        Characterization of Post-Rainy Season Grown Indigenous and Exotic Germplasm Lines of Sorghum for Morphological and Yield Traits

        Sorghum is a major staple crop and vital for the marginal farmers in Asian and African countries. Landraces or germplasm lines adapted to biotic and abiotic stresses are the prime source of adaptive traits in the crop breeding programs. In order to assess the genetic variability, 141 exotic germplasm lines and 36 popular varieties were evaluated for eight agro-morphological traits. Wide range of values were observed for grain yield (6.6-124.4 g/plant), seed index (1.5-7.1 g/100 seeds), panicle length (8-44 cm) and panicle width (7.25 cm). High genotypic and phenotypic coefficient of variation was observed for grain yield per plant. The values of broad sense heritability in the sorghum core population ranged from 0.75 to 0.99. Seed weight, days to flowering, panicle width and leaf area were significantly correlated with grain yield (P ≤ 0.01). Seed weight was positively correlated with panicle width (correlation coefficient 0.31), while negatively correlated with panicle length (correlation coefficient -0.15). Cluster analysis resolved all the genotypes into four major clusters. Among germplasm lines, TSG-313 had high seed weight of 7.05 g/100 seeds, while TSG-325 had highest grain yield of 124.4 g/plant as against control variety. Germplasm lines with high heritability scores would help us to utilise them in recombination breeding.

        Cell death in culture: Molecular mechanisms, detections, and inhibition strategies

        Mammalian cell cultures are widely used in the biopharmaceutical industry to produce monoclonalantibodies, vaccines, growth factors, etc. Cell death is an essential biological process for physiologicalgrowth and development, but it is a major problem in biopharmaceutical production in bio-industry. Celldeath within bioreactor occurs due to various intracellular and extracellular stresses. These stressesnegatively affect the culture longevity, overall product quality, and yield. Among all cell death types,apoptosis accounts for most of the cellular death in the bioreactor. The implementation and developmentof various strategies to prevent the cellular death are crucial for robust bioprocess development. Celldeath during culture can be prevented or inhibited by supplementing media with specific chemicals,synthetic inhibitors, and genetic cell engineering approaches. In this review, we classified and describeddifferent types of cell death and their molecular mechanisms and summarized the cell death inhibitionapproaches implemented to inhibit cell death for various applications.

        Variability of Azadirachtin in Azadirachta indica (neem) and Batch Kinetics Studies of Cell Suspension Culture

        Seeds of neem were collected from different parts of India and analyzed for their azadirachtin content by High Performance Liquid Chromatography (HPLC). In order to assess the effects of genotypic and geographical variation on azadirachtin content in cell cultures, callus development was attempted in the seeds containing high and low concentration of azadirachtin. The concentration of azadirachtin in callus cultures was significantly affected by the explant source. Seed kernels with higher azadirachtin content produced higher azadirachtin content in callus cultures and lower azadirachtin content was seen in callus cultures produced from seed kernels with low azadirachtin content. The protocol for development of elite stock culture of Azadirachta indica was established with the objective of selecting a high azadirachtin-producing cell line. The highest azadirachtin-producing cell line was selected and the effects of different media and illumination conditions on growth and azadirachtin production were studied in shake flask suspension culture. Detailed batch growth kinetics was also established. These studies provided elite starter culture and associated protocols for cultivation of A. indica plant cell culture in the bioreactor.

        Production of Azadirachtin from Plant Tissue Culture: State of the Art and Future Prospects

        With increasing awareness towards environment-friendly and non-toxic pesticide azadirachtin obtained from neem tree (Azadirachta indica) is gaining more and more importance. Its broad-spectrum activity, peculiar mode of action. eco-friendly and non-toxic action towards beneficial organisms has offered many advantages over chemical pesticides. All currently use commercial formulations based on azadirachtin contains azadirachtin extracted from seeds of naturally grown whole plants which is labour intensive process depending upon many uncontrollable geographical and climatic factors. Plant tissue culture can be a potential process for the production, offering consistent, stable and controlled supply of this bioactive compound, However the research on tissue culture aspects of production are in preliminary stage and requires culture and process optimization for the development of a commercially viable process. This review states the present status and future challenges of plant tissue culture for azadirachtin production.

