        消癌散의 항암효과 및 血管新生抑制에 미치는 영향

        Cancer, which is expressed in various forms, is one of the leading causes of human death. Soamsan (SAS) is composed of ten medicinal herb, the prescription was made according to the principles of Oriental traditional medicine based on the concept of synergic effects and interaction of among the components. SAS has been used for the cancer therapy, but the mechanism of it's effect is not well known. In the present study, the cytotoxic effect of the SAS water extract on cancer cell lines was investigated by the method of MTT in A549 cell lines and the anti-angiogenic effect was shown in the assay of chorioallantoic membrane (CAM) and in the cornea of rat administerd orally with SAS water extraction. The viability of A549 cell lines was not affected by the whole extract of SAS but the n-Hexan fraction of SAS water extract showed strong cytotoxicity which was not seemed to be done by the apoptotic mechanism. SAS water extract showed inhibition effects of angiogenesis induced in the cornea of rat and CAM assay. As the above results, it is suggested that SAS can be a candidate for new prescription for cancer therapy.

        蘇木의 에틸아세테이트 추출물이 B16/F10 흑색종세포의 멜라닌 합성에 미치는 효과

        Caesalpinia sappan L. has been commonly used as analgesic, a cure for contusion and sprain as well as a remedy for thrombosis in the oriental medicine. The main constituent of Caesalpinia sappan L. is brazilin having a flavonoid structure. Melanin is a main pigment found in skin, hair and eyes, and melanin synthesis is controlled by a cascade of enzymatic reactions regulated at the level of tyrosinase. The aim of this study was to examine the effect of Caesalpinia sappan L. on proliferation and melanogenesis of B16/F10 melanoma cells. After 48h treatment of cells with various concentrations of EtOAc extract, the cells exhibited a dose-dependent inhibition in their proliferation without apotosis. Therefore, the growth retardation may be due to the cell arrest, not due to cytotoxicity. And there were estimated total melanin contents as a final product and activity of tyrosinase, a key enzyme, in melanogenesis. EtOAc extract increased the melanin contents and tyrosinase activity in low concentration (0.01-10㎍/ml), whereas it decreased those in high concentration. In conclusion, it was observed that EtOAc extract of Caesalpinia sappan L. differently regulates melanization of cells dependent of its concentrations.

        白朮抽出液이 멜라닌 生成에 미치는 影響

        皮膚의 색깔은 여러 因子에 의하여 決定되는데 특히 멜라닌은 表皮에서 特殊하게 分化된 멜라닌 細胞에서 合性이 되어 피부의 색을 결정하는 중요한 인자가 된다. 韓醫學에서의 皮膚는 身體內部의 臟器와 밀접한 關聯이 있는데 특히 肺와 밀접한 聯關을 가지고 있으며, 白斑症과 같은 皮膚色素 缺乏이나 皮膚의 美白效果를 나타낼 수 있는 藥材들이 많이 소개되어져 있다. 따라서 본 硏究에 있어서는 現在 皮膚疾患에 應用될 수 있는 韓藥材를 검색하고자 白朮 抽出物로부터 멜라닌 生成 抑制 效果를 確認하였다. 白朮의 抽出液이 皮膚의 멜라닌 色素 形成에 어떠한 影響을 나타내는지를 糾明하기 위하여 먼저 MTT assay를 실시하여 처리농도를 결정하고 실제 melanogenesis에 대한 影響을 判斷할 수 있는 最終産物인 melanin의 定量測定을 施行하였으며, 아울러 Tyrosinase 遺傳子의 발현에 대한 影響과, 멜라닌 合成의 기전 중 JNK(c-Jun N-terminal kinase) 및 전사활성인자(transcriptional activator)인 AP-1 의 活性 抑制를 測定하였던 바 白朮 抽出液은 B16 mouse 흑색종 細胞의 色素生成을 抑制시켰고, α-MSH에 의해 活性化 되는 JNK活性도 다소 抑制시켰으며, AP-1과 같은 전사활성인자는 不活性化됨을 나타내어 白朮에 의한 멜라닌 細胞의 멜라닌화에 대한 이러한 抑制 作用이 認定되었다. Recently many efforts are focused to understand the mechanical insights of melanogenesis to develop the therapeutic agents for hyper-and hypo-pigmentation. This work is designed to evaluate the effciencies of Rhizoma Atractylodis macrocephalae, which has been used against hyper-pigmentation for many decades. Rhizoma Atractylodis macrocephalae alone decreases the tyrosinase promoter activity of B16 mouse melanoma cells in a dose-dependent manner. Pretreatment of the cells with Rhizoma Atractylodis macrocephalae also markedly suppresses the increase of α-MSH(10 nM)-induced melanogenesis as well as tyrosinase promoter activity at 10㎍/ml. Parallel with these results, α-MSH significantly increases the phosphotransferase activity of cJun N-terminal kinase1 (JNK1)/stress activated protein kinase (SAPK) (1.5 fold), which is returned to control level by pretreatment of Rhizoma Atractylodis macrocephalae. In addition, pretreated Rhizoma Atractylodis macrocephalae profoundly inhibits the ability of the nuclear proteins from B16 cells to bind with AP-1 nucleotide binding consensus sequences. These results indicates that Rhizoma Atractylodis macrocephalae inhibits α-MSH induced melanogenesis of B16 mouse melanoma cells via suppression of phosphotransferase activity of JNK1 and transcriptional activation of AP-1.
