Gateway cloning system을 이용한 목화진딧물 cDNA Library Construction

목화진딧물의 방제에 RNA interference(RNAi)를 이용하여 새로운 시각으로 새로운 방제를 시도하고자 한다. RNAi를 이용하여 목화진딧물의 방제에 이용할 target유전자들을 선발하기 위하여 gateway system을 이용한 목화진딧물 cDNA library를 제작하였다. 그 결과 RNAi에 적합한 약 100~400bp의 insert를 확인하였으며, blast search 및 EST database 비교분석 결과, 목화진딧물 관련 유전자임을 확인하였고, 최종적으로 8.4x105 titer의 목화진딧물 cDNA library를 완성하였다. 이러한 cDNA library는 att site를 가지는 TRV(Tobacco rattle virus) RNA2 vector에 LR recombination한 다음 Agrobacterium tumefacience(GV2260)에 transformation하였다. Agro-infiltration을 통하여 RNAi기작이 진단된 오이 에 목화진딧물을 접종하여 섭식시켰다. 섭식을 통한 살충 또는 기피효과를 bio-assay함으로써 target유전자를 선발하는 데 이용될 수 있을 것으로 사료된다.
Construction of a cDNA library of Aphis gossypii Glover for use in RNAi

Aphis gossypii Glover is an important insect pest that functions as a viral vector and mediates approximately 45 different viral diseases. As part of a strategy for control of A. gossypii, we investigated the functions of genes using RNAi. To this end, a cDNA library was constructed for various genes and for selecting appropriate targets for RNAi mediated silencing. The cDNA library was constructed using the Gateway cloning system with site‐specific recombination of bacteriophage λ. It was used to carry out single step cloning of A. gossypii cDNAs. As a result, a cDNA library with a titer of 8.4 × 106 was constructed. Since the sequences in this library carry att sites, they can be cloned into various binary vectors. This library will be of value for various studies. For later screening of selected genes, it is planned to clone the library into virus‐induced gene silencing (VIGS) vectors, which makes it possible to analyze gene function and allow subsequent transfection of plants. Such transfection experiments will allow testing of RNAi‐induced insecticidal activity or repellent activity to A. gossypii, and result in the identification of target genes. It is also expected that the constructed cDNA library will be useful for analysis of gene functions in A. gossypii.
Fanconi 증후군 치료 후 골동통이 호전된 다발성 골수종 1예

Adult Fanconi syndrome is characterized by variable abnormalities caused by renal proximal transport defects, resulting in glycosuria, aminoaciduria, bicarbonaturia, uricosuria and phosphaturia. A 57-year- old man with κ-light chain multiple myeloma, undergoing chemotherapy with prednisolone and melphalan for 17 month, was admitted with spontaneous femoral neck fracture and was consulted due to polyuria and refractory metabolic acidosis immediately after hemiarthroplasty. The laboratory values showed normal anion gap metabolic acidosis with normal urinary anion gap, hypokalemia, hypouricemia, hypophosphatemia at the time of consultation. After partial correction of acidemia, the fractional excretion of HCO3- was 11.9%, it was interpreted as proximal renal tubular acidosis. 24-hour urine collection showed increased level of excretion for most aminoacids. Diffuse osteopenia and multiple compression fractures on spine were detected on radiological examinations. Also, osteoporosis and osteomalacia was suggested during his clinical course. After the diagnosis of Fanconi syndrome was made, treatment was started with sodium bicarbonate, potassium citrate, calcitriol, calcium carbonate along with phosphate rich diet. Laboratory abnormalities were corrected and refractory multiple bone pain was ameliorated with these treatment.

친환경농자재를 이용한 톱다리개미허리노린재의 친환경적 방제

Recently, the rate of cultivation of such as hairy vetch was increased as a way of reducing the current soil problem. However, the occurrence of insect pest such as bean bugs, Riptortus pedestris, were increased in the field of green manure crops and their injuries to hairy vetch were observed. While minimizing insecticide use for the environmental friendly agriculture, the control of the bean bug can be utilized environmental friendly agricultural materials (EFAMs) in the green manure crop fields. The control effects of some EFAMs and most of chemical insecticides to the bean bug were significantly high. As a result of direct spray of EFAM that contained sophora extract or neem extract, the control effects of six EFAMs were higher than 70% at 120 hours after treatments. Among them, three EFAMs were showed over 90% of control effects. On the other hand, most of chemical insecticides were showed 100% of control effect against the bean bug at 48 hours after treatment. As mortality effects of EFAMs were sloely observed until 120h after treatment, we must use selected EFAMs at the beginnibgs of occurrence and entrance in the field of green manure for effective control of bean bugs.
콜레마니진디벌(Aphidius colemani) 촉각에 분포하는 감각기의 종류 및 외부미세구조적 특징

