개 고밀도 단일염기다형(SNP)칩을 이용한 군견 고관절 이형성증에 대한 전장유전체 연관 연구

Objectives : Canine hip dysplasia (CHD) is a malformation in the development of the hip joint between acetabulum and femoral head that results in osteoarthritis. The highest incidence of CHD occurs in military working dogs (MWDs) such as German Shepherd and Belgian Malinois. Although CHD is a genetic disease, its diagnosis has been only judged by clinical findings in aged dogs. In this study, a single nucleotide polymorphism (SNP) was investigated in the blood of MWDs to develop a concise and exact detection method for CHD at early ages. Methods : 70 MWDs (57 GSDs and ` BSDs) aged between 2-15 years old were assigned as based on OFA scores measured by X-ray diagnosis. Blood samples from 70 MWDs were collected in each EDTA tube, extracted using a Wizard genomic DNA purification kit (Promega, Madison, WI, USA), and analyzed concentrations and purities at absorbances of 260 nm and 280 nm using a ND-1000 spectrophotometer (NanoDrop Technologies, CA, USA). Each sample was genotyped on a Canine HD BeadChip (Illumina Inc., San Diego, CA, USA) containing 173,662 loci distributed across the entire canine genome. Results : Using the Plink program, 137,123 SNP markers were finally selected through a filtering process. When a genome-wide threshold level was determined using Bonferroni correction (i.e. 1/137,123=7.30×10-6), three significant SNP markers were found on CFA4. Conclusions : Three markers (BICF2S2352810, BICF2P832958 and BICF2P308917) were selected as a candidate marker for CHD test, which is a significant SNP marker among other SNPs. The present study was conducted to establish the relationship between CHD and marker using GWAS. Genotype of 3 SNPs on BICF2S2352810, BICF2P832958 and BICF2P308917 has indicated a normal for CHD through A, G, and C, respectively

The purpose of approximate analyses may be to compare the performance of alternative proposals for the structures, or to .determine the deflections and major member farces in the chosen structures so as to allow it to be properly proportioned. One virtue of this approximate analysis for drift is that it easily allows the drift contributions of the individual frame members to be seen, there by providing guidance as to which members should be increased in size to most effectively reduce an excessive total drift or story drift. This study presents approximate hand calculation of drift for the braced frames changing the eccentricity ratio(e/L) and results is compared with the computer analysis. Results show that the drift calculated by the approximate analysis is some larger than computer analysis, but that is acceptable.