        Bioprocess Strategies and Recovery Processes in Gibberellic Acid Fermentation

        Gibberellic acid (GA3) is a commercially important plant growth hormone, which is gaining much more attention all over the world due to its effective use in agriculture and brewing industry. Industrially it is produced by submerged fermentation technique using Ascomycetous fungus Gibberella fujikuroi. Solid state and immobilized cell fermentation techniques had also been developed as an alternative to obtain higher yield of GA3. This review summarizes the problems of GA3 fermentation such as production of co-secondary metabolites along with GA3, substrate inhibition and degradation of GA3 to biologically inert compound gibberellenic acid, which limits the yield of GA3 in the fermentation medium. These problems can be overcome by various bioprocessing strategies e.g. two - stage and fed batch cultivation processes. Further research on bioreactor operation strategies such as continuous and / or extractive fermentation with or without cell recycle / retention system need to be investigated for improvement in yield and productivity. Down stream processing for GA3 isolation is also a challenge and procedures available for the same have been critically evaluated.

        Male Oxidative Stress Infertility (MOSI): Proposed Terminology and Clinical Practice Guidelines for Management of Idiopathic Male Infertility

        Despite advances in the field of male reproductive health, idiopathic male infertility, in which a man has altered semen characteristics without an identifiable cause and there is no female factor infertility, remains a challenging condition to diagnose and manage. Increasing evidence suggests that oxidative stress (OS) plays an independent role in the etiology of male infertility, with 30% to 80% of infertile men having elevated seminal reactive oxygen species levels. OS can negatively affect fertility via a number of pathways, including interference with capacitation and possible damage to sperm membrane and DNA, which may impair the sperm’s potential to fertilize an egg and develop into a healthy embryo. Adequate evaluation of male reproductive potential should therefore include an assessment of sperm OS. We propose the term Male Oxidative Stress Infertility, or MOSI, as a novel descriptor for infertile men with abnormal semen characteristics and OS, including many patients who were previously classified as having idiopathic male infertility. Oxidation-reduction potential (ORP) can be a useful clinical biomarker for the classification of MOSI, as it takes into account the levels of both oxidants and reductants (antioxidants). Current treatment protocols for OS, including the use of antioxidants, are not evidence-based and have the potential for complications and increased healthcare-related expenditures. Utilizing an easy, reproducible, and cost-effective test to measure ORP may provide a more targeted, reliable approach for administering antioxidant therapy while minimizing the risk of antioxidant overdose. With the increasing awareness and understanding of MOSI as a distinct male infertility diagnosis, future research endeavors can facilitate the development of evidence-based treatments that target its underlying cause.

        Effect of Dietary Fiber Enrichment and Different Cooking Methods on Quality of Chicken Nuggets

        The effect of dietary fiber enrichment (wheat bran) and cooking methods (oven, steam and microwave) on functional and physico-chemical properties of raw nuggets formulation as well as nutritional, color and textural properties of chicken nuggets were analyzed in this study. Among different cooking methods used for nuggets preparation, steam cooked nuggets had significantly (p<0.05) higher water holding capacity (56.65%), cooking yield (97.16%) and total dietary fiber content (4.32%) in comparison to oven and microwave cooked nuggets. The effect of cooking methods and wheat bran incorporation was also noticed on textural properties of the nuggets. Hardness, firmness and toughness values of oven and steam cooked nuggets were significantly (p<0.05) higher than microwave cooked nuggets. Among nuggets prepared by different cooking methods, cohesiveness of microwave cooked nuggets was found to be significantly (p<0.05) highest, whereas, oven cooked nuggets had significantly (p<0.05) highest gumminess and chewiness values. Steam cooked nuggets were found to be better among all nuggets due to their higher cooking yield and dietary fiber content.

        Study on diuretic activity and electrolytes excretion of methanol extract ofLippia nodiflora (Verbenaceae) in rats

        In the Indian traditional medicine, Lippia nodiflora (Verbenaceae) whole plant is claimed to possess powerful diuretic activity. However, the diuretic potential of this plant is not yet investigated. The aim of this study was to evaluate the diuretic potential of methanol extract of Lippia nodiflora (MELN) in rats. Control (0.9% saline solution, 25 ml/kg, b.w) or urea (1 g/kg b.w) or frusemide (5 mg/kg b.w) and different concentrations of MELN (200 and 400 mg/kg b.w) were intraperitoneally administered (n = 6 per each treatment group) to hydrated rats and their urine output was monitored over a period of 5 h and 24 h after drug administration. The diuretic responses with its electrolyte excretion potency of the extract were highly remarkable in comparison with control animals. The extract at doses of 200 and 400 mg/kg shows a significant increase in volume of urine with increase in Na+, Ca2+ and Cl- excretion accompanied by the excretion of K+ in dose dependent manner. This study suggests that the active component(s) in MELN had similar diuretic effect to that of frusemide. These results validate the traditional use of Lippia nodiflora as a diuretic agent.
