      • NF1-B, C bind and repress the TSHβ subunit gene

        TSH β-subunit is a subunit of the TSH, which is produced by thyrotropes of the anterior pituitary grand. TSH β-subunit gene is elaborately regulated by thyrotrope-specific transcription factors. In this study, EMSA and DNase I protection assays were employed to characterize the interactions between nuclear proteins of the thyrotropes and the -577/-565 region of the TSH β-subunit promoter. Interestingly enough, multiple sequence alignments showed that this -577/-565 region is highly conserved among several mammalians including human, chimpanzee, rat, mouse and dog. In addition, transcription factor cluster analysis identified the NF1 binding sequences in these conserved regions. Based on these observations, we demonstrated that NF1 physically interact with the -577/-565 region of the TSH β-subunit promoter and functionally repress the transcription. Taken together, these results suggest that transcription factor NF1 is involved in the TSH β-subunit gene regulation in thyrotropes of the anterior pituitary gland.
