There have been a lot of problems unsolved on the survival and functional stability of genetically engineered microorganisms (GEMs), when they are released into the environments for any purposes of function. In this study, three GEM strains constructed from 4-chlorobiphenyl (4CB)-degrading Pseudomonas sp. DJ-12 were examined for their survival and degradation stability of 4-hydroxybiphenyl by exposing them in sterile distilled water (SDW), filtered autoclaved river water (FAW), and filtered river water (FW), separately. The GEM strains showed no difference among themselves in survival and degradation stability during 30 days exposed in any waters, but their viabilities were appeared in order SDW>FAW>FW. The recombinant plasmids harboring in the GEM strains were relatively stable during 30 days exposed in SDW and FAW, but they began to be destroyed after 10-day exposure in FW. These results indicate that non-sterile FW has some biotic factors affecting survival and functional activity of the GEM strains. The most cells of the GEM strains survived in the waters retained degradation activity of 4-hydroxybiphenyl. Therefore, the GEM strains examined in this study showed high stability by themselves in their survival and degradation function during exposure in the water environments.
