        Clostridium acetobutylicum 무포자 변이주와 야생주의 Protonmotive force 와 용매 생산의 상관 관계

        Wild strain and asporogenous mutant of Clostridium acetobutylicum were used in a study to investigate the effect of organic acids on the maintenance of protonmotive force and on solventogenesis. In the presence of 50 mM acetate both of the strains maintained the protonmotive force of-174--232 mV and the internal pH of 5.46-6.52. The protonmotive force determination in the presence of 25 mM acetate and 25 mM butyrate showed that normal magnitude of protonmotive force was maintained by the acidogenic culture of the mutant and by the wild strain. On the other hand the solventogenic culture of mutant strain maintained the protonmotive force of -135--148 mV in the presence of 25 mM acetate and 25 mM butyrate. From these results it is concluded that the higher solventogenic ability of the mutant strain is due to the increased permeability of butyrate into the cell.

        일차산물 부티르산이 Clostridim acetobutylicum 의 Protonmotive force 에 미치는 영향

        In order to understand the acid tolerance mechanism in Clostridium acetobutylicum against the undissociated weak acids, a two stage continuous culture of C. acetobutylicum KCTC 1037 was made to supply acidogenic and solventogenic phase culture for a study on the protonmotive force and fermentation profile in the presence of butyrate, the acidic fermentation product. Undissociated butyric acid was permeable to the membrane of acidogenic cell, while the influx of the acid was minimal into the solventogenic cell. Acidogenic cell had higher pH gradient and protonmotive force than solventogenic cell. The protonmotive forces decreased linearly with the increase in butyrate from 25 to 75 mM. Acidogenic culture maintained the internal pH of 5.6-6.2 in the presence of 75 mM butyrate, and the internal pH of the solventogenic cultures was 5.3-5.7 in the similar conditions. Though the acidogenic culture was found to maintain the protonmotive force in the presence of 75 mM butyrate in the short term experiment, a long term incubation showed that the acidogenic culture was unable to ferment sugar in the similar conditons. On the other hand the solventogenic culture maintained active protonmotive force and fermented sugar in the presence of 75 mM butyrate. The protonmotive force of acidogenic culture was dissipated by 75 mM butyrate in the presence of ATPase inhibitor, dicyclohexylcarbodiimide, whilst the protonmotive force of solventogenic culture was maintained a similar condition. From these results, it is concluded that the acidification of cytoplasm by butyric acid is prevented by proton pumping action of the membrane bound ATPase which maintains the protonmotive force for a short-term period in the acidogenic cultures, and that solventogenic culture possess the ability to reduce the influx of organic acid in long term fermentation. It is suggested that C. acetobutylicum maintains the protonmotive force by the investment of ATP during the fast-fermenting acidogenesis; on the other hand, the solventogenic culture can reduce the influx of the acidic fermentation products to maintain the protonmotive force.

        해리되지 않은 분자상의 유기산에 대한 Clostridium acetobutylicum의 내산성 특징을 연구하기 위하여, C. acetobutylicum KCTC 1037과 그 균주의 용매 생성능이 향상된 변이주인 C. acetobutylicum KCTC 1724의 protonmotive foroe를 측정하였다 C. acetobutylicum의 protonmotive force는 50 mM의 아세트산이 있을 때 산 생산기 세포들은 -189~-232 mV, 용매 생산기 세포들은 -174~-204mV로 유지되었다. 25 mM의 아세트산과 부티르산이 있을 때 용매 생산기 세포들의 protonmotive force는 -135~-163 mV로 낮아졌는데 변이주 KCTC 1724의 pH gradient는 야생주 보다 더욱 감소하여, 부티르산의 투과성이 야생주보다 높은 것으로 측정되었다. 변이주에서 용매 생산성이 높은 것은 용매생산의 전구물질이 되는 유기산이 세포안으로 쉽게 들어갈 수 있기 때문으로 생각된다.
