넙치 전 암컷 집단의 생산을 위한 연구 IV. 성분화 기간 중 수온에 의한 성 결정

사육 수온이 넙치의 성비에 미치는 영향을 조사하기 위해 일반 넙치 및 자성발생성 2배체 넘치 자어를 대상으로 부화 후 35일(전장 1.2 cm)부터 100일(전장 12 cm)까지 사육 수온 18, 21, 24 및 $27^{\circ}C$로 나누어 2회 반복하여 실험하였다. 사육 수온이 높을수록 암컷의 비율은 낮게 나타나 사육 수온과 암컷 비율에 있어 역상관관계를 보였다. 자성발생성 2배체 실험군의 경우 $18^{\circ}C$에서 $94.4\%$의 암컷을 보인 반면 $27^{\circ}C$에서는 $57.6\%$로 나타나 암수비가 1 : 1이었다(P<0.01). 사육 수온에 따른 생존율은 일반 넙치 및 자성발생성 2배체 넙치 모두 18, 21, 24 및 $27^{\circ}C$ 순으로 높았으나 $27^{\circ}C$를 제외한 나머지 실험군은 유의차가 없었으며 $18^{\circ}C$를 제외한 나머지 실험군의 체중 성장은 유의차가 없었다(P>0.05). We investigated the effects of rearing water temperatures during sex differentiation period on sex ratios in olive flounder, Paralichthys olivaceus. Control and gynogenetic diploid juvenile flounder reared at water temperature of 18, 21, 24 and $27^{\circ}C$ for 65 days from 35 to 100 days after hatching. Fish were sampled to examine sex ratios at 195 or 260 days after hatching. Female ratios of control and gynogenetic diploid flounder declined rapidly as water temperature increased. Sex ratio of gynogenetic diploid reared at $27^{\circ}C$ was very close to 1 : 1 ratio (P<0.01), The survival rates of control and gynogenetic diploid reared at $27^{\circ}C$ were different from other water temperature groups. The growth of body weight of control and gynogenetic diploid reared at $18^{\circ}C$ were different from other water temperature groups.

넙치 양식의 생산성을 높이는 방법의 일환으로 수컷에 비해 성장이 빠른 암컷만의 생산을 위하여 먼저 참돔 정자를 이용한 자성발생성 이배체의 유도 조건, 성장, 생존율 및 성비를 조사하였다. 그 결과 자성발생성 이배체 넙치는 $4,800ergs/mm^2$의 자외선으로 불활성화 시킨 참돔 정자와 성숙된 넙치 난을 인공 수정시켜 수정 후 3분부터 수온 $2^{\circ}C$에서 45분 동안 제 2극체 방출의 억제에 의하여 유도되었다. 유도된 자성발생성 이배체의 성장과 생존율은 부화후 200일째까지는 대조군과 유의차가 없었으나, 부화후 300일째는 자성발생성 이배체의 성장이 대조군보다 양호하였다. 또한 자성발생성 이배체의 암컷 비율은 $92.1\%$로 대조군의 $43.6\%$에 비하여 월등히 높았다. Gynogenetic diploid of olive flounder, Paralichthys olivaceus were induced by cold shock to fertilized eggs with red sea bream, Paragus major sperm that had been genetically inactivated with 4,800 ergs/$mm^2$ ultraviolet (UV) rays. Cold shock to the eggs at $2^{\circ}C$ for 45 minutes proved to be optimum condition to retain the second polar body. At this treatment, hatching rates of normal fry obtained were more than $33.8\%$. No different growth rates were observed up to 200 days after hatching between control and gynogenetic diploid offsprings. However, body weights of gynogenetic diploids were significantly heavier than that of control 300 days after hatching (p> 0.05). A proportion of female in gynogenetic diploid was significantly higher than that in the control (p< 0.01).
