It was attempted to observe the effect of garlic on the hepatic purine metabolizing enzymes in this study. The activities of adenosine deaminase, guanase, and uricase in rats were not changed significantly following the feeding of 5% garlic juice. Whereas, garlic juice inhibited significantly the hepatic xanthine oxidase activity compared to control group with the lapse of treated-period. The urate level of serum in rats was significantly decreased by the treatment of garlic juice. The above inhibitory effect of garlic was greater in boiled garlic juice than fresh garlic juice-treated group. These results indicated that, according to the chemical properties of allicin which is unstable in heat, other components than allicin in garlic may regulate the hepatic purine metabolizing enzymes.

간 aldehyde oxidase 활성에 마늘 성분이 어떼한 영향을 주는가를 관찰할 목적으로 in vivo 및 in vitro 실험을 하였다. 마늘 수침액을 mouse에 섭취케 하였을때 간 aldehyde oxidase의 활성은 유의성 있게 감소하였다. 이와같은 마늘성분에의한 aldehyde oxidase 활성억제작용이 어떤 성분에 기인된 것인지를 확인하기 위하여 마늘로부터 추출한 allicin분획과 allicin을 열분해시킨 분획 및 butanol분획을 이용하여 토끼간으로부터 정제한 aldehyde oxidase의 활성을 비교 관찰 하였을때 3분획 모두 본 효소의 활성을 현저히 억제시켰으며 이와 같은 억제작용은 열처리한 allicin분획에서 더 강하게 나타났다. 한편 본 효소와 기질간의 반응속도에 마늘성분이 어떠한 영향을 미치는가를 검토하였을때 상기 3분획 모두 $V_{max}$에는 영향이 없었으나 $K_m$치를 증가시키는 경쟁적 저해현상을 나타내었다. When pyrazinamide is used in the treatment of tuberculosis, the measurement of pyrazinoic acid which is an intermediate metabolite of pyrazinamide in body is required in order to prevent its associated side-effects, especially that of hyperuricemia. Effects of vitamin B$_6$ on pyrazinoic acid metabolism were studied in this experiment. The activity of hepatic pyrazinoic acid oxidizing enzyme in the presence of pyridoxal was powerfully inhibited, and the pattern was competitive inhibition type. Whereas, its enzyme activity was significantly increased by the treatment of pyridoxal, and the characteristics of the increase may include induction of enzyme proteins. As mice received pyrazinoic acid(300mg/kg) after pyridoxal-pretreatment(40mg/kg) once daily for 4 days, the blood level of pyrazinoic acid and uric acid was decreased significantly.

Pyrazinamide, an amide of pyrazinoic acid, is widely used in combination with other drugs for the treatment of tuberculosis. It was attempted to observe the effect of pyridoxal on the pyrazinamide induce hyperuricemia in this study. It was observed that the values of serum transminases were not changed in mice injected pyrazinamide and pyrazinoic acid, respectively (100, 200, and 300mg/kg) compared with control. Blood urate levels were increased in mice treated with these drugs in a dose dependent manner. After pyrazinoic acid was administered intraperitoneally at a dose of 300mg/kg pretreated with 50mg/kg of pyridoxal once daily for 4 days, the blood levels of uric acid and pyrazinoic acid were decreased significantly.

Alcohol 性 肝 損傷에 미치는 人蔘의 影響 : Alcohol 投與後 血淸 isocitrate dehydrogenase 活性에 미치는 人蔘의 影響

The purpose of this study was to investgate the effect of Panax Ginseng on serum isocitrate dehydrogenase activity in kreb's cycle after ethanol consumption. Ginseng butanol extract significantly increased the serum isocitrate dehydrogenase activity which is inhibited by ethanol treatment in mouse. It was observed that ethanol did not change directly the serum isocitrate dehlrdrogenase activity in vitro test. Ginseng butanol fraction increased the enzyme activity according to ginseng dose-dependent in low concentration, while high concentration of ginseng extract showed inhibition effect in vitro test. It was also observed that ginseng butanol fraction decreased km value of serum isocitrate dehydrogenase. These results suggest that some components of ginseng regulate the serum isocitrate dehydrogenase activity which is induced by ethanol consumption.
마늘 成分이 Xanthine Oxidase 活性에 미치는 影響

Allium sativum L. has been used as a food, drug, spice and preservative from ancient times. A dose, 1/kg of ethanol produced experimental hyperuricemia in rat. Allium sativumt.L were tested for their ability to alter the hepatic xanthine oxiBase activity and the blood level of uric acid in the ethanol·treated rat. Treatment of allium sativum L. 1% and 5% for 90 days markedly decreased the xan-thine oxidase activity in normal rat liver. When 1% of allium sativum L. treated for 5days and 10 days, markedly decreased the xanthine oxidase activity in the ethanol-treated rat liver. It was also observed that allium sativum L. reduced the blood concentration of uric acid in experimentally induced hyperuricemia by alcohol treatment. In vitro, it was found that a low concentration of allium sativum L. in the reaction mixture increased the hepatic xanthine oxidase activity, while in a high concentration was not changes of xanthine oxidase activity of normal rat. These results suggest that allium sativum L. may be some therafeutic effect on theohyperuricemia syndrome.
Testosterone에 의한 승홍의 신독성 유발에 의한 연구

Mercuric chloride, inorganic compound, is one of the most important drugs that has been used in the field of argriculture, antisyphilitica and anticeptics, but it is not used clinically at present. We have studied the effect of testosterone on the mercuric chlorideinduced nephrotoxicity. Renal lipid peroxide concentration of male rat treated with mercuric chloride was significantly increased in comparison with that of the female rat, it showed similar effects on testosterone pretreatment. Changes in renal catalase and glutathione peroxidase activities were not siginificantly different in testosterone-treated groups. But, renal xanthine oxidase and aldehyde oxidase activities of testrosterone-treated group given mercuric chloride significantly increased in comparison with that of the testosterone-treated alone. Animals treated with testosterone prior to mercuric chloride showed more severe damage on histological observations than those treated with testosterone only. Consequently, we suggest that the mercuric chloride-induced nephrotoxicity might be renal lipid peroxide generating enzyme system by testosterone.
