        α- Phenyl - cysteine 유도체의 합성에 관한 연구

        Through hydrolysis of hydantoin with barium hydroxide amino acid can be prepared. By applying this method new four α-phenyl-cysteine derivatives were synthesized.

      • 잠재적 COX-2의 억제작용이 있는 1,5-Diarylhtdantoin 유도체의 합성

        For the development of new COX-2 inhibitors, 1,5-diarylhydantoins 5a~5c and 1,5-diaryl-2-thiohydantoin 6a~6c were synthesized from commercially available phenylacetic acids through esterification, bromination, C-N bond formation and cyclization. Esters 2a~c were efficiently synthesized from the starting materials la~c by refluxing in absolute methanol for 3 hours with catalytic concentrated sulfuric acid. Bromination of 2a~c was carried out with use of N-bomosuccinimide at rt in dichloromethane. The bromine of 3a~c was substituted with aniline in ethanol or N,N-dimethylformamide to provide 4a~c. Hydantoins and 2-thiohydantoins were synthesized from 4a~c by treatment of potassium isocyanate or potassium thiocyanate in dil-ethanol with triethylamine.

        With the increasing concern about drug abuse among young people, information is needed that will help to assess the incidence and the nature of drug abuse in Korea. There are only a few studies of drug use among junior and senior high school students and this report was planned to add another sample. The author performed a questionnaire survey on drug use among 4201 junior and senior high school students in Seoul. He found that 20.1% of the students had used drugs at least once in the past and stimulants were used most extensively. The author also found that the students with a bad home background and had school record and juvenile delinquents had used more drugs.

      • (±)-2-(p-Haloaryl)-2-hydroxyacetic acid 유도체의 합성

        2-Hydroxyacetic acids are often used as starting compounds in organic synthesis. There are a lot of synthetic methods of 2-hydroxyacetic acid derivatives. but often it is not easy to synthesize some derivatives. In this study condensation reaction of diethyl oxomalonate with aromatic hydrocarbons was applied. In the condensation reaction of 4′-halo-1.1′-biphenyls (Cl. Br) with diethyl oxomalonate in the presence of tin(Ⅳ) chloride diethyl 2-〔(4′-halo-1.1′-biphenyl)-4-yl〕-2-hydroxymalonates were formed and then the corresponding esters were hydrolyzed with potassium hydroxide to the 2-hydroxydicarboxylic acids. By decarboxylation of the corresponding 2-hydroxymalonic acids in c-HCl at 100℃ the final products (±)-2-〔(4′-halo-1.1′-biphenyl)-4-yl〕-2-hydroxyacetic acids were prepared.

      • 잠재성 소염진통작용이 있는 Hydantoin-3-Acetic Acids 및 그 유도체 합성

        Through the Bucherer-Bergs method, new 5-alkylthio (sulfonyl)-methyl-p-x-phenylhydantoins were prepared. Hydantoin-3-acetates were synthesized by the reaction of the hydantoins and ethyl chloroacetate and through the subsequent hydrolysis of the 3-acetates with dilute sodium hydroxide hydantoin-3-acetic acids were obtained. These products are expected to exhibit antiinflammatory and analgetic activities.

        Garlic (Allium sativum L) is one of the oldest cultivated plants and has been used throughout the world as food supplement and folk medicine for thousands of years. In modern times a number of garlic derived products are introduced on the market as health food supplement in ever growing scale. In 1844 German chemist Wertheim investigated the garlic first time chemically and thereafter many kinds of organosulfur compounds were isolated and their biological activities were elucidated scientifically. The main biological activities are antibacterial, antifungal, antithrombotic, cholesterol-lowering, antineoplastic and hepatoprotective activities. Chemical works as well as therapeutic and preventive effects of garlic are reviewed.

      • 5-Phenyl-4,5-dihydro-1H-benzazepin-2 one의 합성

        The first of benzodiazepines was chlordiazepoxide, which was marketed as tranquilizer under the trade name Librium. Now benzodiazepine derivatives are prescribed for insomnia and muscle relaxants as well as to treat anxiety. As benzodiazepines used widely, drug abuse becomes a serious social problem because of psychological and physical dependence and over deaths. It is necessary to develop the in vitro diagnostic reagents for the detection of small amount of abused drugs in urines. Because accuracy and convenience for point of care testing systems are indispensable, immunological determinations are fit for our requirements, which react with specific drug antigens and are able to detect small amount of drugs in urine. A benzazepinone derivative(4), which will be used as a component of the reagents for detecting benzodiazepine drugs or their metabolite in urine, was synthesized from 5-phenyltetrahydrofuran-2-one(1) as a key intermediate.

        Aryloxyallylthiopyridazine 유도체 합성

        Allylthio group of the sulfur compounds of garlic oil plays an important role for prevention and treatment of hepatic diseases induced by toxic substances or carbon tetrachloride. Thus allylthio group as pharmacologically active group was introduced into pyridazine heterocycle ring. Aryloxyallylthiopyridazine derivatives were synthesized and their hepatoprotective activities were screened in rat. The activities of these compounds were weaker than alkoxyallylthiopyridazine derivatives, which exhibit a superior hepatoprotective effect.

      • 3-Alkylthio-6-allylthiopyridazine 유도체 합성 및 SK-Br3 인간유방암세포와 Hep-G2 인간간암세포에 대한 항암작용

        Allylthio group of allicin and other organosulfur compounds which are isolated from garlic is considered as a pharmacophore, a key structure component of the molecule which is due to biological activities. In the foregoing studies various 3-alkoxy-6-allylthiopyridazine derivatives (K-compounds) were synthesized and their biological activities were tested in test animals. They showed good hepatoprotective activities on the carbon tetrachloride-treated mouse and aflatoxin B1-treated rats and chemopreventive activities on hepatocarcinoma cells in rat as expected. Now two pyridazine derivatives whose oxygen atom at 6-position of 3-alkoxy-6-allylthiopyridazine is displayed by sulfur(S) were synthesized and their activities were synthesized and their activities were tested in vitro against SK-Br3 human breast cancer cells and Hep-G2 human liver cancer cells. Thio-K6 and Thio-K16 showed good chemopreventive activities on hepatocarcinoma and breast cancer.
