RP-HPLC에서 α-Hydroxy Carboxylic Acid류의 머무름거동에 관한 연구

This study is an investigation of the effect of organic modifier volume in mobile phase, temperature and pH on the retention behavior in RP-HPLC. The retention data, capacity factors(k') of ten α-hydroxy acids were measured in the mobile phase with various compostion of methanol-phosphate buffer solution(v/v) at the same time that pH value was adjusted to 2.4, 4.4 and 6.4 respectively, and regulated the temperature from 10°C to 50°C with using c18 column. As expected, the decrease in retention time was mostly obtained as higher pH values than pKa values of solutes. That is explained by the increasing concentration of the ionized species when pH increase, but some of them did not coincide with that tendency. The effect of temperature on retention was investigated by the linerity of van't Hoff plot (R〉0.900), on the whole, at a lower volume fraction of organic modifier. And hydrophobicity index, log kw' obtained with experimental and the theoretical method showed an excellent correlation. Maximum retention time of these solutes is Mostly no more than double that of dead time, therefore another hydrophobicity index φ0 values were determined and discussed in several solute system.