Several species of the genus Aphidius are used in biological control programs against aphid pests throughout the world and their behavior and physiology are well studied. While the importance of sensory organs in their behavior is understood, their antennal structure remains largely unknown. In this study, the external morphology and distribution of the antennal sensilla on the antennal of both female and male adults of A. colemani were described using scanning electron microscopy (SEM). Generally, the filiform antennae of males ($l,515.2{\pm}116.5\;{\mu}m$) are longer than females ($l,275.1{\pm}103.4\;{\mu}m$). Antennae of this species are made up of radicle, scape, pedicel and flagellomeres. Male and female antennae differed in the total number of flagellomeres as 15 in males and 13 in females. The result of SEM observation was characterization and grouping of seven types of receptors into morphological classcs: Bohm bristles, Sensilla placodea, 2 types of S. coeloconica and S. trichodea, and S. basiconica as with a tip pore and with wall pores. In addition, the possible functions of the above sensilla types arc discussed in light of previously published literature; mechanoreception (Bohm bristles, S, coeloconica I & II and S. trichodea) and chemoreception (S. placodea, S. basiconica type I & II). Aphidius속(Hymenuptera: Braconidae)에 속하는 일부 종은 주로 생물학적 방제 인자로서 사용되고 있으며, 행동학적 생태에 대하여 연구가 많이 되어있다. 행동에 있어서 신경기관을 중요성을 알고 있음에도 불구하고 촉각의 구조는 널리 알려져 있지 않다. 본 연구에서는 콜레마니진디벌의 암수 성충의 촉각에 분포하는 감각기를 주사전자현미경(SEM)을 이용하여 분류하고, 외부형태를 관찰하였다. 일반적으로 콜레마니진디벌의 촉각은 염주상(filiform)의 형태로 수컷($1,515.2{\pm}116.5\;{\mu}m$)이 암컷($l,275.1{\pm}103.4\;{\mu}m$)보다 더 길다. 촉각은 촉각근절(radicle), 밑마디(scape), 자루마디(pedicel, 채찍마디(flagellomeres)로 구분된다. 암 수컷의 채찍마디의 수에서 수컷은 15마디, 암컷은 13마디로 차이가 나타났다. 콜레마니진디벌의 촉각을 SEM을 이용하여 관찰한 결과, 총 7 종류의 감각기 즉; Bohm bristles, Sensilla placodea와 두 가지 타입의 S. coeloconica, S. trichodea, 또 tip pore 또는 wall pores가 있는지 여부에 띠라 두 가지 S. basiconica를 확인할 수 있었다. 이전에 연구들을 바탕으로 각 감각기들의 기능을 추측해볼 때 Bohm bristles과 S. coeloconica type I & II, S. trichodea 등은 기계적감각을, S. placodea, S. basiconica type I & II는 회학적감각을 담당할 것으로 추정된다.
EPG를 이용한 담배가루이(Bemisia tabaci)의 섭식행동과 Cyantraniliprole에 대한 섭식변화

Feeding behaviors of the sweet potato whitefly, Bemisia tabaci, and changing of their feeding behaviors were recorded and analyzed with an Electrical Penetration Graph (EPG) analysis against cyantraniliprole. The characteristic patterns of feeding behaviors investigated were as follows; NP (non probing), pathway phase waveform, waveform of feeding xylem. In results, B. tabaci did not almost feeding the phloem. And It was not appeared potential drop during recording time. It was predominantly indicated pathway activity waveform and waveform of xylem feeding. After treatment of cyantraniliprole with recommended concentrations to tomato plants, EPG waveforms were recorded during 3 hours. Cyantraniliprole treatment showed longer time to first feeding xylem than untreated (P=0.043). It was showed shorter duration of first feeding xylem than untreated (P=0.017). And it was showed longer NP (non-probing) time than untreated. Consequently, It was supposed that cyantraniliprole was effect of anti-feeding or avoidance to B. tabaci.

딸기수출재배단지에서 발생하는 점박이응애, 진딧물류, 작은뿌리파리의 분포를 파악하여 방제에 활용하고자 조사를 수행하였다. 진주시 수곡면에 위치한 딸기재배단지의 농가 20곳(토경재배 15곳, 양액재배 5곳)을 선정하여 2주 간격으로 3개월간 조사를 실시하였다. 재배방법에 따른 해충발생의 차이는 뚜렷하지 않았고, 전반적으로 점박이응애는 발생하였으나 진딧물과 작은뿌리파리 유충은 발생이 미비하였다. 작은뿌리파리 성충은 시간이 경과됨에 따라 점차 감소하는 추세로 기온에 영향을 받는 것으로 사료되며, blue sticky trap보다 yellow sticky trap에서 더 많이 채집되었다. 따라서 점박이응애, 진딧물류는 천적을 활용하고, 작은뿌리파리는 yellow sticky trap을 이용하여 방제하는 것이 효율적일 것이라 사료된다.
