultry, Two pathology-based techniques, in situ RT-PCR and in situ hybridization (ISH) were applied to formalin-fixed, paraffin-embedded tissues from chickens naturally infected with velogenic ND virus (VNDV). Two pairs of primers and a probe for ISH and in situ RT-PCR, respectively, were selected from highly conserved region of matrix gene of NDV. The ISH experiment was carried out using MicroProbe$^{TM}$ capillary action system within 2 hours. In situ RT-PCR was performed using MicroProbe$^{TM}$ capillary action system and GeneAmp In Situ PCR system. With ISH and in situ RT-PCR, viral nucleic acid was detected in the central nervous system of chickens from infected with neurotropic velogenic Newcastle disease virus (NVNDV), whereas viral nucleic acid was detected in various organs or tissues of chickens from infected with viscerotropic velogenic Newcastle disease virus (VVNDV). In the NVND group, positive signals were characteristically defined in the cytoplasm of neuron, vascular endothelial cells, and perivascular mononuclear macrophages in the central nervous system. One of NVND group, chicken from one farm exhibited positive signals in the bronchial epithelium. The VVND group chickens showed positive reaction in the macrophages, vascular endothelium, and bronchiolar epithelium. Markedly, viral nucleic acid was detected in the macrophages of morphologically normal tissues which were peripheral or located in distant areas from lesions. The central nervous system of chickens infected with VVND virus had positive signals in the vascular endothelial cell, perivascular mononuclear macrophages and some neuron. The number and intensity of the positive cells by in situ RT-PCR were more and stronger, respectively, in comparison with those by ISH. Particularly, positive reaction was detected in macrophages infiltrating in cardiac muscle by in situ RT-PCR, but not obtained by ISH. Therefore, these results demonstrated that ISH is a rapid diagnostic method for detection of NDV and in situ RT-PCR can be used as an efficient method for detection of low viral load infection or subclinical viral infection of NDV.

        중국인 디아스포라 베이다오 北島의 시적 이미지 연구

        본고는 중국인 디아스포라 망명시인으로 불리는 베이다오 北島 시에 나타난 이미지를 연구한 논문이다. 베이다오는 2005년 8월 제2회 서울 국제문학포럼에 참여하며 국내의 독자들에게 널리 알려지고 있는 당대 중국의 저명한 시인이라고 할 수 있다. 그는 북경에서 출생하여 문화대혁명 때 홍위병 운동에도 참여했었고, 건설현장에서 일도 했던 경력을 소유하고 있다. 하지만 그는 1978년 『오늘 今天』이란 문학잡지를 출간하며 전통과 사회주의 현실주의 문학에 반대하는 전위적인 아방가르드 문학을 추구하였지만 반체제 시인으로 낙인찍혀 서구로 망명하여 지금은 미국에 망명중인 시인이다. 본고에서는 그의 시문학 세계에 대한 전체적인 조망 속에서 시인의 디아스포라적 인생역정을 바탕으로 시적 이미지의 변화과정을 연구해 보았다. 초기 시에는 주로 역사에 대한 회의와 반항, 그리고 영웅적 이미지가 「회답 回答」이란 시를 통해서 나타났으며, 서구로 망명한 이후의 시세계에서는 새로운 낯선 이국적 세계에 대한 발견과 향수의식이 주로 나타났다. 그리하여 그의 시에서는 고향과 조국에 대한 향수 이미지를 시적 형상화하며 자기정체성을 추구해 나가는 시적 변화과정이 잘 나타나고 있다. 필자는 베이다오 시의 이미지 변화과정을 통해 중국 당대시의 끼친 영향과 특징을 잘 이해해 볼 수 있었다. This is a thesis on the images that appear in the poems by Beidao (北島), who is known to be an exiled poet in the Chinese Diaspora. As he participated in the 2nd 'Seoul International Forum for Literature' held in August, 2005, he is widely known amongst Korean readers as one of the most famous Chinese poets of the contemporary period. Born in Beijing, he took part in the Red Guards during the Cultural Revolution and also worked as a construction worker. He published a literary magazine, entitled “Today” (今天) in 1978, to pursue avant-garde literature against the traditional Socialist realism. Branded as a dissident poet, however, he sought a political asylum in the West and is now living in exile in the US. In this study was examined the transformation process of the poetic images based upon his life in the Chinese Diaspora, in the light of his entire world of poetry. In his early poems, such as “Answer”, were represented his skepticism about History, rebellion and heroic images. Since his exile in the West, his works have shown some of his newly found discoveries in the new, unfamiliar and exotic world and his nostalgia for his own homeland. In his poems, therefore, is well represented a poetic transformation process that forms nostalgic images for his homeland and country and searches for his own identity. Through the transformation process of the poetic images by Beidao, I was able to understand his influences on the China's contemporary poetry and its characteristics.

      • 유식세포 분석법에 의한 진도개 전파성 성기육종의 DNA Ploidy 유형분석

        Transmissible venereal tumor(TVT) is a naturally occurring contagious neoplasm which can be transmitted by mechanical contact during mating in dogs and transplanted as intact viable cells to dogs and other members of canine family such as coyotes, jackals, wolves, and foxes. The incidence of this tumors tends to increase in Korean native Jindo dogs. This is probably due to the high density and unrestrained management system. With time, TVT reaches the maximum size and then tends to regress spontaneously unless individuals are immunologically compromised. It consists of different types of cells depending on the stage. In this study, 10 tumors were selected from Jindo dogs. These were histologically calssified into three stages; progressive, steady-state, and regressive. Mitotic figures were counted, and their histological appearance at each stage is compared with their DNA ploidy. Histologically, 5 tumor cases were calssed as the progressors, 3 cases as the steady-state tumors, and 2 cases as regressors. Progressors were composed of round cells with large nuclei containing conspicuous nucleoli and frequent mitotic figures. A few spindle-shaped cells and inflammatory cells including mainly lymphocytes, a few neutrophils and macrophages were also seen. In the steady-state tumors, there was an increased number of spindle shaped cells and mitotic figures were rare. Six tumors were diploid and four were aneuploid with the variation coefficient of 7.02. Two of five progressive tumors were aneuploid. Two of three steady-state tumors were aneuploid while both tumors at the regressive stage were diploid. Progressive and steady-state tumors had a much larger S/G2M fraction and a higher mitotic index than regressive tumors. Two tumors which persisted for more than one year were aneuploid. These results suggest that the progressive and steady-state tumors had more active cell division than the regressive neoplasms.
