A New Cytotoxic Spiroketal from the Myxobacterium Sorangium cellulosum

A new spiroketal, spirodienal B (1), was isolated from culture extracts of S. cellulosum. Its structure was elucidated by spectroscopic analysis. In addition to 1, the strain produced a second, closely related antibiotic, spirodienal. Both compounds demonstrated potent cytotoxicity against human tumor cells. Against human tumor cells such as HCT-15, the activity of 1 was more than 4000 times stronger than that of doxorubicin in terms of IC50.
Antioxidative Iridoid Glycosides and Phenolic Compounds from Veronica peregrina

Eight iridoid glycosides and four phenolic compounds were isolated from the EtOAc soluble fraction of Veronica peregrina MeOH extract as the radical scavengers for antioxidant activity. The compounds were identified as protocatechuic acid (1), luteolin (2), veronicoside (3), minecoside (4), specioside (5), amphicoside (6), catalposide (7), 6-O-cis-p-coumaroyl catalpol (8), p-hydroxy benzoic acid methyl ester (9), verproside (10), verminoside (11), and chrysoeriol 7-glucuronide (12) by spectroscopic analysis. All compounds except for 1 and 2 were isolated for the first time from this plant. The antioxidant activity was evaluated by the ORAC(Oxygen Radical Absorbance Capacity) assay, which measures scavenging activity against peroxy radicals induced by 2,2'-azobis (2-methoxypropion-amidine) dihydrochloride, and the ORAC value is expressed as relative trolox equivalent. Compounds 2, 4, 5, 6, 8, and 12 exhibited potent antioxidant activity, and compounds 1, 11 had similar activity with trolox, whereas the other compounds showed weaker activity than trolox.
Cytotoxic Phenolic Compounds from Chionanthus retusus

Cytotoxicity-guided fractionation and purification of MeOH extract from Chionanthus retusus Lindl. et Paxton resulted in the isolation of compounds. Fourteen phenolic compounds were isolated from the EtOAc soluble fraction, and their structures were determined by spectroscopic analysis. Isolated compounds were identified as phillygenin (1), scopoletin (2), pinoresinol (3), kaempferol (4), aromadendrin (5), 2-(4-hydroxyphenyl)ethanol (6), 3,3',5,5',7-pentahydroxyflavanone (7), luteolin (8), quercetin (9), apigenin (10), chrysoeriol (11), phillyrin (12), oleuropein (13), (7R,8R)-guaiacylglycerol (14). All compounds except for 12 and 13 were isolated for the first time from this plant and genus Chionanthus. Anti-proliferative effect of isolated compounds were evaluated by the sulforhodamin B assay against four human tumor cell lines (A549, SK-OV-3, SK-MEL-2, and HCT15). Compounds 4 and 8-10 were significantly active with ED50 values of 1.84-6.35 μg/mL. Also, compounds 1, 3, 5 and 7 revealed cytotoxic effects at concentrations below 30 μg/mL.
In vitro 3D skin model using gelatin methacrylate hydrogel

Interests in in vitro skin models have been growing. Collagen, which is a main scaffold material for in vitro 3D skin models, has weak mechanical properties, often resulting in undesirable contraction. The physiological characteristics of the skin models often depend on the matrix in which cells are cultured. In this study, we developed a 3D skin model using gelatin methacrylate. The mechanical and transport properties were studied, and attachment and growth of fibroblasts and keratinocytes were examined. Fibroblasts preferred softer matrix, whereas HaCaT cells preferred harder matrix of gelatin methacrylate. This study provides information for developing in vitro skin models.

Four steroids and one flavonol glycoside were isolated from the ethanol extract of the whole plant of Aconitum pseudolaeve var. erectum. Their structures were identified as ${\beta}-sitost-4-en-3-one$, 22-dihydro-stigmast-4-en-3,6-dione, ${\beta}-sitosterol$, ${\beta}-sitosterol-3-O-{\beta}-D-glucopyranoside$ and $kaempferol-3-O-{\beta}-D-glucopyranoside(astragalin)$ on the basis of spectral data.

Abstratc-Urtica angustifolia has been used to cure diabetes etc. as a folk medicine. The anit-diabetic effect of U. angustifolia was investigated by the experimental diabetic model in rats. U. angustifolia extracts were administered before and after injection of alloxan(50 mg/kg, i.v.), and the blood glucose and serum cholesterol levels were determined at 3 days after alloxan injection. The increased blood glucose and serum total cholesterol levels in the diabetic rats induced by alloxan were significantly decreased by administration of MeOH and BuOH extracts, relatively. And reduced body weight was recovered with the treatments of each extract. The MeOH and BuOH extracts of U. angustifolia showed remarkable anti-diabetic effect, and MeOH extract showed more strong anti-diabetic effect than BuOH extract.
