In order to detect the aflatoxins in home-made Takju and peanut butter, the samples were collected in Chungbuk region and cleaned up Sep-pak silica cartridge. Aflatoxins were detected by thin layer chromatographic and high performance liquid chromatographic behavior. Determination was carried out by thin layer densitometer. The results were as follows; 1. Aflatoxin B; was detected in 78% of the home-made Takju, and the highest concentration was 1.2 ppb and average 0.36 ppb. 2. Aflatoxins were not detected in any peanut butter smaples. 3. Clean-up method by Sep-pak silica cartridge was more efficient and economical than column chromatography of AOAC method.
간장장해와 Ketosis시에 Cytochrome P450IIE의 Regulatory Mechanism에 관한 연구

In order to elucidate the alteration of drug-metabolizing enzymes and mechanism in the animal with hepatic injury and ketosis, the regulation of P450IIE was studied in the rats with heaptic injury caused by CCl$_4$ and with ketosis caused by streptozotocin and high-fat diet. P450IIE expression in liver was examined by the combination of enzyme activities, Western immunoblot, and mRNA Northern blot analyses using specific polyclonal antibody and cDNA probe for P450IIE. Enzyme activity and amounts of immunoreactive P450IIE were rapidly decreased in a time-dependent manner after a single dose of CCl$_4$ . However, the decreases in P450IIE enzyme activity and immunoreactive protein by CCl$_4$ were not accompanied by a decline in its mRNA level. The data thus suggested a post-translational reduction of P450IIE by CCl$_4$. The enzyme activities (aniline hydroxylase) in hepatic microsomes were elevated about 2-3-fold by streptozotocin and feeding with a high fat diet. This increases in enzyme activities were also accompanied by 3-fold increases in immunoreactive P450IIE protein and its mRNA. Our data thus indicated that P450IIE induction during the ketosis appears to be due to pretranslational activation.

The genotoxicity of combinations of four p-oxybenzoic acids (methyl paraben, ethyl paraben, isopropyl paraben, butyl paraben) and benzoic acid had been evaluated. The in vitro Ames test using Salmonella typhimurium (TA 98, TA 100, TA 1535, TA 1537) and the in uiuo micronucleus assay using mouse peripheral blood were performed. Methyl paraben plus benzoic acid, ethyl paraben plus benzoic acid, and ethyl paraben plus butyl paraben slightly increased the frequency of micronucleated reticulocytes in the high doses, but were negative in Ames test with Salmonella typhimurzum with and without rat liver microsomal activation. The other combinations tested were negative in Ames test and did not show any clastogenic effect in micronucleus test. These results suggest that genotoxicity can produced by the combinations of p-oxybenzoic acid.

6-(4-Iodophenyl)amino-7-chloro-5,8-quinolinedione (RCK9) was evaluated for antifungal activities. The MIC values of RCK9 were determined against A. flavus, C. albicans, C. neoformans and F. oxysporium. The RCK9 showed generally potent antifungal activities against the tested fungi. Acute oral toxicity studies of RCK9 were carried out in ICR mice of both sexes. These acute oral toxicities of RCK9 were low and LDSO values were over 2,850 mg/kg in ICR mice. The genotoxicities of RCK9 had been evaluated. RCK9 was negative in Ames test with Salmonella typhimurium and chromosomal aberration test in CHL cells. The clastogenicity was tested on the RCK9 with in vivo mouse micronucleus assay. RCK9 did not show any clastogenic effect in mouse peripheral blood and was negative in mouse micronucleus assay. The results indicate that RCK9 has no genotoxic potential under these experimental conditions.
Effects of Diazinon on the Murine Host Defense System

Diazinon which is one of the most heavily used organophosphate pesticide in Korea, was examined for its effects on the marine host defense system. Immunotoxicological assay parameters adopted in this study were carbon clearance for macrophage function, susceptibility to tumor challenge, and pathotoxicological indicators. Subchronic exposure of pesticide to rodents resulted in the suppression of immune functions, enhancement of susceptibility to tumor challenge, and moderate histological changes of lymphoid organ without any significant alterations of clinical status.
PCB ( Aroclor 1254 ) 가 한국산 담수어류에 (淡水魚類) 미치는 영향

The purpose of this study was to elucidate the adverse effects of Aroclor 1254 on domestic freshwater fish. The acute toxicity in terms of LC_(50)(mg/L) in 96 hours was evaluated for the freshwater fish, Oryzias latipes, Cyprinus carpio and Misgurnus anguillicaudatus. 96-hr LC_(50) values(that concentration kills half of the firth in 96 hours) were 0.053mg/L for newly hatched Oryzias latipes but above 200mg/L for adult Oryzias latipes. Concentrations of Aroclor 1254 above 100mg/L were not lethal to Misgurnus anguillicaudatus but half of Cyprinus carpio were dead after 96 hours in 100mg/L concentration. To assess the danger of Aroclor 1254 to aquatic invertebrates, bioassays were conducted with Daphnia magna. Fifty percent of hatched Daphnia magna died at 0.048mg/L concentration after 96 hours. It is suggested from this study that further study on Aroclor 1254 or other PCBs should be focussed en the mortality of the fish larvae with chronic exposure at low concentrations.

2010. 7. 1 이후 건조(K/L, Keel Laying)되는 120m 이상 또는 주 화재구역(Main Fire Zone)을 3 개 이상 가지는 여객선에 이른바 SRtP 법규(SOLAS Ⅱ-1 reg.8-1, SOLAS Ⅱ-2 reg.21, 22) 적용이 시행되었다. 이 법규에 의하면 여객선의 주요 의장시스템들은 사고분계점(Casualty Threshold) 이하의 화재나 침수상황에서도 작동하도록 설계되어, 선박이 안전하게 귀항할 수 있도록 해야 한다. 또한 본 법규의 승인을 위해서는 선박의 주요 의장시스템들이 위 사항을 만족한다는 것을 체계적인 엔지니어링 분석을 통해 증명하여야 한다. SRtP 법규의 증명은 널리 알려진 FMEA(Failure mode and effect analysis)나 FTA(Fault Tree Analysis) 혹은 BDD(Binary Decision Diagram)과 같은 방법론들이 사용될 수 있으나, SRtP 가 적용되는 의장시스템의 수가 방대하고 시스템 간 연계가 복잡하여 분석자가 일일이 분석하거나 트리를 구성하여야 하는 위 방법론들을 적용하기에는 어려움이 있다. 따라서, 본 연구에서는 SRtP 평가를 위한 새로운 방법론과 이를 이용한 SRtP 평가 프로그램을 제안하였다. 제안된 방법론에서는 작동조건을 가지는 장비모델을 이용하여 SRtP 분석을 수행함으로써 분석자의 개입을 최소화하였고 이를 통해 시스템 분석을 자동화하였다. 또한 본 논문에서는 제안된 SRtP 평가프로그램의 구성과 여기에 사용된 새로운 SRtP 분석 방법론 그리고 개발된 프로그램을 이용한 Fuel oil system 의 SRtP 분석결과를 살펴보았다.
