This study was desinged to investigate the early changes of submandibular gland in diabetic rats induced with alloxan monohydrate. For this study, 45 Sprague-Dawley rats (mean body weight of IOOg) were used as the experimental animals, and divided into two experimental groups as follows. Control group, Diabetic group induced alloxan monohydrate (185mg/kg). The experimental animals were sacrificed at 12 hours, 1, 3 and 7 days after the experiments, by vascular perfusion with 2.5% glutaraldehyde fixative solution. Their submandibular glands were removed, and prefixed with 2.5% glutaraldehyde for three hours. The specimens were rinsed with phosphate buffer solution (pH 7.2-7.4) and postfixed with 1% osmium tetroxide at 4°C for 2 hours. Thereafter the specimens were dehydrated within alcohol series, cleared with prophylene oxide and embedded within Epon 812 resin. Thin sections were made, and stained with 1% toluidine blue. The submandibular glands were observed under light microscope. The results were as follows; 1. Submandibular gland under diabetes mellitus was increasingly changed in ductal tissues comparable to acinar tissues in early stage. 2. The proportion of area occupying acinar and ductal tissues of submandibular gland under diabetes mellitus was decreased. 3. A intercalated ductal tissues of submandibular gland under diabetes mellitus were gradually, atrophied with time, but striated ductal tissues were expanded. 4. In striated ductal tissues of submandibular gland"under diabetes mellitus, dense-rounded bodies were gradually accumulated with time.
