        PromoterWizard: An Integrated Promoter Prediction Program Using Hybrid Methods

        Promoter prediction is a very important problem and is closely related to the main problems of bioinformatics such as the construction of gene regulatory networks and gene function annotation. In this context, we developed an integrated promoter prediction program using hybrid methods, PromoterWizard, which can be employed to detect the core promoter region and the transcription start site (TSS) in vertebrate genomic DNA sequences,an issue of obvious importance for genome annotation efforts. PromoterWizard consists of three main modules and two auxiliary modules. The three main modules include CDRM (Composite Dependency Reflecting Model) module, SVM (Support Vector Machine) module, and ICM (Interpolated Context Model)module. The two auxiliary modules are CpG Island Detector and GCPlot that may contribute to improving the predictive accuracy of the three main modules and facilitating human curator to decide on the final annotation.

        분자생물학에서 컴퓨터는 많은 수의 분자배열을 짧은 시간 내에 비교 정렬하는데 이용되어 왔다. 이를 위해, 다른 접근방법들과 몇 가지 점에서 차이를 보이는 새로운 알고리즘이 제안되었다. MAlign이라 명명된 이 알고리즘은 비교 배열들간의 동시 비교를 효과적으로 수행할 수 있는 방법을 도입, 전체적 상동성을 검색할 수 있게 고안되었다. 동시 비교성의 결여는 기존의 알고리즘들이 갖는 문제점 중의 하나이었는데 이것을 consensus 또는 compacted sequence를 비교 과정에 도입함으로써 해결하였으며, 상동성 벡터 (homology vector) 개념을 적용하여 각 중간 배열들의 상동성 값을 벡터전사의 과정으로 비교함으로써 전체적인 비교를 용이하게 하였다. 몇몇 실험결과로부터, 다중배열 비교에 있어서는 일대일 대응 배열에서 나타난 높은 상동성값이 유지되는 것과 상동성이 높아질수록 이러한 결과가 더욱 두드러짐을 알 수 있었다. 이러한 정근방법을 이용하여 Copper 결합 단백질들과 세균의 신호 발생 단백질들에 대한 다중정렬 결과가 제시되었다. One application of computer in molecular biology has been the use of its power for comparing a large number of molecular sequences in very short time. For this purpose, a new algorithm is proposed which differs in several aspects from other approaches. Our algorithm, called MAlign, is designed to seek global homology by introducing an effective way to make simultaneous comparisons among test sequences. One problem in previous algorithms which were limited in its ability to compare sequences simultaneously has been solved by introducing intermediate consensus or compacted sequences and including them for comparison. In addition, a homology vector concept was applied to provide uniform representation for each intermediate, which makes global comparison easier. Several test results indicate that high homology values obtained from pairwise alignment are maintained after multiple alignment of those sequences, which is more apparent in higher homology values. Sample alignment results using this approach for three different copper binding proteins as well as bacterial signaling proteins are presented.

        국내의 유전체 연구도 2000년을 시작으로 본격화되었으며, 미생물의유전체 서열 결정을 수주 이내에 완료할 수 있는 시설을 갖추고 있는 벤처기업도 이미 등장하였다. 한편, 이들 과제의 산물인유전체정보로부터 유용한 학문적 결과와 산업적 결과를 얻기 위해 유전체후속(post-genome) 프로젝트들이 최근에 진행되고 있다.

        Investigation of Live Load Defl ection Limit for Steel Cable Stayed and Suspension Bridges

        Long span bridges such as steel cable stayed and suspension bridges are usually more fl exible than short to medium span bridges and expected to have large deformations. Defl ections due to live load for long span bridges are important since it controls the overall heights of the bridge for securing the clearance under the bridge and serviceability for securing the comfort of passengers or pedestrians. In case of sea-crossing bridges, the clearance of bridges is determined considering the height of the ship master from the surface of the water, the trim of the ship, the psychological free space, the tide height, and live load defl ection. In the design of bridges, live load defl ection is limited to a certain value to minimize the vibrations. However, there are not much studies that consider the live load defl ection and its eff ects for long span bridges. The purpose of this study is to investigate the suitability of live load defl ection limit and its actual eff ects on serviceability of bridges for steel cable-stayed and suspension bridges. Analytical study is performed to calculate the natural frequencies and defl ections by design live load. Results are compared with various design limits and related studies by Barker et al. (2011) and Saadeghvaziri et al. (2012). Two long span bridges are selected for the case study, Yi Sun-Sin grand bridge (suspension bridge, main span length = 1545 m) and Young-Hung grand bridge (cable stayed bridge, main span length = 240 m). Long-term measured defl ection data by GNSS system are collected from Yi Sun-Sin grand bridge and compared with the theoretical values. Probability of exceedance against various defl ection limits are calculated from probability distribution of 10-min maximum defl ection. The results of the study on the limitation of live load defl ection are expected to be useful reference for the design, the proper planning and defl ection review of the long span bridges around the world.

        수평관에서 프로판, 이소부탄, HFC134a를 포함한혼합냉매의 풀비등 열전달계수

        Nucleate pool boiling heat transfer coefficients(HTCs) were measured with one nonazeotropic mixture of Propane/Isobutane and two azeotropic mixtures of HFC134a/Isobutane and Propane/HFC134a. All data were taken at the liquid pool temperature of 7℃ on a horizontal plain tube with heat fluxes of 10kW/m2 to 80kW/m2 with an interval of 10kW/m2 in the decreasing order of heat flux. The measurements were made through electrical heating by a cartridge heater. The nonazeotropic mixture of Propane/Isobutane showed a reduction of HTCs as much as 41% from the ideal values. The azeotropic mixtures of HFC134a/Isobutane and Propane/HFC134a showed a reduction of HTCs as much as 44% from the ideal values at compositions other than azeotropic compositions. At azeotropic compositions, however, the HTCs were even higher than the ideal values due to the increase in the vapor pressure. For all mixtures, the reduction in heat transfer was greater with a larger gliding temperature difference. Stephan and Korner's and Jung et al's correlations predicted the HTCs of mixtures with a mean deviation of 11%. The largest mean deviation occurred at the azeotropic compositions of HFC134a/Isobutane and Propane/HFC134a.
