범가자미, Verasper variegatus 수컷에서 estradiol-17β에 의해 유도된 vitellogenin의 면역학적 특성

Vitellogenin(Vg) was purified from plasma of estradiol-17β(E₂)-treated spotted flounder, Verasper variegatus, by precipitation with cold distilled water, followed by fractionation using a Sepharose CL-6B column chromatography. E₂-induced protein was identified as Vg by SDS-PAGE and western blot analysis. The molecular weight of the purified Vg was estimated 175kD as determined by SDS-PAGE. In order to measure the Vg level, monoclonal antibodies against Vg were produced by hybridoma technique. Purified Vg was immunized into Balb/c mice and then the spleen cells from mice were fused with NS-1 myeloma cells. The hybridoma cells were screened by enzyme-linked immunosorbent assay (ELISA) and subcloned by limiting dilution. The hybridoma clones which secreted antibodies highly reactive to the purified Vg were designated as 4D6. Its specificity was demonstrated by western blot from plasma of untreated, E₂-treated male fish, and purified Vg.
넙치 전 암컷 집단의 생산을 위한 연구 IV. 성분화 기간 중 수온에 의한 성 결정

사육 수온이 넙치의 성비에 미치는 영향을 조사하기 위해 일반 넙치 및 자성발생성 2배체 넘치 자어를 대상으로 부화 후 35일(전장 1.2 cm)부터 100일(전장 12 cm)까지 사육 수온 18, 21, 24 및 $27^{\circ}C$로 나누어 2회 반복하여 실험하였다. 사육 수온이 높을수록 암컷의 비율은 낮게 나타나 사육 수온과 암컷 비율에 있어 역상관관계를 보였다. 자성발생성 2배체 실험군의 경우 $18^{\circ}C$에서 $94.4\%$의 암컷을 보인 반면 $27^{\circ}C$에서는 $57.6\%$로 나타나 암수비가 1 : 1이었다(P<0.01). 사육 수온에 따른 생존율은 일반 넙치 및 자성발생성 2배체 넙치 모두 18, 21, 24 및 $27^{\circ}C$ 순으로 높았으나 $27^{\circ}C$를 제외한 나머지 실험군은 유의차가 없었으며 $18^{\circ}C$를 제외한 나머지 실험군의 체중 성장은 유의차가 없었다(P>0.05). We investigated the effects of rearing water temperatures during sex differentiation period on sex ratios in olive flounder, Paralichthys olivaceus. Control and gynogenetic diploid juvenile flounder reared at water temperature of 18, 21, 24 and $27^{\circ}C$ for 65 days from 35 to 100 days after hatching. Fish were sampled to examine sex ratios at 195 or 260 days after hatching. Female ratios of control and gynogenetic diploid flounder declined rapidly as water temperature increased. Sex ratio of gynogenetic diploid reared at $27^{\circ}C$ was very close to 1 : 1 ratio (P<0.01), The survival rates of control and gynogenetic diploid reared at $27^{\circ}C$ were different from other water temperature groups. The growth of body weight of control and gynogenetic diploid reared at $18^{\circ}C$ were different from other water temperature groups.
