돼지 난포란의 체외 성숙시 성선 자극 호르몬의 첨자가 난자 성숙에 미치는 영향

This study was carried out to investigate the effects of gonadotropins added during in vitro maturation of porcine oocytes. The follicular oocytes were cultured in TCM-199 medium and Ham's F-10 medium containing different combination of gonadotropins. The results of emerging rate of germinal vesicle(GV), germinal vesicle breakdown(GVBD), metaphase-I(M-I) and metaphase-II(M-II) stages by 1% aceto-orcein staining and Hoechst 33342 DNA labelling were as follows. The oocyte maturity were observed in the emerging rate of GVBD and M-II stages after cultures in media hormones added. The optimal concentrations for oocytes maturity with a single treatment of FSH, hCG and LH were indicated to 0.5㎍/mi, 2IU/ml and 2IU/ml, respectively. The rates of GVBD and M-II stages were showed higher in that order LH(62.7%), hCG(51.7%) and FSH(35.3%). When the oocytes were matured for 44hrs in the medium containing of FSH+hCG, FSH+LH, hCG+LH and each treatment of the hormones, the maturity rates were figured in 70%, 63.3%, 58.6% and same 60% for each. Finally, in comparison wth the oocytes maturity by the emerging rate between TCM-199 medium and Ham's F-10, the former (49.5%) is insignificantly a similar tendency to the latter(48.5%). These results demonstrate that the addition of gonadotropins to the culture medium has a significant influence on the rate at which porcine oocytes undergo maturation.
